-Demands for sustainable agricultural systems have forested the development of biological control techniques, such as the use of the egg parasitoid Trichogramma Westwood. However, the arduous identification of the parasitoid at the species level, due to the tiny size and the morphological similarities is an obstacle to increasing its use. Molecular markers are useful to reach the specimen genome and avoid environmental effects that could misguide identification. Several molecular markers techniques are available and the ISSR technique has been used to differentiate close individuals, due to its high polymorphism level, reproducibility and low cost. The objective of this study was to measure the level of genetic differentiation among five lines of Trichogramma, using ISSR markers: three belonging to the species T. pretiosum Riley, one to T. atopovirilia Oatman & Platner and one to T. bruni Nagaraja. Morphological identification of the parasitoids was conducted at ESALQ/USPPiracicaba, SP. After DNA removal and standatization, PCR reactions were performed with 26 ISSR primers; 11 of them were selected because they presented greater polymorphism and consistency. Molecular data were converted into a binary matrix and analyzed (NTSYS v. 2.1). The 11 primers produced 172 polymorphic sections. Genetic similarity ranged from 19% to 96%, showing that the ISSR technique can efficiently identify DNA polymorphism in Trichogramma. Results also indicate important inter and intra-specific variations among the parasitoid lines.KEY WORDS: Molecular biology, microhymenopteran, identification, biological control RESUMO -A demanda por processos agrícolas racionais, em detrimento da utilização de estratégias convencionais, tem contribuído para o desenvolvimento de técnicas de controle biológico, como a utilização de parasitóides de ovos do gênero Trichogramma Westwood. Um entrave para sua utilização é a dificuldade na identificação das espécies, devido ao diminuto tamanho e à similaridade morfológica. Os marcadores moleculares acessam o genoma, evitando o efeito ambiental e conseqüentemente erros de identificação. Várias técnicas estão disponíveis e a técnica ISSR vem sendo empregada para a diferenciação rápida entre indivíduos próximos, devido ao elevado grau de polimorfismo, reprodutibilidade e baixo custo. RESUMOAs temperaturas baixas que ocorrem durante o cultivo do arroz na Região Sul do Brasil causam prejuízos ao rendimento e à qualidade dos grãos de arroz. Genótipos tolerantes a baixas temperaturas seriam uma alternativa para minimizar tais prejuízos. Desse modo, o presente trabalho teve como objetivo avaliar a tolerância ao frio em famílias mutantes de arroz (geração M 4 ) nos períodos, vegetativo e reprodutivo, como forma de identificação de variabilidade genética para o caráter. Para a avaliação das famílias mutantes quanto à tolerância ao frio, no período vegetativo, foram utilizadas 10 sementes de cada família colocadas em bandejas celuladas, sendo as plântulas mantidas em casa de vegetação à temperatura de 13 ºC por 10 dias em sala climatizada, com fotoperíodo de 12 horas. A avaliação do período reprodutivo consistiu na aplicação de frio na antese, sendo caracterizada por um estresse de 15 ºC por sete dias. Pelos resultados verificou-se que no período vegetativo, seis famílias tiveram 0-20% de redução na estatura, com um elevado grau de tolerância. ABSTRACT CHARACTERIZATION OF RICE FAMILIES FOR COLD TOLERANCE IN THE VEGETATIVE AND REPRODUCTIVE PHASESLow temperatures that occur during the cultivation of rice in the Southern region of Brazil cause reductions in to the yield and quality of the rice grain. Genotypes tolerant to low temperatures would be an alternative to minimize these problems. In the present work cold tolerance was evaluated in rice families (M 4 generation) in the vegetative and reproductive phases, in order to identify possible genetic variability. For the evaluation in the vegetative stage, 10 seeds from each family were placed in cell trays and kept at 13 ºC for 10 days in controlled chamber with 12 hour day light. The evaluation at the reproductive stage consisted of the application of low temperature at anthesis, by a stress of 15 ºC for 7 days. The results suggest that in the vegetative phase, 6 families presented 0-20% reduction in stature, showing a high degree of tolerance. For the reproductive phase there were no families showing higher tolerance than the tolerant control.
RESUMOO presente trabalho teve por objetivo verificar o efeito de diferentes períodos de luz ultravioleta sobre a similaridade genética do fungo simbionte de formigas cortadeiras pertencentes às espécies Acromyrmex ambiguus, Acromyrmex crassispinus e Acromyrmex lundi, através da técnica AFLP. Os fungos foram inoculados em meio de cultura Pagnocca e mantidos em câmaras climatizadas à temperatura de 25 + 1ºC e escotofase de 24h, por um período de 15 dias. Posteriormente, foram expostos à luz ultravioleta por períodos de: 0, 5, 10, 15, 20, 25 e 30 minutos. Aos 45 dias após o tratamento, foi realizada a extração do DNA e padronização, realizando-se as reações de PCR por meio de 24 combinações de primers AFLP, das quais quatro foram selecionadas por apresentarem maior polimorfismo e consistência. Os dados moleculares foram transformados em matriz binária e analisados pelo programa estatístico NTSYS v. 2.1. As quatro combinações de primers utilizadas geraram 152 bandas polimórficas. As populações de fungos originárias de diferentes espécies de Acromyrmex apresentaram uma similaridade genética média de 37%, sendo que a indução de mutação através da luz ultravioleta permitiu obter isolados mais dissimilares. ABSTRACT The present research was aimed at verifing the effect of different periods of ultraviolet light on the development of the symbiont fungus of leaf cutting ants belonging to the species Acromyrmex ambiguus, Acromyrmex crassispinus andAcromyrmex lundi, through the AFLP technique. The fungus were inoculated in Pagnocca's medium and maintained in climatized cameras at 25 + 1ºC and 24 hours of darkness, for a period of 15 days. After that, they were exposed to ultraviolet light for 0, 5, 10, 15, 20, 25, and 30 minutes. At 45 days after the treatment, the extraction and standardization of DNA was performed. PCR reactions were tested using 24 AFLP primer combinations, from which four were selected because they presented larger polymorfism and consistence. The molecular data were transformed in binary mattrix and analyzed by the statistical program NTSYS v. 2.1. The four combinations of primers generated 152 polymorfic bands. The fungus populations from different Acromyrmex species presented a medium genetic similarity of the 37%, and the mutations induced by the ultraviolet light allowed to obtain more dissimilar isolates.
