RÉSUMÉCe travail a pour objectif d'établir les conditions optimales de régénération de plantes fertiles à partir de suspensions cellulaires embryogènes en vue de leur utilisation pour une transformation génétique par bombardements de protoplastes. Ce travail réalisé sur des variétés de Pennisetum glaucum de Côte d'Ivoire a permis de mettre en évidence les facteurs qui influent (taille et âge des cals, milieu de conditonnement, et âge de la suspension cellulaire) sur la régénération de plantes fertiles. Il ressort de cette étude que la taille des cals et les milieux de conditionnement utilisés n'ont aucun effet sur le taux de régénération de plantes fertiles. Cependant, l'âge de la suspension cellulaire obtenue à partir de cals embryogènes et l'âge du cal initiateur de la suspension cellulaire influent sur le taux de régénération. Le taux de plantes normales régénérées est élevé durant les deux premiers mois de culture de la suspension cellulaire initiée à partir des cals de 2-3 mois (60 à 70%). Les plantes régénérées présentent les mêmes caractéristiques de croissance végétative (taille, longueur de la chandelle, nombre de barres, longueur de l'épi) que les plantes témoins. Pour le rendement, les plantes régénérées portent moins de grains (412) que les plantes témoins (715).
ABSTRACT.Many problems were kipt out with immature embryos and shoot explants pieces used ; principally : Calli were initiated in plant flowering periode (every 110 days), and the low percentage of embrogenesis what either the source used. So for Pennisetum glaucum, new embryogenic calli sources were explored from calli initiation to fertile plant regeneration achieving. About 90 % of dissected shoot apices, seedlings and seeds developed calli. From meristems position, 6 mm of shoot and 20 mm of root developed 50 to 100 % of calli. No embryogenic calli were obtained from root. More than 80 % of dissected apices led to embryogenic calli. Maintained on MS(1. 1. 2,5), MS(5) and N6(1.100.25) culture media, calli embryogenic potential and fertile plants regeneration were conserved for more than 12 months. Characteristics of regenerated plants were similar to control. REGENERATION DE PLANTE A PARTIR DE LA CULTURE DE TISSUS DE MIL (Pennisetum glaucum (L) R.)
Les difficultés liées à utilisation de l'embryon immature et des explants de tige dans les cultures de tissus sont nombreuses ; dont l'initiation
L'activité parentale, surtout maternelle et sa régulation ne sont pas l'apanage des seuls vertébrés. Chez les arachnides, les exemples abondent. C'est le cas de Coelotes terrestris, une araignée terricole de la famille des Agelenidae qui nourrit ses petits pendant au moins quatre semaines. Mais compte tenu des difficultés d'observation en milieu naturel, une étude expérimentale a été menée en laboratoire dans le but de comprendre les interactions entre la femelle et ses jeunes. Ainsi, cinq catégories de femelles ont été mises en confrontations forcées avec des jeunes homospécifiques. Les interactions de ces confrontations ont été enregistrées et analysées. Les résultats montrent qu'en présence des jeunes, l'expression des « sursauts » (SST) traduit vraisemblablement une attitude agressive des femelles vis-à-vis des jeunes car, ce sont les femelles en préémergence (pré-ponte, incubante et sub-adulte) connues pour être agressives ou "tueuses" de jeunes qui présentent significativement la manifestation de ce comportement. Par contre, l'activité de « tissage » (TIS) peut être qualifiée de « comportement maternel ». Son expression semble traduire une certaine tolérance et/ou reconnaissance des femelles à l'égard des jeunes. En effet, ce sont les femelles en post-émergence (nourricière et post-dispersion) déjà montrées comme les plus tolérantes qui posent significativement cet acte (de tissage).
