Исследованы подходы к генной терапии атеросклероза путем невирусного переноса гена аполипопротеина Al (ароАІ) -основного белкового компонента антиатерогенных липопротеинов -в клетки печени. Сконструирован плазмидный вектор для экспрессии полноразмерного гена. ароАІ человека в клетках млекопитающих. Конструкция содержит последовательности бактериальной плазмиды, позволяющие ей реплицироваться в клетках Escherihia coli, и кассету для экспрессии гена ароАІ, фланкированную инвертированными терминальными повторами аденоассоциированного вируса человека. Для доставки векторной ДНК использован синтетический полимер полиэтиленимин (ПЭИ, 25 кДа). Показано>, что в результате ПЭИ-опосредованной трансфекции клеток в культуре аналогичным вектором, содержащим репортерный ген зеленого флуоресцирующего белка, 50-70 % клеток экспрессируют введенный ген. На экспериментальных животных, которым вводили конструкцию с геном ароАІ человека прямой инъекцией в печень, продемонст рировано, что белок АроАІ человека тестируется в плазме крови в течение 15 сут после доставки гена. ПЦР-анализ суммарной ДНК печени подопытных животных показал наличие расчетного фрагмента гена apoAJ человека у крыс через 15 дней после введения гена.
Клонированы фрагменты суммарной ДНК генома человека из лейкоцитов и клеток плаценты, способные обеспечивать автономное состояние клонирующего вектора pYF40 (в который они заключались) в Saccharomyces cerevisiae. Получены два фрагмента соответствующих молекулярных масс: 6,5 (из лейкоцитов) и 3,4 тыс. п. н. (из клеток плаценты), обладающие высокой трансформирующей способностью в пекарских дрож жах (Ю 1 -10 3 трансформантов на 10 мкг плазмидной ДНК). Выявлена продолжитель ная экспрессия гена аро-А1 человека в составе экстрахромосомных рекомбинантных ДНК, содержащих клонированные автономно реплицирующиеся АР-последовательности в культуре клеток.
