Using of natural sources that have anti-inflammatory activity for the prevention and treatment of degenerative diseases began to be further explored. An investigation on the anti-inflammatory activity of the aqueous extract of guava leaves (Psidium guajava L.) from Sawangan, Depok on white male rats of Sprague-Dawley strain had been carried out on the carrageenan-induced paw edema method. To examine the effect of guava extract on subcutan at different doses of 125, 250 and 500 mg/kg of body weight (BW). Indometacine at dose of 10 mg/kg BW was used as a positive control. Observations were made during five hours with an interval of one hour. These results demonstrate that the percentage of inflammation or edema (% E) optimal at the 4th hour and then decreased at the 5th hour, while the percentage of optimal inhibition occurred at the 5th hour. Guava extract at 125, 250 and 500 mg/kg BW reduced inhibitory percentage activities by 40.81, 55.45 and 43.61% (p< 0.05) respectively. In conclusion, this study suggests that guava extract has anti-inflammatory properties by decreasing edema level.Keywords: Anti-inflammatory, guava leaves, edema.
Katalase (EC 1.11.16) adalah merupakan enzim antioksidan yang bertindak sebagai biokatalisator dengan mengubah H2O2 menjadi H2O dan O2. Enzim ini berperan penting dalam meregulasi metabolisme H2O2. Hidrogen peroksida adalah merupakan sebuah molekul kecil hasil samping dari metabolisme. Kelebihan H2O2 akibat defisiensi Katalase, menyebabkan kerusakan yang signifikan pada protein, DNA, RNA dan lipid sehingga hal ini juga berperan pada patogenesis berbagai penyakit, termasuk diabetes. Dalam studi in silico ini, perubahan yang terjadi pada mutasi SNP (single nucleotide polymorphism) C (330) T (rs 10011179) dari gen CAT akan diselidiki, termasuk hubungannya dengan risiko peningkatan resiko diabetes. Untuk itu dilakukan pencarian bioinformasi, diantaranya informasi tentang gen, protein dan penelusuran informasi penting lainnya yang dapat digunakan sebagai informasi pendukung penelitian. Analisis bioinformatika gen CAT dilakukan dengan menggunakan beberapa program open source dari situs web. Pada penelitian ini ternyata terjadi mutasi misssense SNP Ala110Val (C330T, rs1001179) yang ditemukan pada struktur 3D CAT pada urutan asam amino ke-110 (C330T) yang mengganggu fungsi amidasi protein CAT. Mutasi ini menyebabkan berkurangnya ekspresi gen CAT, yang berakibat pada berkurangnya aktivitas CAT sebagai biokatalis dan sebagai enzim antioksidan. Hal ini terkait dengan peningkatan risiko diabetes tipe 1.
Diabetes Mellitus (DM) merupakan penyakit metabolik dengan ciri hiperglikemia yang terjadi karena kelainan sekresi insulin, kerja insulin, atau keduanya. COVID-19 adalah penyakit menular yang disebabkan oleh Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) yang petama kali ditemukan di kota Wuhan, China pada bulan Desember 2019. COVID-19 yang disertai dengan diabetes mellitus diketahui mempunyai resiko prognosis yang lebih buruk daripada yang tidak disertai diabetes meliitus. Penelitian ini bertujuan untuk mengetahui apakah terdapat peningkatan mortalitas dalam kasus COVID-19 yang disertai diabetes mellitus.Jenis penelitian ini menggunakan Literature Review dengan menggunakan jurnal yang diambil dari database sebanyak 20, dengan keyword pencarian jurnal yaitu “COVID-19, Mortalitas, dan Diabetes”. Limitasi pencarian jurnal yaitu 2 tahun, berhubungan dengan COVID-19 yang merupakan penyakit baru. Penyaringan jurnal menggunakan PRISMA Flow Diagram, sehingga didapatkan jurnal yang sesuai dengan kriteria yang telah ditetapkan.Didapatkan 20 jurnal yang relevan dengan kriteria yang telah ditetapkan, dengan 18 jurnal mendukung hipotesis alternatif, dan 2 jurnal tidak mendukung. Jurnal yang paling mendukung hipotesis alternatif dengan Odd Ratio sebesar 12.234 (Cl 95% 4.126-36.272), p0.026 oleh Albitar, Ballouze, Ooi, et al., 2020, ditemukan juga bahwa diabetes tipe 1 dan tipe 2 yang merupakan penyerta COVID-19, mempunyai pengaruh yang besar dalam menaikan resiko kematian pasien COVID-19, dibuktikan oleh jurnal Barron, Bakhai, Kar, et al., 2020 dengan diabetes tipe 1 (HR 1.61(1.32-1.96)) dan diabetes tipe 2 (HR 1.61(1.54-1.67)), p0,0001, ditemukan juga bahwa pasien COVID-19 dengan riwayat diabetes mellitus juga mempunyai resiko kematian yang tinggi, dibuktikan oleh jurnal dari Yu, Lei, Li, et al., 2020 dengan (OR 2.34(1.45-3.76)), p0.005
Changes in lifestyle and modernization have caused a shift in the pattern of diseases from communicable to non-communicable diseases (NCD). The NCD category includes diabetes, hypertension, obesity, coronary heart disease. Non-communicable diseases are a challenge during the Covid-19 pandemic because they are comorbid, which will aggravate the disease. Therefore it is necessary to educate on a healthy lifestyle to avoid these new infectious diseases. The method was carried out through webinars with the topic of an active lifestyle, teachings on faith and introduction to herbs to PKK cadres (Family Welfare Empowerment – FWE) in Ciseeng village. The metabolic health characteristics have normal values of 0%, 26%, 100%, 74% for waist circumference (WC), body mass index (BMI), systolic and diastolic blood pressure, respectively. Knowledge of healthy lifestyle from the webinar increased from 51.30% to 85.96% (p <0.05). The conclusion is that the metabolic health of respondents is considered at risk, while webinar activities reduce the risk of comorbidities.
