Collar rot is a disease difficult to control that has hindered passion fruit cultivation in many regions of Brazil. Therefore, this study aimed to find genetic resistance to the fungus Fusarium solani in Passiflora species using the multivariate analysis methodology to discriminate the most resistant species. The following fourteen Passiflora species were assessed: P. quadrangularis, P. nitida, P. foetida, P. tenuifila, P. alata, P. setacea, P. cincinnata, P. mucronata, P. micropetala, P. suberosa, P. morifolia, P. eichleriana, P. edulis and P. coccinea. These plants were arranged in a casualized block design with 14 treatments (species), three replications and three plants per plot. The reactions of the inoculated species of Passiflora were evaluated with the use of 12 resistance traits. The generalized Mahalanobis distance was used to form groupings by the UPGMA method. 3D projection with the canonical variables and quantification of the relative contribution of characters were also conducted. The UPGMA method revealed the formation of three distinct groups of species, which composed the susceptible, moderately resistant and resistant groups. The groups formed by threedimensional dispersion were similar to those formed by the dendrogram. The following traits contributed most to genetic diversity: inoculation time until the lesion reached more than 50% of the circumference of the injured stem and area under the curve of the expansion of the lesion width. The use of the set of traits and their joint assessment through multivariate analysis allowed greater accuracy in the inference of the most resistant species, mainly P. nitida and P. cincinnata.
Palavras-chave: fungos endofíticos, plantas medicinaisABSTRACT: Endophytic fungi associated with medicinal plants. With the use of medicinal plants in infusions, syrups, dyes, unguents, among other forms, it is expected that endophytic fungi, present inside the plants but not causing diseases, become components of these products, especially when used in natura. In addition, endophytic fungi can produce toxic substances to the users or even modify the plant metabolism, altering the medicinal composition and properties, as well as the quality of the stored and commercialized product. Therefore, the aim of this study was to isolate and identify the endophytic flora from eleven randomly chosen medicinal species. Pure cultures were obtained from the fungi Phomopsis, Colletotrichum, Pestalotia, Trichoderma, Fusarium, Nigrospora and Glomerella endophytically occurring in Plectranthus barbatus, Vernonia condensata, Pfaffia paniculata, Foeniculum vulgare, Cymbopogon citratus, Cymbopogon nardus, Cordia curassavica, Maytenus ilicifolia, Punica granatum, Morus nigra and Bauhinia forficata. The plant species that presented the highest number of endophytic fungi were Vernonia condensata, Punica granatum and Morus nigra. All fungi recovered in this study showed strictly endophytic features, not manifesting pathogenicity in their host species. Among the detected fungi, special attention must be given to the genus Fusarium, since a wide range of species of this genus are known to produce mycotoxins and constitute important post-harvest pathogens.
This work was carried out with the objective of performing a molecular and aggressiveness characterization of F. solani and F. oxysporum f.sp. passiflorae collected in the Pantanal, Cerrado and Amazon biomes. We selected the most aggressive isolates for use in breeding programs aiming resistance to Collar Rot and Fusariosis. For inoculation of the isolates of F. oxysporum f.sp. passiflorae the washed root method was used. The molecular characterization of the isolates was carried out by partial sequencing of the Transcribed Internal Spacer of the rDNA region. The isolates of F. solani formed two distinct groups in relation to aggressiveness. Among all isolates, FSUNEMAT 40 and FSUNEMAT 46 were the most aggressive. The model with K=2 was taken as the best model to explain the genetic structure of the F. solani populations, with clear combinations of genes from both gene pools. There were three groups with respect to the aggressiveness of the isolates of F. oxysporum f.sp. passiflorae, with the isolated FOUNEMAT 22 being the most aggressive. In view of these results, the isolates of F. solani collected in P. edulis in the state of Mato Grosso presented a high molecular variability independent from the biome of origin, and this was also observed in the tests of aggressiveness. The results indicate the need to consider the molecular variability and the aggressiveness of the pathogens in the evaluation of genotypes of sour passion fruit in programs of selection of resistant cultivars.
