Cultured keratinocytes are frequently employed for studies of epidermal lipid metabolism. Interpretation of experimental data may be complicated by donor to donor variability, the relatively short culture lifetime and variations between passages, problems that are not encountered to the same extent with immortalized cell lines. The present study was undertaken to compare the lipid composition and synthesis of normal human adult keratinocytes (NHAK) with HaCaT cells, a long-lived, spontaneously immortalized human keratinocyte line, in relation to proliferation and differentiation. No differences between the two cell types were observed: a) in total lipid content; b) in the distribution of major lipid classes during growth at 50%, 75% and 100% confluence; c) in cultures grown at 0.6 mM calcium, at which differentiation is retarded, or at 1.6 mM calcium, at which some differentiation takes place; d) in the incorporation of [14C] acetate into cellular lipids at confluence, or e) in the fatty acid composition of major cellular lipid classes. At 100% confluence NHAK and HaCaT cells differ in their cholesterol metabolism. At all stages of growth, cholesterol synthesis in HaCaT cells is more LDL-dependent than in NHAK. Furthermore, NHAK become less LDL-dependent at confluence whereas HaCaT cells do not. HaCaT cells also revealed a significantly larger fraction of phosphatidyl-ethanolamine, -serine and -inositol at 0.6 mM calcium concentration than NHAK. These findings suggest that HaCaT cells do not differentiate as well as NHAK in vitro and may therefore serve as a model for the study of lipid metabolism in cells defective in terminal differentiation.
There is a need in many studies on the epidermal permeability barrier for practical and reliable separation of all major fractions of human stratum corneum lipids. Various methods have been described including thin layer chromatography, high performance thin layer chromatography and iatroscan. However, none of these methods seems to be applicable for an inexpensive, rapid and reliable analysis of a large number of samples. Here, such a method for the separation and quantification of all major stratum corneum lipid fractions is presented. This method employs the one-dimensional separation of stratum corneum lipids using thin layer chromatography. The systems used herein are based upon various modifications of the solvents, solvent ratio and developing distance of each system. For quantification of chromatographed and charred lipids a Desaga densitometer is used. The system described here had been successfully applied to 550 samples of human stratum corneum lipids.
Little is known about the aetiology and pathogenesis of the different types of inherited and acquired palmoplantar keratodermas. We describe a condition of painful palmoplantar keratoderma with an altered stratum corneum lipid pattern which may be responsible for the excessive cornification. Plantar stratum corneum lipids were analysed by quantitative thin-layer chromatography. Serum lipids, and the activities and gene loci of the enzymes serum steroid sulphatase and arylsulphatase C were also determined. Examination revealed that both the stratum corneum and the serum cholesterol sulphate (CS) content were significantly elevated in comparison with the stratum corneum cholesterol ester content. The cholesterol content was unchanged compared with controls. Serum activities of steroid sulphatase and arylsulphatase C were decreased, but not to the extent found in recessive X-linked ichthyosis. Their gene loci did not show any deletions. This unique distribution of stratum corneum sterol derivatives, reflected by the elevated serum CS concentration, may contribute to the altered structural and functional properties of intercellular lipid lamellae within the stratum corneum of this type of keratoderma.

Es wird angenommen, daß bei den ü ber 60 jä hrigen Frauen mehr als 70 % mindestens eine behandlungsbedü rftige Dermatose aufweisen. Die Erkrankungen reichen von ausschließlich kosmetischen Beeinträ chtigungen wie Faltenbildung durch Elastizitä tsverlust ü ber schweren Pruritus bis zur Entstehung maligner Melanome. Mit steigendem Alter nehmen wegen chronischer Sonnenexposition besonders die Lichtschä den in Form der Elastose mit leichterer Verletzlichkeit der Haut sowie benigne und maligne Neoplasien zu. Hä ufig werden erste Anzeichen einer Hauterkrankung, insbesondere Frü hzeichen von malignen Neoplasien der Haut, von den Patienten nicht erkannt oder beachtet. Um die Prä vention und rechtzeitige Behandlung von Dermatosen zu verbessern, sollen die charakteristischen Verä nderungen der alten Haut dargestellt werden und ein Ü berblick ü ber die Klinik der wesentlichen Dermatosen der ä lteren Haut gegeben werden.Die Haut ist das Organ, welches dem Altern am sichtbarsten unterworfen ist. Der physiologische Alterungsprozeß der Haut setzt mit etwa 30 Jahren ein und schreitet individuell unterschiedlich schnell fort, abhä ngig von zahlreichen endogenen und exogenen Faktoren. Die endogene Form der Hautalterung findet sich nur an streng lichtgeschü tzten Kö rperregionen. Sie entspricht der biologischen oder "Zeitalterung" der Haut. Bei regelmä ßigem Einfluß exogener Faktoren auf die Haut kommt es zur sog. "Umweltalterung", wobei das Sonnenlicht den grö ßten Einfluß hat. Man geht davon aus, daß 90 % des Umweltalterns der Haut durch die UV-Strahlung verursacht wird und spricht deshalb von der "Lichtalterung" der Haut. Zeitalterung der HautDie klinischen Kennzeichen der Zeitalterung der Haut sind wohlbekannt und umfassen Schlaffheit, Elastizitä tsverlust, Faltenbildung, Trockenheit und fleckige Pigmentverschiebungen. Die verschiedenen Strukturen der Haut sind unterschiedlich stark betroffen, wobei die Oberhaut oder Epidermis meist bis ins hohe Alter eine weitgehend konstante Zahl an Zellschichten aufweist. Die wesentlichen Verä nderungen betreffen vor allem die Unterhaut oder Dermis sowie die dermoepidermale Grenzzone [10]: Es findet sich eine Abflachung der Grenzzone mit verminderter Verankerung zwischen Dermis und Epidermis. Die Unterhaut ist ebenfalls verdü nnt mit reduzierter Zell-und Gefä ßdichte.
