The effects of trans-resveratrol (1) were evaluated in acute nociception models induced by capsaicin or glutamate in mice, in an attempt to further characterize its mechanism of action. The oral administration of 1 (50 and 100 mg/kg) reduced significantly the licking behavior elicited by capsaicin (1.6 μg/paw) or glutamate (10 μmol/paw). The co-administration of 1 into the mouse paw (200 μg/site) markedly prevented glutamate-induced licking, without affecting capsaicin responses. In addition, the intrathecal (it) injection of 1 (150 to 600 μg/site) greatly reduced the licking behavior caused by capsaicin, but not glutamate. Similarly, the intracerebroventricular injection of 1 (300 μg/site) caused a potent inhibition of capsaicin-induced nociception, while the glutamate responses remained unaffected. However, the co-administration of 1 (300 μg/site) reduced the biting behavior induced by spinal injection of glutamate (30 μg/site, it), leaving capsaicin (6.4 μg/site)-induced biting unaltered. Notably, the oral administration of 1 (100 mg/kg) inhibited significantly the capsaicin-induced increase of c-Fos and COX-2 labeling in the spinal cord and COX-2 expression in the cortex, but failed to affect c-Fos and COX-2 expression in the glutamate model. This study has explored the effects of 1 in both the capsaicin and glutamate models, extending current knowledge on the analgesic effects of trans-resveratrol.
INTRODUÇÃO: A terapia antirretroviral combinada (TARV) foi introduzida no Brasil em 1996, como parte da política nacional de acesso gratuito aos serviços de saúde e medicamentos. Infelizmente, o seu uso contínuo tem sido associado com mudanças na distribuição da gordura corporal e com alterações metabólicas que podem aumentar a morbidade e mortalidade nesta população. O treinamento físico tem sido estudado como uma estratégia eficaz de intervenção não farmacológica para melhorar os parâmetros de aptidão física relacionados à saúde e para minimizar os efeitos indesejáveis da infecção pelo HIV e/ou o uso prolongado da TARV, no entanto, há poucos estudos sobre o treinamento físico, síndrome lipodistrófica e cardiologia.OBJETIVO: Avaliar o risco de doença isquêmica cardíaca em sujeitos HIV/AIDS em uso de TARV praticantes de treinamento concorrente com séries simples.MÉTODOS: Quatorze sujeitos foram avaliados através da circunferência abdominal, pressão arterial sistólica (PAS) e diastólica (PAD), colesterol total (CT), HDL, LDL, triglicerídeos (TG) e glicemia. Para a estimativa do risco coronariano em 10 anos utilizou-se o Escore de Framingham.RESULTADOS: A maioria dos sujeitos situou-se dentro dos valores de referência para as variáveis analisadas, exceto para os valores de LDL e TG. Treze sujeitos (92,7%) ficaram abaixo dos 10% de risco coronariano em 10 anos, e apenas um (7,3%) estava em risco moderado. Houve correlação significativa entre o tempo de treinamento e a PAS.CONCLUSÃO: Sugere-se a realização de ensaios clínicos randomizados para avaliar os mesmos desfechos deste estudo.
Objectives: The present study has tried to analyze the individual and combined expression of the three markers-p53, Ki-67 and bel-2-in HPV associated pre-malignant lesions of the cervix. Methods: Thirty-four cases of high-grade lesions, 21 low-grade cases and 18 normal cases were histologically assessed. HPV-DNA presence and the viral types were determined through PCR and RFLP, respectively. Marker expression was performed by immuno-histochemistry. Results: With regards to viral types, HPV with high oncogenic potential represented the majority, with just one case reporting the low oncogenic potential for viral infection (HPV 6). HPV was the most frequent and could be associated with the high-grade lesion group, however, no significant associations were found between the viral ones and the marker expression. When the expression location, beyond the basal layer, was taken into account, the three markers proved to be good indicators of the lesion grade. The markers' combined expression: p53(−), Ki-67(+), bcl-2(−) and p53(−), Ki-67(−), bcl-2(−) was statistically significant when associated with the lesion grade. Conclusions:The results suggest that the p53, Ki-67 and bel-2 markers can contribute to differentiate the lesion grades, whether in its isolate form or in combined form.
Background: Metallurgical workers are a high-risk population as a function of their occupational exposure to toxicants, such as heavy metals, aromatic hydrocarbons and silicate. Objective: To investigate the occurrence of biological effects among workers exposed to toxicants in various areas of a metallurgical company in Caxias do Sul, Rio Grande do Sul, Brazil. Methods: Cross-sectional study with 31 workers from 10 areas who responded a questionnaire on occupational and lifestyle habits. Blood and toxicology tests were compared to the questionnaire results. Results: About 19.4% (n=6) of the sample did not use all the required personal protective equipment (PPE). Reduction of red blood cell parameters and leukocyte abnormalities were found, including leukocytosis, neutrophilia and eosinophilia, as well as in the blood lead levels. The following significant associations were found on analysis per work area: smoking and neutrophilia (p=0.000); work in the mold section and leukocytosis (p=0.023) and eosinophilia (p=0.007); and more than 10 years of work and leukocytosis (p=0.008). Conclusions: The data suggest that length of work, type of exposure and use of PPE might cause changes in the results of laboratory tests. RESUMO | Introdução: Metalúrgicos compõem uma população de risco pela sua exposição ocupacional a toxicantes, como metais pesados, gases, hidrocarbonetos aromáticos e silicato. Objetivo: Avaliar a existência de efeitos biológicos em trabalhadores expostos a toxicantes de vários setores de uma metalúrgica de Caxias do Sul, Rio Grande do Sul. Método: Foi realizado um estudo transversal com 31 trabalhadores de 10 setores em que foi aplicado um questionário sobre hábitos ocupacionais e de vida. Exames hematológicos e toxicológicos foram comparados com os resultados do questionário. Resultados: 19,4% (n=6) dos trabalhadores não usaram todos os equipamentos de proteção individual (EPIs). Foi possível observar diminuição dos parâmetros do eritrograma e alterações na série branca, como leucocitose, neutrofilia e eosinofilia, e no chumbo sérico. Evidenciaram-se valores diminuídos na série vermelha e leucocitose dos trabalhadores que não faziam uso adequado dos EPIs. Quando avaliado o setor de trabalho, pôde-se evidenciar as seguintes associações significativas: fumantes e neutrofilia (p=0,000); setor da moldagem e leucocitose (p=0,023) e eosinofilia (p=0,007); mais de 10 anos de trabalho e leucocitose (p=0,008). Conclusões: Os dados sugerem que o tempo de trabalho, o tipo de exposição e o uso de EPIs podem alterar exames laboratoriais. Introdução: As investigações sobre treinamento físico com pessoas que vivem com HIV/Aids enfatizam os componentes aeróbico, força e concorrente em séries múltiplas. Objetivo: Avaliar o treinamento concorrente com séries simples nos parâmetros imunológico, virológico, cardiorrespiratório e muscular em pacientes com HIV/Aids. Métodos: Séries temporais, com a participação de 6 homens e 1 mulher que faziam uso da TARV. O parâmetro imunológico foi avaliado por citometria de fluxo; o virológico, pelo teste HIV-1 RNA 3.0 (bDNA); o cardiorrespiratório, por meio do protocolo de rampa e o muscular, pelo teste de 15 repetições máximas e número máximo de flexões de tronco em 1 minuto. O treinamento foi realizado três vezes por semana durante 12 semanas, com reavaliações e incrementos mensais. A estatística contemplou valor absoluto, mediana e amplitude e o teste t de Wilcoxon no programa SPSS, versão 20.0 (p < 0,05). Resultados: No parâmetro imunológico, houve aumento do número de linfócitos T CD4+ (p = 0,034). No parâmetro virológico, a carga viral em 2 participantes diminuiu (495 para 51 cópias/ml e 72 para não detectável [< 50 cópias/ml]) e 5 mantiveram-na não detectável. No cardiorrespiratório verificou-se aumento do VO2máx absoluto (p = 0,028) e da carga máxima de trabalho (Watt) no cicloergômetro (p = 0,015). A carga de trabalho aumentou na função muscular de todos os exercícios: voador (p = 0,018), roldana alta (p = 0,017), pressão de pernas (p = 0,018), roscas bíceps (p = 0,016) e tríceps (p=0,017) e número máximo de abdominais (p = 0,018). Conclusão: O treinamento melhorou os parâmetros cardiorrespiratório e muscular e não causou efeitos deletérios nos parâmetros imunológico e aumento na carga viral.
