Sheep breeding has been important in agribusiness, transforming the Brazilian productive scenario. However, it is still deficient due to the damages caused by infectious diseases. Leptospirosis is a severe disease with global distribution, caused by bacteria from the Leptospira genre affecting both humans and animals. The general infection is unapparent, or its clinical signs, when present, are similar to other infections. Brucellosis is an infectious disease caused by bacteria from the Brucella genre responsible for reproductive disorders in animals, especially ruminants. The purpose of this paper was to seroepidemiological study of Leptospira spp. and Brucella ovis in sheep and dogs from nonmechanized rural properties from the northwestern region in the state of Paraná, Brazil. In order to detect anti-Leptospira antibodies, microscopic agglutination (MAT) was performed. For anti-Brucella antibodies, the agar gel immunodiffusion assay (AGID) was performed. From the total 542 samples from sheep sera analyzed, 11.25% were considered reagent to Leptospira spp. and 18.26% to Brucella ovis. From the 36 dog samples, 25% were reagent to MAT and AGID. From the 32 properties analyzed, 75% were considered positive for leptospirosis and 56.25% for brucellosis. Antibodies against the most probable serovars were Hardjo (34.42%) and Butembo (44.44%) in sheep and dogs, respectively, and the variable exchange of animals among properties was associated to leptospiric infection (p=0.028) in sheep. Leptospirosis and brucellosis are present in the sheep herd and dogs in the rural properties studied, and such result is a warning of the zoonotic importance and the need to establish sanitary programs directed to these animal species. Bacterial resistance is a primary public health concern worldwide. Within this context, pets and breeding animals act as reservoirs for multidrug-resistant bacteria (MR), such as those producing extended spectrum beta-lactamases (ESBL) and those presenting plasmid-mediated quinolone resistance (PMQR). The aim of this study was to detect the presence of ESBL and PMQR in members of the Enterobacteriaceae family, isolated from healthy sheep and dogs from non-intense farming rural properties in the Umuarama region of Paraná, Brazil. A total of 81 oral and rectal swabs from dogs and sheep from 11 small rural properties were analyzed. These swabs were inoculated in tubes containing brain heart infusion broth (BHI), and the resulting cultures were inoculated on MacConkey agar (MAC) supplemented with 10 μg/mL cefotaxime for the selection of ESBL producers. The cells were also plated on MAC supplemented with 50 μg/mL nalidixic acid for selecting quinolone-resistant enterobacteria. The bacterial isolates were subjected to biochemical identification tests, antibiograms, double-disk synergic tests, and polymerase chain reaction analysis for resistance-inducing genes (bla ESBL , qnr, and genes encoding efflux pump and acetylases). Four (5.00%) bacterial isolates (3 Escherichia coli and 1 Morganella morganii) resistant to cephalosporins and/or quinolones were identified; of these, three (75%) isolates were from sheep and one (25%) from a dog. These findings indicate the presence of MR bacteria in the normal microbiota of the animals studied. Bovine mastitis is an inflammation of the mammary gland in response to invasion by opportunistic agents. Due to the high economic importance of dairy production and the complexity related to animal health, the objective of this work was to identify and evaluate the antibacterial resistance profile of samples of mastitis milk, milking hand and milking equipment from small rural dairy farms belonging to the northwest region of the state of Paraná, Brazil. Five small, non-technical dairy farms in the municipalities of Boa Esperança, Juranda and Tapejara, all belonging to the northwest region of the state of Paraná, Brazil, were selected. The properties had Holstein and/or crossbred herds, carried out a bucket-by-foot milking system and all had the presence of animals with subclinical mastitis confirmed by the California Mastitis Test. Samples of sterile swabs from the milking insufflators, the milking hand and milk samples were collected—and later, isolation tests and phenotypic characterization of the samples, sensitivity tests to antimicrobials and phenotypic tests for the detection of beta-producing strains were performed with extended-spectrum beta-lactamase (ESBL), molecular identification of Staphylococcus aureus isolates and mecA gene research. Of the 199 samples collected from the 15 selected properties in the municipalities of Boa Esperança, Tapejara and Juranda, 72 (36.20%) were classified as multiresistant. Isolated from milkers’ hands and milking machines, which phenotypically produce extended-spectrum beta-lactamase (ESBL), the presence of the mecA gene was also observed in 11 isolates of Staphylococcus spp. of milk samples, machines and milking hands. Mastitis can be spread to the herd through the milking process by the milkers’ instruments and hands, and adequate management measures can prevent its transmission and the conscious use of antibiotics decreases the prevalence of multidrug-resistant pathogens. In this work, different pathogenic bacteria were detected in mastitic milk, milking equipment and milking hand with a high percentage (36.20%) of isolates classified as multidrug resistant. In addition, the presence phenotypically (ESBL) and molecularly (mecA gene) of isolates carrying resistance genes was also verified. These results directly reflect on the health of the animals, the health of the workers and the health of the respective environment, which can enable the continuity of the propagation of the etiological agents involved in the mastitis infection. The awareness of producers and workers on these properties about the disease, transmission, sanitary aspects and adequate management and treatment are essential for improving milk production and production efficiency.
Os Staphylococcus spp. têm sido descritos como integrantes rotineiros da microbiota do ser humano em superfície de regiões mais úmidas do corpo, enquanto algumas espécies e subespécies apresentam predileção pela região urogenital. Também são considerados oportunistas e frequentemente tem sido causa de infeção em pacientes com comprometimento do sistema imunológico ou em uso de dispositivos invasivos em pacientes hospitalizados. No entanto, nos últimos anos, houve uma disseminação dessas cepas pela comunidade, o que possivelmente pode estar relacionado a pressão seletiva pelo uso inadequado dos antibióticos. As infecções causadas por Staphylococcus spp. meticilina resistente tem causado sérios problemas dentro e fora do ambiente hospitalar. Dentro da comunidade causa colonização de pessoas aumentando a chance de óbito em todos os sexos e faixa etária e em gestantes têm sido consideradas importantes fontes de microrganismos patógenos pelos fatores de virulência apresentados por essa bactéria e pelas condições fisiológicas e anatômicas da gestante, tornando-se, dessa forma, uma ameaça de propagação de infecções por cepas multirresistentes.
