Inflammasomes integrate cytosolic evidence of infection or damage to mount inflammatory responses. The inflammasome sensor NLRP1 is expressed in human keratinocytes and coordinates inflammation in the skin. We found that diverse stress signals induce human NLRP1 inflammasome assembly by activating MAP kinase p38: While the ribotoxic stress response to UV and microbial molecules exclusively activates p38 through MAP3K ZAKα, infection with arthropod-borne alphaviruses, including Semliki Forest and Chikungunya virus, activates p38 through ZAKα and potentially other MAP3K. We demonstrate that p38 directly phosphorylates NLRP1 and that serine 107 in the linker region is critical for activation. NLRP1 phosphorylation is followed by ubiquitination of NLRP1PYD, N-terminal degradation of NLRP1, and nucleation of inflammasomes by NLRP1UPA-CARD. In contrast, activation of NLRP1 by nanobody-mediated ubiquitination, viral proteases, or inhibition of DPP9 was independent of p38 activity. Taken together, we define p38 activation as a unifying signaling hub that controls NLRP1 inflammasome activation by integrating a variety of cellular stress signals relevant to the skin.
This work aimed to study the removal of malachite green dye from wastewaters through adsorption using raw corn straw (RCS) and ultrasound-assisted modified corn straw (MCS). RCS and MCS were prepared and characterized in detail. The characterization indicated that RCS and MCS presented favorable structures for malachite green adsorption and that the ultrasound treatment provided a disorganization of the adsorbent's crystalline regions and also caused the formation of cavities and protuberances. The adsorption study was performed by equilibrium isotherms, kinetic curves, thermodynamic parameters, and application in real effluents composed of dye mixtures and inorganic compounds. The Elovich model was suitable for the adsorption kinetics and the Freundlich model was appropriate to represent the equilibrium. The maximum experimental adsorption capacities were 200 mg g for RCS and 210 mg g for MCS, obtained at 328 K. MCS was more effective than RCS to treat real effluents, attaining around 92% of color removal.
In this research, chitin and rice husks were used as adsorbents to recover cobalt from leached solutions of lithium-ion batteries. The waste materials were obtained and characterized. At first, the adsorption study was performed in batch systems using synthetic solutions, where the pH effect, kinetic, equilibrium, and thermodynamic aspects were investigated. Later, in the best conditions, chitin and rice husks were applied to recover cobalt from real leached solutions of lithium-ion batteries. For both materials, the adsorption was favoured at pH ¼ 6.0. The maximum adsorption capacities were 50.0 mg g À1 and 17.6 mg g À1 for chitin and rice husks, respectively, obtained at 318 K. The Co þ2 recovery percentages from real leached solutions were 95.0 % and 40.0 % for chitin and rice husks respectively. These results revealed that chitin can be used as an alternative and low-cost waste material to recover Co þ2 from real leached solutions of lithium-ion batteries.
Physicochemical and thermodynamic interpretations of malachite green (MG) adsorption on raw (RCS) and modified corn straw (MCS) are detailed in this work. Experimental isotherms were constructed at different temperatures (from 298 to 328 K). To interpret these curves, three models named multilayer model with saturation, double layer model with two energies, and monolayer model with one energy were developed based on the principle of grand canonical ensemble in statistical physics. The double layer model with two energies was the more adequate to represent the adsorption process. Thermodynamic functions such as configurational entropy (Sa), free adsorption enthalpy (G), and internal energy (Eint) were studied. The number of MG molecules per site (n) ranged from 0.33 to 2.33 and the adsorbed quantity at saturation (Qasat) ranged from 164.15 to 528.19 mg g−1 and both increased with the temperature. The density of the receptor sites (NM) ranged from 248.71 to 104.88 and decreased with the temperature. For RCS and MCS, the first and second energies, ϵ1 and ϵ2, increased with the temperature, with ϵ1 being higher than ϵ2. The adsorption energies of MCS (7.77–11.19 kJ mol−1) were higher than the adsorption energies of RCS (6.17–10.88 kJ mol−1). The adsorption process occurred by physisorption and the disorder increased at low equilibrium concentrations, but decreased at high concentrations. The free enthalpy and internal energy described with care the spontaneity of the studied system.

We found that diverse stress signals converge on the activation of p38 kinases to initiate human NLRP1 inflammasome assembly: UV irradiation and microbial molecules that initiate the ribotoxic stress response critically relied on the MAP3 kinase ZAKα to activate p38 and ultimately human NLRP1. Infection with insect-transmitted alphaviruses, including Semliki Forest, Ross River, and Chikungunya virus, also activated NLRP1 in a p38-dependent manner. In the absence on ZAKα, inflammasome assembly was maintained, although at reduced levels, indicating contribution of other upstream kinases. NLRP1 activation by direct nanobody-mediated ubiquitination was independent of p38 activity. Stimulation of p38 by overexpression of MAP2 kinases MKK3 or MKK6 is sufficient for NLRP1 activation, and NLRP1 is directly phosphorylated by p38. Taken together, we define p38 activation as a unifying signaling hub that controls NLRP1 inflammasome activation by integrating a variety of cellular stress signals relevant to the skin.
RESUMO -O presente trabalho teve como objetivo estudar a remoção do corante verde malaquita de águas residuais através da adsorção utilizando palha de milho in natura e tratada via ultrassom. O modelo de pseudo-segunda ordem foi adequado para a cinética de adsorção e o modelo de Freundlich foi apropriado para representar o equilíbrio. A máxima capacidade de adsorção encontrada para os experimentos com efluente sintetizado em concentração de 200 mg L -1 foi de 257,1 mg g -1 para a palha de milho in natura e de 283,8 mg g -1 para o adsorvente modificado, gerando 66,6 % e 73,3 % de remoção, respectivamente, enquanto para os experimentos com o efluente real obteve-se 56,3 % e 63,2%, respectivamente. INTRODUÇÃOCom o desenvolvimento industrial, agravam-se os problemas ambientais devido à eliminação de rejeitos tóxicos. Efluentes industriais contendo corantes sintéticos são gerados em diferentes setores industriais, tais como têxtil, papel, couro, plástico e processamento mineral, e o seu despejo é uma das principais fontes de poluição aquática (Khataee et al., 2015). Esses rejeitos, quando eliminados sem um tratamento adequado, representam perigo ao meio ambiente (Srinivasan e Viraraghavan, 2010). Existem diversos métodos de tratamento para a remoção de corantes de efluentes aquosos. Uma das técnicas mais eficientes é a adsorção, devido ao seu baixo custo, baixa necessidade energética e simples operação (Esquerdo et al., 2014). Devido à necessidade de um adsorvente presente em larga escala, reutilizável e de baixo custo, resíduos agroindustriais vêm sido pesquisados na remoção de corantes. Neste trabalho, objetivou-se estudar a adsorção do corante Verde Malaquita, utilizando palha de milho in natura e modificada via ultrassom. MATERIAL E MÉTODOS Desenvolvimento do adsorventeA palha de milho foi obtida em uma propriedade rural no estado do Rio Grande do Sul. O material passou por lavagem com água destilada, secagem em estufa por 24 h a 40 °C, moagem e peneiramento até atingir a faixa granulométrica de 68 a 75 μm. Em seguida, o pó resultante foi lavado novamente com água deionizada, até atingir pH 7, e seco em estufa por 24 h a 40 °C. A modificação via ultrassom da palha de milho realizou-se em um processador ultrassônico (UP400S, Hielscher, Alemanha) de 400 W, equipado com um sonotrodo de titânio,
RESUMO -O presente trabalho teve como objetivo investigar a recuperação de cobalto a partir de soluções lixiviadas de baterias utilizando a adsorção com casca de arroz. Primeiramente, utilizando-se soluções sintéticas de cobalto, foram estudados, o efeito do pH, a cinética e o equilíbrio da adsorção. Após, na condição mais apropriada obtida com as soluções sintéticas, a casca de arroz foi testada em soluções reais lixiviadas de baterias. Os resultados mostraram que o pH 6 foi o mais adequado para a adsorção. O modelo de pseudo-segunda ordem foi apropriado para a cinética de adsorção e o modelo de BET foi adequado para representar o equilíbrio. A capacidade máxima de adsorção experimental foi de 17,6 mg g -1 . Percentuais de remoção maiores que 95% foram obtidos utilizando-se 1,2 g/50 mL de adsorvente, mostrando que a casca de arroz é um adsorvente promissor para a recuperação de cobalto de soluções lixiviadas de baterias íon-lítio. INTRODUÇÃOA partir da década de 90, por razões ambientais e de eficiência, foram introduzidas no mercado as baterias íon-lítio, que passaram a ter grande consumo em escala mundial. Contudo, a expansão desse mercado trouxe como consequência um aumento na produção de lixo eletrônico, o qual é tóxico devido à presença de metais pesados. Assim tornou-se necessário o desenvolvimento de processos de reciclagem adequados para este tipo de resíduo (Busnardo et al., 2007). A reciclagem de baterias tornou-se interessante não só por conta de questões ambientais, mas também devido à presença de materiais valiosos, que recuperados poderiam gerar um retorno econômico. No caso das baterias íon-lítio, um dos metais mais valiosos é o cobalto (Busnardo et al., 2007), o qual, é relativamente raro e prejudicial à saúde humana em concentrações elevadas (He et al., 2011). A reciclagem de baterias íon-lítio esgotadas envolve, em maioria, técnicas hidrometalúrgicas. Uma das etapas no processamento destas baterias é a lixiviação ácida, a fim de que os metais sejam extraídos e estejam em solução aquosa. Posteriormente, utilizam-se técnicas para a remoção dos metais dessa solução resultante da lixiviação (Gurung et al., 2012).Dentre os diversos métodos utilizados para a remoção de metais pesados de soluções aquosas (Kumar and Acharya, 2013), a adsorção vem se destacando, sendo recomendada para soluções menos concentradas.
