The study of the human immune response to hepatitis B virus (HBV) has been hampered by the lack of an adequate model to evaluate the hepatitis B surface antigen (HBsAg) specific cell response. Thus, this study was conducted to perform an in vitro analysis of the antigenic properties of recombinant HBsAg and demonstrate the influence of variables such as culture time, antigen concentration and cell density on lymphocyte proliferation. Peripheral blood mononuclear cells (PBMCs) were isolated from the venous blood of vaccinated individuals, and in vitro cellular immune responses were evaluated using an HBsAg-specific proliferation assay. Lymphoproliferative responses were detected in culture systems, despite the lack of serum antibodies. Optimal results were obtained when lymphocytes were stimulated at a seeding density of 4×10(6) cells/mL, with 50 ng/mL of recombinant HBsAg protein vaccine for 3 days. Data from the present study may contribute to the development of an adequate system to evaluate the cellular immune responses to HBsAg in vaccine recipients.
The question of whether booster doses are required to maintain long-term protection against hepatitis B virus (HBV) after primary vaccination remains to be determined. Thus, the aim of this study was to evaluate the immune memory responses to hepatitis B surface antigen (HBsAg) challenge in vaccinated individuals through an in vitro-specific stimulation assay. Peripheral blood mononuclear cells (4 × 10(6) cells/ml) were stimulated with 50 ng/ml of recombinant HBsAg. In vitro anamnestic antibody responses, as shown by detection of high avidity antibody in culture supernatants, were found 13-18 years after primary vaccination and were not correlated with serum antibodies (r = -0.177; P = 0.377). In addition, the findings from this study indicate that immune memory against hepatitis B was well preserved in 40.0% and 60.0% of vaccinees with anti-HBs levels less than 10 IU/L or lacking serum antibodies altogether, respectively. In conclusion, the data suggest the presence of immunological memory in vaccinated individuals, including those who showed anti-HBs <10 IU/L or undetectable antibody.
O objetivo deste relato de experiência é apresentar a utilização de metodologias ativas mediadas por tecnologia em tempos de pandemia na disciplina Integração Básico-Clínica II do curso de Medicina. A experiência é caracterizada como uma possibilidade de contribuição ao desenvolvimento docente para o mundo pós-pandemia. Em um grupo de cinco professores e 40 estudantes, a disciplina Integração Básico-Clínica II foi adaptada emergencialmente para atividades mediadas por tecnologia. Os conteúdos trabalhados propiciaram a integração dos conhecimentos disciplinares à clínica e trouxeram inúmeros desafios pedagógicos neste momento inédito de como ensinar, aprender e sobreviver a uma pandemia. Em 16 encontros via plataforma institucional foram utilizadas exclusivamente metodologias ativas de ensino-aprendizagem, com estratégias diversificadas. O modelo de aprendizagem virtual não é algo novo, apesar de inédito para esse grupo de discentes e para esta disciplina e seu valor está em possibilitar repensar a educação e seus mecanismos, num extenuante exercício de criatividade para enfrentar novos desafios surgidos decorrentes de situações extremas. A aplicação de metodologias ativas associado ao feedback dos discentes foi imprescindível para a tomada de decisões na continuidade da integração dos conteúdos, levando à construção de uma disciplina agradável e que contribuiu para o processo formativo de todos os envolvidos. O maior aprendizado foi a necessidade de adaptação e criatividade, permitindo contribuições à construção da identidade docente. A práxis foi fortalecida e reinventada, sendo a adaptação e criatividade, atributos considerados necessários para o desenvolvimento docente para o mundo pós-andemia.
Nas últimas décadas houve um aumento significativo das ISTs entre a população da faixa etária superior a 60 anos. O objetivo foi avaliar o perfil sorológico e as representações sociais sobre a Sífilis em idosos. Para avaliar o perfil sorológico desse grupo foi realizado o teste imunocromatográfico treponêmico. Os testes foram realizados em 133 participantes, com idade entre 60 e 84 anos de ambos os sexos, com resultado não reagente em 100% dos casos. As representações sociais da sífilis para os idosos é de se tratar de uma doença perigosa, porém alguns dos entrevistados não a associaram como uma doença. Além disso, a maioria deles relaciona o uso de preservativos como método de prevenção de doenças, porém com baixa adesão ao uso. Os nossos resultados apontam que os idosos são vulnerais a sífilis.
Introduction: Medical students have an occupational risk for hepatitis B (HB). This study sought to determine anti-HBs and anti-HBc IgG levels in vaccinated students, check their seroconversion, and correlate this with vaccination. Methods: One hundred and forty-three students' blood samples and their vaccination schedules were analyzed. Results: 65.7% were positive for anti-HBs; however, anti-HBs was absent in 34.3%. Only two samples were positive for anti-HBc IgG. Conclusions: More than 30% of students did not have minimum protective levels. Comparing HBV vaccination and anti-HBs reactivity, the majority of reactive individuals received their last dose within the past 16 years.
Some animals have an important relationship with fungal infections, and searching for pathogens in animal samples may be an opportunity for eco-epidemiological research. Since studies involving wildlife are generally restricted, using samples from road kills is an alternative. The aim of this study was to verify whether pathogenic fungi of public health importance occur in wildlife road kills from Santa Catarina State, Brazil. Organ samples (n = 1063) from 297 animals were analysed according to Polymerase Chain Reaction (PCR) using universal primers to detect fungi in general and, subsequently, using primers specific to Paracoccidioides brasiliensis, Histoplasma capsulatum and Cryptococcus spp. There were 102 samples positive for fungal species. Eight samples were positive for P. brasiliensis, three samples were positive for Cryptococcus spp. and one sample had coinfection by these two fungi. No sample was positive for Histoplasma spp. according to the molecular detection. Genetic sequencing allowed the identification of Fungal sp. in 89 samples, Cryptococcus neoformans in two samples and Aspergillus penicillioides in three samples. This study shows the importance of wild animals in the epidemiology of fungal infections and assists in the mapping of pathogen occurrence in a region that was not previously evaluated.
The present study aimed to explore the influence of the gut-brain axis on neuroendocrine and immunological modulation in neurological and psychiatric disorders. This systematic review followed the Preferred Reporting Items for Systematic Reviews and Meta-analyzes (PRISMA) guidelines, and searches were conducted in the electronic databases PubMed and SciELO using combinations of descriptors “Gastrointestinal Microbiome”, “Neurosecretory Systems”, “Immune Response”, “Nervous System Diseases” e “Mental Disorders”. From the 144 studies generated by crossing the descriptors, 32 of them were excluded because they were duplicated in the databases, 13 because they were not related to the objectives of the review, and another 29 because they were not on eligibility criteria. Therefore, 70 studies were included in the present review. Communication between the GI tract and the CNS occurs via the neuronal, endocrine, and immunological pathways through a) the production of neurotransmitters, b) the tryptophan metabolism, c) the modulation of the immunological activity in the CNS and the SNE, d) production of short chain fatty acids, e) the production of intestinal hormones, and f) the production of branched chain amino acids.
Introdução: A criação das melíponas ou popularmente conhecidas como “abelhas sem ferrão” e a utilização do mel e seus derivados como recurso terapêutico para prevenir e tratar enfermidades, é uma prática antiga, com relatos desde os primórdios da civilização egípcia. A crescente busca e estudos realizados acerca do mel produzido por abelhas sem ferrão ocorre devido à sua composição química relacionada à efeitos antissépticos, antimicrobianos, anti-inflamatórios, anticancerígenos e cicatrizantes. Objetivos: Revisar a literatura dos últimos 5 anos, sobre as propriedades imunológicas, utilização e os benefícios do mel de abelhas sem ferrão. Material e métodos: a busca da literatura foi realizada em plataformas como Pubmed, SciELO e Portal CAPES, usando as palavras-chave “abelhas sem ferrão”, “mel” e “propriedades bioativas”. Resultados: O mel de abelhas sem ferrão possui alto teor de açúcares, baixo pH, sais minerais, vitaminas, e em menores quantidades proteínas e flavonoides. Tais compostos, conferem ao mel, atividade antimicrobiana inibitória contra bactérias do gênero Staphylococcus aureus, Bacillus sp, Proteus spp., Escherichia coli e Pseudomonas aeruginosa. Além da alta viscosidade do mel que proporciona uma barreira física em feridas capaz de evitar a exsudação e neutralizar intermediários reativos do oxigênio. Seu uso experimental em feridas contaminadas de ratos induziu a formação de granulações no tecido, estimulou o crescimento epitelial e a síntese de colágeno reduzindo o tempo de cicatrização; ademais estimulou a produção das citocinas TNF-α, IL1β e IL-6. O mel pode ser empregado no tratamento de doenças que envolvam o estresse oxidativo, uma vez que se mostrou eficaz na captura direta do radical livre DPPH e AAPH, e apresentou atividade anticancerígena inibindo a expressão da proteína inflamatória ICAM-1 e VCAM-1 em células endoteliais. Conclusão: Observa-se, com esses dados, que o mel de abelhas sem ferrão é rico em compostos bioativos que melhoram a resposta imunológica frente a bactérias patogênicas, reações inflamatórias estimulando a produção de citocinas e contribuem no tratamento e prevenção de enfermidades relacionas ao estresse oxidativo, sugerindo o desenvolvimento de novas linhas de pesquisa para avaliar sua aplicação.
