This work aimed at evaluating the effects of the supplementation of starter diet with Arg on breast muscle development in broilers and the activation of satellite cells and the aggregation of myofibrillar protein. Male Cobb chicks (n = 990) were randomly assigned to 1 of 5 treatments in a complete random design. Measurements of 33 chicks per treatment were made in 6 repetitions. The treatments consisted of a basal diet with 1.390% digestible Arg (without supplementation) and 4 dietary levels of Arg (1.490, 1.590, 1.690, and 1.790%) with Arg:Lys ratios of 1.103, 1.183, 1.262, 1.341, and 1.421, respectively. Arginine supplementation was used only in the starter phase (1 to 21 d). Dietary supplementation with Arg had a positive effect (P < 0.05) on breast and breast fillet weight on d 7 and 21 and on myofiber diameter on d 14 and 21. However, no effect was observed (P > 0.05) on the protein:DNA ratio, which demonstrates that Arg does not interfere with the mitotic activity of the satellite cells. Independently from mechanism, Arg affected muscle growth in the starter phase positively. Dietary supplementation with Arg in the starter phase had no effect (P > 0.05) on the carcass yield of broilers on d 42. Diet supplementation with Arg at levels above the ones recommended for the starter phase may be necessary for improved muscle development in broilers.
This trial aimed at evaluating the effect of organic trace mineral supplementation of commercial layer diets on productive performance and egg quality. One-hundred-ninety-two Hy Line W36 white 69-w-old layers were distributed into a completely randomized design with three treatments, and eight replicates, with eight birds each. Treatments consisted of a basal diet supplemented with inorganic trace minerals (R1), and two others experimental diets containing 0.250 ppm (R2) and 0.500 ppm (R3) of an organic source of zinc, manganese, and selenium. Feed intake (g/bird/day), feed conversion ratio (kg/dozen egg and kg/kg egg), egg weight (g), egg production (%), thin and cracked eggshells (%), specific gravity (g/mL), Haugh Units, total egg solids (%), yolk yield, white and shell yields (%), eggshell thickness, and egg Se content were evaluated Tukey's test analyzed differences among means at 5% of probability using PROC GLM in SAS (2000). Although not significant as compared to the non-supplemented diet, improvements on relative cracked-plus-thin shells were observed with the use of organic mineral blend. The addition of the organic blend to the diet at 0.250 kg/ton resulted in (p<0.05) higher total egg solids. Also, as compared to eggs from control group, fresh and dried yolk yields were higher with the dietary inclusion of the organic mineral blend at 0.250 and 0.500 kg/ton
Intense bone remodeling takes place in both subchondral and trabecular bone in the knee following ACL transection. This process seems to occur around week 12, although Oc and NO appeared to be involved earlier at 8 weeks. These results suggest that not only subchondral but also trabecular bone metabolism is altered in this OA model.
Though used occasionally, systemic therapies in severe childhood psoriasis have not been systematically investigated. Cyclosporin A (CysA) is effective in adults with severe psoriasis but there are no extensive data regarding the efficacy and safety of its use in childhood psoriasis. In this paper, we describe six children aged between 11 months and 13 years (average: 7.6 years) treated with CysA microemulsion formulation for severe psoriasis, who had been unresponsive to other treatments. The CysA dose ranged from 2 to 4 mg/kg/day, for periods varying from 8 to 105 weeks (mean: 54 weeks). Dose tapering was gradual after lesion improvement and adjusted according to clinical response. Adjuvant therapy with topical steroids, vitamin D3 ointments, coal tar preparations or anthralin was used in all children. Acitretin was used in three patients for short periods. The children were regularly monitored for serum renal and liver function and blood pressure. Improvement of skin lesions was achieved after between 4 and 30 (mean: 12) weeks of treatment, with complete remission in three children. Relapse of lesions occurred in the other children during CysA reduction, but they responded to a dose increase. The treatment was found to be well tolerated and with no significant side-effects. CysA can be used in carefully selected and monitored patients and may represent an alternative tool for severe episodes of psoriasis in children, when other therapies are unsuccessful.
This study aimed at evaluating the influence of Glutamine (Gln) and Vitamin E (VE) supplementation on the performance and immune response of broilers. A completely randomizes experimental design with a 2 x 3 (VE x Gln) factorial arrangement was used. VE was supplemented at 10 and 500 mg/kg feed, with or without Gln (1%) addition, and two periods of supplementation in the starter diets (1-7 and 1-14 days of age), with five replicates of 50 birds each. The analyzed parameters were: live performance (weight gain, feed intake, and feed conversion ratio); relative weights of the spleen, bursa, and thymus; antibody titers (with sheep red blood cells suspension -SRBC) and cutaneous basophilic hypersensivity (CBH). Data were submitted to the analysis of variance, and means were compared using the test of Tukey. Treatments did not influence (P>0.05) live performance parameters or antibody titers. VE reduced (P=0.01) CBH, with the level of 10 mg VE/kg allowing higher cell proliferation as compared to 500 mg VE/kg. As to lymphoid organs, only the spleen was affected (P=0.035) by Gln, which resulted in higher spleen relative weight when fed during the first week of age. Results showed that 10 mg VE/kg with Gln (1-7 days) promoted better immune responses.
ABSTRACT. Due to the lack of a complete urea cycle, uricotelic species, such as broilers, are not able to synthesize de novo arginine (Arg), thus depending exclusively on dietary Arg. High levels of dietary lysine (Lys) increase the demand for Arg because of the antagonistic relationship between these amino acids. The Arg-Lys antagonism promotes an expressive increase in the renal Arg activity and consequently induces the degradation of Arg and the decrease in the activity of glycine amidinotransferase, an enzyme that uses Arg in the synthesis of muscle creatin. Arg is considered an important modulator of immunological and physiological processes. The degradation of Arg produces ornithine, a precursor of polyamines that are key to cell division, DNA synthesis, and cell cycle regulation. Arg participates in the synthesis of nitric oxide (NO), a highly reactive free radical in cells and membranes and participates in several cell processes, including in neurotransmission and immune response. Arg is also considered a potent secretagogue of insulin, growth hormone, and IGF-I in the blood stream. Exclusively vegetarian diets may not provide an adequate supply of Arg, which is required for maximum production and for the immune system of current broiler lineages.Key words: carbamoyl phosphatase synthetase, creatine, immune system, nitric oxide, polyamines, urea cycle.RESUMO. Devido à falta de um completo ciclo da uréia, espécies uricotélicas como os frangos de corte são incapazes de sintetizar arginina (Arg) de novo, por isso dependem exclusivamente da Arg dietética. Níveis elevados de lisina (Lys) dietética aumentam a exigência de Arg devido à relação antagônica entre estes aminoácidos. O antagonismo entre Arg e Lys promove expressiva elevação da atividade da arginase renal e conseqüentemente induz à degradação da Arg e leva à diminuição da atividade da glicina amidinotransferase, enzima que utiliza Arg na síntese de creatina muscular. Arg é considerada importante modulador da imunidade e de processos fisiológicos. A degradação de Arg gera ornitina, precursor das poliaminas que têm papel-chave na divisão celular, síntese de DNA e regulação do ciclo celular. A Arg é utilizada na síntese de óxido nítrico (NO), um radical livre altamente reativo, permeável às células e membranas que participa de vários processos celulares, incluindo a neurotransmissão e a imunidade. A Arg é também considerada um potente secretagogo da insulina, hormônio do crescimento e IGF-I. This study investigated the effects of arginine (Arg) supplementation on broiler breeder egg production and egg quality. Male (30) and female (360) Ross ® breeders, totaling 390 birds, were studied. A completely randomized design with five Arg levels (0.943%, 1.093%, 1.243%, 1.393%, 1.543% digestible Arg) and six replicates of 12 females and one male per experimental unit was applied. The following performance and egg quality were evaluated: lay percentage, albumen and yolk contents, average egg weight, egg specific gravity, and eggshell percentage and thickness. Hatchability and and embryo mortality were also determined. The applied dietary digestible Arg levels quadratically influenced egg production (p<0.05), with the highest production obtained when 1.262% digestible Arg was supplemented. Egg weight linearly increased (p<0.05) with digestible Arg dietary level; however, egg specific gravity linearly decreased (p<0.05). Hatchability was not affected (p<0.05) by digestible Arg level. The supplementation of broiler breeder diets with Arg improved egg production and egg weight without any effect on hatchability. Further research is needed to determine the effect of dietary Arg supplementation on the performance of the progeny.
The effects of starter diet (days 1 to 21) supplemented with arginine (Arg) on the production performance and duodenum and jejunum mucosa morphometry of broilers were studied. Male Cobb broiler chickens (990) were randomly assigned to one of ϐive treatments in a complete random design. Measurements of 33 chicks per treatment were made in six repetitions. The treatments consisted of a basal diet with 1.390% digestible Arg (no supplementation) and four dietary levels (1.490%, 1.590%, 1.690%, and 1.790%), providing a relationship with lysine of 1.103; 1.183; 1.262; 1.341 and 1.421%, respectively. From the age of 22 days on, all birds received conventional grower diet. The data were submitted to regression analysis by polynomial decomposition of the degrees of freedom in relation to the levels of Arg. The Arg supplementation increased (P<0.05) the live weight and the feed conversion ratio without increasing the feed intake of the birds. However, no effect was observed (P>0.05) in the growth phase (days 22 to 42) in the absence of the Arg supplementation. The supplementation of Arg over of NRC recommendation during the starter phase may be necessary for the expression of the maximal weight gain potential in birds. No effect (P<0.05) of Arg dietary supplementation was observed either on small intestine weight and length at any age. However, the duodenum villus:crypt ratio increased and the crypt depth decreased in the ϐirst week in response to increasing dietary Arg. It is concluded that broiler Arg dietary supplementation in the starter diet improved production performance and small intestine morphometry, especially in the ϐirst week.
