The Muller F element (4.2 Mb, ~80 protein-coding genes) is an unusual autosome of Drosophila melanogaster; it is mostly heterochromatic with a low recombination rate. To investigate how these properties impact the evolution of repeats and genes, we manually improved the sequence and annotated the genes on the D. erecta, D. mojavensis, and D. grimshawi F elements and euchromatic domains from the Muller D element. We find that F elements have greater transposon density (25–50%) than euchromatic reference regions (3–11%). Among the F elements, D. grimshawi has the lowest transposon density (particularly DINE-1: 2% vs. 11–27%). F element genes have larger coding spans, more coding exons, larger introns, and lower codon bias. Comparison of the Effective Number of Codons with the Codon Adaptation Index shows that, in contrast to the other species, codon bias in D. grimshawi F element genes can be attributed primarily to selection instead of mutational biases, suggesting that density and types of transposons affect the degree of local heterochromatin formation. F element genes have lower estimated DNA melting temperatures than D element genes, potentially facilitating transcription through heterochromatin. Most F element genes (~90%) have remained on that element, but the F element has smaller syntenic blocks than genome averages (3.4–3.6 vs. 8.4–8.8 genes per block), indicating greater rates of inversion despite lower rates of recombination. Overall, the F element has maintained characteristics that are distinct from other autosomes in the Drosophila lineage, illuminating the constraints imposed by a heterochromatic milieu.
Facilities housing captive animals are full of staff who, every day, interact with the animals under their care. The expertise and familiarity of staff can be used to monitor animal welfare by means of questionnaires. It was the goal of our study to examine the association between chimpanzee (Pan troglodytes) welfare, happiness, and personality. To these ends we collected two waves of welfare and subjective well-being ratings of 18 chimpanzees housed at the Edinburgh Zoo and one set of ratings of 13 chimpanzees housed at Fundació Mona. Ratings were made on a welfare questionnaire that included 12 items related to stress, psychological stimulation, and behavioural indicators of negative and positive welfare states, and a 4-item subjective well-being questionnaire. In addition, ratings were made on the 54-item Hominoid Personality Questionnaire and an abbreviated version of this scale consisting of 37 antonym pairs. We used generalizability theory to test whether welfare ratings generalized across items, raters, chimpanzees, and time. We then assessed the validity of the welfare and subjective well-being questionnaires by examining their associations with behaviour. Finally, we tested whether the welfare and subjective well-being ratings were associated with personality. Welfare ratings generalized across items, raters, chimpanzees, and time. Principal components analysis and regularized exploratory factor analysis indicated that ten welfare items and all four subjective well-being items formed a single dimension (welfareSWB).LASSO regression found that lower welfareSWB was associated with regurgitation, coprophagy, urophagy, and decreased proximity to nearest neighbour. A linear model that adjusted for age, sex, and facility, indicated that higher Extraversion and lower Neuroticism were related to higher welfareSWB. Welfare ratings were reliable and associated with subjective well-being and personality, demonstrating that staff ratings are a valid and potentially valuable tool for chimpanzee welfare assessment.
26Welfare questionnaires, which allow people who are familiar with individual animals 27 to rate the welfare of the animals, are an underutilised tool. We designed a 12-item welfare 28 questionnaire and tested its reliability and associations with subjective well-being (SWB), 29 locomotor stereotypy, and personality traits. The welfare questionnaire included questions 30 relating to physical health, stress and coping, satisfaction with social relationships, 31 psychological stimulation, and the display of positive and negative welfare indicators. We 32 collected ratings of 66 brown capuchins (Sapajus apella) living in three facilities. Each 33 capuchin was rated on the welfare questionnaire by an average of 2.8 raters. The interrater 34 reliability of the welfare questionnaire items ranged from ICC(3,k) 0.51 to 0.86. A principal 35 components analysis indicated that the 12 welfare items loaded onto one component. We 36 repeated this process with the welfare and subjective well-being items and found all the items 37 were defined by a single component (welfareSWB). We then conducted three sets of analyses, 38 one predicting the welfare component, one predicting the SWB component, and predicting the 39 welfareSWB component. The independent variables were frequency of locomotor stereotypy, 40 personality, age, and sex; facility was included as a random effect. In models including 41 stereotypy, age, and sex we found frequency of stereotypy to be significantly associated with 42 all three predicted components (p<0.01). After controlling for stereotypy (b=-0.25, p=0.17), 43 age (b=-0.54, p=0.01), and sex (b=-0.32, p=0.07), the personality traits of Sociability (b=1.02, 44 p<0.001), Assertiveness, (b=0.63, p<0.001), and Attentiveness (b=0.54, p=0.01) were 45 associated with higher scores on the joint welfareSWB component; Neuroticism was 46 negatively associated (b=-0.60, p=0.01). Previous studies of nonhuman primates have found relationships between health and individual differences in personality, behavior, and social status. However, despite knowing these factors are intercorrelated, many studies focus only on a single measure, for example, rank. Consequently, it is difficult to determine the degree to which these individual differences are independently associated with health. The present study sought to untangle the associations between health and these individual differences in rhesus macaques (Macaca mulatta). We studied 85 socially housed macaques at the Oregon and California National Primate Research Centers, and used veterinary records to determine the number of injuries and illnesses for each macaque. We measured personality using 12 items from a well-established primate personality questionnaire, performed focal observations of behaviors, and calculated dominance status from directional supplant data. All twelve personality questionnaire items were reliable and were used to represent five of the six personality dimensions identified in rhesus macaques-Dominance, Confidence, Openness, Anxiety, and Friendliness (also known as Sociability). Following this, we fit generalized linear mixed effects models to understand how these factors were associated with an animal's history of injury and history of illness. In the models, age was an offset, facility was a random effect, and the five personality dimensions, behavior, sex, and dominance status were fixed effects. Number of injuries and illnesses were each best represented by a negative binomial distribution. For the injury models, including the effects did improve model fit. This model revealed that more confident and more anxious macaques experienced fewer injuries. For the illness models, including the fixed effects did not significantly improve model fit over a model without the fixed effects. Future studies may seek to assess mechanisms underlying these associations.
One way to gain insights into personality evolution is by comparing the personality structures of related species. We compared the personality structure of 240 wild white-faced capuchin monkeys to the personality structure of 100 captive brown capuchin monkeys. An ancillary goal was to test the degree to which different personality questionnaires yielded similar personality dimensions. Both species were rated on a common set of 26 antonym pairs. The brown capuchin monkeys were also rated on the 54-item Hominoid Personality Questionnaire. Our cross-species comparisons revealed three personality dimensions---Assertiveness, Openness, and Neuroticism---shared by brown and white-faced capuchins, suggesting that these dimensions were present in the common ancestor of these species. Our comparison of the dimensions derived from the antonym pairs and the Hominoid Personality Questionnaire revealed that three common dimensions were identified by both questionnaires. In addition, the dimension Attentiveness was only identified using the Hominoid Personality Questionnaire. These results indicate that major features of capuchin personality are conserved and that the structure of some traits, such as those related to focus, persistence, and attention, diverged. Further work is needed to identify the evolutionary bases that led to the conservation of some dimensions but not others.
Puppy assessments for companion dogs have shown mixed long-term reliability. Temperament is cited among the reasons for surrendering dogs to shelters. A puppy temperament test that reliably predicts adult behavior is one potential way to lower the number of dogs given to shelters. This study used a longitudinal design to assess temperament in puppies from 8 different breeds at 7 weeks old (n = 52) and 6 years old (n = 34) using modified temperament tests, physiological measures, and a follow-up questionnaire. For 7-week-old puppies, results revealed (a) puppy breed was predictable using 3 variables, (b) 4 American Kennel Club breed groups had some validity based on temperament, (c) temperament was variable within litters of puppies, and (d) certain measures of temperament were related to physiological measures (heart rate). Finally, puppy temperament assessments were reliable in predicting the scores of 2 of the 8 adult dog temperament measures. However, overall, the puppy temperament scores were unreliable in predicting adult temperament.
Aspects of personality in nonhuman primates have been linked to health, social relationships, and life history outcomes. In humans as well as nonhuman primates, facial morphology is associated with assertiveness, aggression, and measures of dominance status. In this study we aimed to examine the relationship among facial morphology, age, sex, dominance status, and ratings on the personality dimensions Confidence, Openness, Assertiveness, Friendliness, Activity, and Anxiety in rhesus macaques ( Macaca mulatta ). We measured facial width-to-height ratio (fWHR) and lower-height/full-height ratio (fLHFH) using photographs from 109 captive rhesus macaques, which observers also assessed for dominance status and personality, and explored the associations among facial morphology, age, sex, dominance status, and personality. fWHR and fLHFH personality associations depended on age category: Assertiveness was associated with higher fWHR and fLHFH, and Confidence was associated with lower fWHR and fLHFH, but all these associations were consistent only in individuals <8 yr. of age. We found fWHR and fLHFH to not be consistently associated with sex or dominance status; compared to younger individuals, we found few associations with fWHR and fLHFH for individuals older than 8 yr., which may be due to limited sample size. Our results indicate that in macaques <8 yr. old, facial morphology is associated with the Assertiveness and Confidence personality dimensions, which is consistent with results suggesting a relationship between fWHR and trait aggression in humans and assertiveness in brown capuchins, all of which implies that fWHR might be a cue to assertive and aggressive traits.
