RESUMO -Este estudo teve como objetivo avaliar o efeito do regulador de crescimento ácido indolbutírico (AIB) no enraizamento de miniestacas apicais e intermediárias de clones de Eucalyptus urophylla x E. globulus e Eucalyptus grandis x E. globulus. O enraizamento das miniestacas foi conduzido durante 30 dias em casa de vegetação climatizada e, em seguida, as mudas foram transferidas para casa de sombra, permanecendo por 10 dias. O delineamento experimental utilizado foi em blocos ao acaso, em arranjo fatorial triplo, constituído de dois tipos de miniestacas (apicais e intermediárias), três concentrações de AIB (0, 2000 e 4000 mg L -1 ) e 11 clones, em três repetições, compostas de parcelas com 12 miniestacas por repetição. Não houve efeito do AIB sobre as características avaliadas. A sobrevivência e o enraizamento das miniestacas apicais foram significativamente superiores. Houve diferença significativa entre clones quanto às características avaliadas, exceto a sobrevivência após 30 dias na casa de vegetação.Palavras-chave: Propagação clonal, Miniestaquia e Ácido indolbutírico. RESUMO -Objetivou-se com o presente estudo avaliar a eficiência da técnica de miniestaquia na propagação vegetativa de progênies de meios-irmãos de angico-vermelho (Anadenanthera macrocarpa (Benth) Brenan) quanto à produção de brotações e sobrevivência das minicepas, enraizamento das miniestacas apicais e intermediárias tratadas com diferentes doses do AIB (0; 2.000; 4.000 e 6.000 mg L -1 ), assim como determinar a velocidade de enraizamento em casa de vegetação. As minicepas foram obtidas a partir de mudas produzidas via sementes de seis progênies de meios-irmãos de angico-vermelho. Com base nos resultados obtidos, as minicepas apresentaram produtividade de 1,2 a 3,7 miniestacas/minicepa/coleta e sobrevivência de 84% a 98% ao longo das seis coletas realizadas. As miniestacas apicais foram superiores em relação às intermediárias, com maior predisposição ao enraizamento, no entanto o AIB não teve efeito significativo sobre o enraizamento das progênies estudadas. Resumo -A micropropagação apresenta um enorme potencial de aplicação para a multiplicação de genótipos de espécies florestais brasileiras de interesse. Os estudos relativos ao cultivo in vitro de tais espécies estão relacionados, principalmente, a ausência de resposta morfogênica às demais técnicas de propagação bem como, a conservação in vitro de germoplasma. A micropropagação via proliferação de gemas axilares corresponde ao principal sistema de propagação in vitro utilizado para a multiplicação de genótipos selecionados, em razão da maior simplicidade ao comparar com a organogênese e embriogênese somática. Entretanto, em vista das espécies florestais nativas serem pouco estudadas, os avanços quanto a propagação in vitro ainda são poucos expressivos. Dessa forma, é necessária a condução de novos trabalhos para promover avanços do cultivo in vitro nestas espécies, bem como o desenvolvimento de novas composições de meios de cultura, testar a interação de reguladores de crescimento ou, até mesmo, sistemas de biorreatores, buscando assim, consolidar a técnica de micropropagação como estratégia aplicável na silvicultura de espécies nativas.
Root explants of a wild passionfruit species (Passiflora cincinnata) and three P. edulis commercial populations ('FB 100', 'FB 200', and 'FB 300') were incubated on Murashige and Skoog (MS) medium supplemented with 4.44 lM 6-benzyladenine (BA) to induce shoot organogenesis. Shoots elongated in liquid medium with 2.89 lM gibberellic acid (GA 3 ) under agitation were rooted in coconut fiber and acclimatized followed by transfer to a greenhouse into pots containing mixture of coconut fiber and Plantmax Ò (1:1). Explant samples were collected during organogenesis and submitted to light and scanning electron microscopy (SEM). Root explants of P. cincinnata responded earlier than those of P. edulis. However, on the third assessment, at 90 days, the genotype 'FB 200' showed shoot number significantly higher than 'FB 100' and 'FB 300', not differing from P. cincinnata.Organogenesis in P. cincinnata and P. edulis occurred via direct pathway, which was confirmed by anatomical studies and SEM. Flow cytometric analysis revealed no variation in DNA content of regenerated plantlets among all genotypes. Nuclear DNA (2C) values (pg) in regenerants of P. cincinnata (2.99 pg) and P. edulis (3.26-3.28 pg) were consistent with DNA amounts of seed-derived control plants.
We addressed a major challenge in the in vitro clonal propagation of Corymbia citriodora, Eucalyptus urophylla and E. benthamii by using an ex vitro adventitious rooting strategy in a mini-incubator. Miniincubators were placed in four environments for rooting. A shade house with no fogging system and a greenhouse with no ventilation but with a fogging environment had the best performance in terms of rooting, root growth and survival of microcuttings. Daily recording of the temperature within each miniincubator in each environment allowed the verification of negative correlations between the maximum average temperature and the survival, adventitious rooting and root growth. The ideal maximum air temperature for the efficient production of clonal plants was 28.4°C (± 5.5°C), and the minimum was 20.3°C (± 6.2°C). E. benthamii was more sensitive to higher temperatures than C. citriodora and E. urophylla. Nevertheless, placing mini-incubators in the shade house with no fogging system resulted in a stable and uniform performance among the three species, with 100.0% survival and 81.4% rooting. Histological sections of the adventitious roots revealed connection with the stem vascular cambium. Therefore, our experimental system demonstrated the potential of mini-incubators coupled with the proper environment to optimize the adventitious rooting performance of microcuttings.
RESGATE VEGETATIVO DE ÁRVORES DE Anadenanthera macrocarpaRESUMO: Objetivou-se, neste trabalho, avaliar o resgate de árvores adultas de angico vermelho (Anadenanthera macrocarpa) pela decepa da árvore e anelamento do caule, bem como a viabilidade da propagação vegetativa da espécie por meio da estaquia. O delineamento experimental utilizado foi o de blocos ao acaso, em arranjo fatorial 2 x 2, constituído de duas técnicas para a indução de brotações basais (anelamento e decepa) e dois tipos de estacas (herbáceas e semilenhosas), com quatro repetições compostas de 12 estacas por parcela. O anelamento do caule e a decepa da árvore foram eficientes na indução de brotações basais em árvores de A. macrocarpa, sendo que a decepa proporcionou maior emissão de brotações basais. As estacas herbáceas apresentaram maiores porcentuais de enraizamento, independente da técnica de resgate. De modo geral, o resgate e a propagação vegetativa de árvores adultas de A. macrocarpa foi tecnicamente eficiente. VEGETATIVE RESCUE OF Anadenanthera macrocarpa TREES ABSTRACT:The objective of this work was to evaluate the vegetative rescue of adult Anadenanthera macrocarpa trees through the induction of epicormic shoots from tree severance and stem ringing, as well as the efficiency of cutting technique on the propagation of the species. The experimental design used was random plots, in a 2 X 2 factorial arrangement, constituted by two techniques for the induction of basal shoots (tree severance and stem ringing) and cuttings types (herbaceous cuttings and semihardwoods cuttings), in four replicates and plots of 12 cuttings per repetition. The tree severance and stem ringing were efficient in the induction of basal shoots in adult A. macrocarpa trees. However, the tree severance provided higher emission of basal shoots. The herbaceous cuttings presented higher rooting percentages, independent of vegetative rescue technique. In general, we conclude that the vegetative rescue and the vegetative propagation of A. macrocarpa adult trees are technically efficient.
