Resumo -O objetivo deste trabalho foi avaliar o efeito de concentrações de extrato de casca de café, óleo essencial de tomilho e acibenzolar-S-metil na germinação, no crescimento micelial e no desenvolvimento in vivo de Cercospora coffeicola, além de caracterizar a efi ciência deles como indutores de resistência, e determinar a atividade da enzima peroxidase e o acúmulo de lignina nos tecidos de cafeeiro. O extrato de casca de café não afetou a germinação, entretanto, inibiu o crescimento micelial proporcionalmente ao aumento das concentrações. O óleo essencial de tomilho inibiu a germinação e o crescimento micelial com o aumento das concentrações. O extrato de casca de café e o acibenzolar-S-metil não afetaram a germinação nem o desenvolvimento dos tubos germinativos, diferentemente do óleo essencial de tomilho. Mudas tratadas com acibenzolar-S-metil, extrato de casca de café e óleo essencial de tomilho, apresentaram picos de atividade da peroxidase aos 2 e 11, 7 e 11 e, 2 e 9 dias após a aplicação dos tratamentos, respectivamente. Os tratamentos não diferiram quanto à concentração de lignina. Extrato de casca de café e acibenzolar-S-metil induziram resistência em mudas de cafeeiro contra C. coffeicola e o óleo essencial de tomilho apresentou efeito tóxico ao patógeno.Termos para indexação: Cercospora coffeicola, Coffea arabica, indução de resistência, microscopia eletrônica de varredura. Coffee berry husk extract, thyme essential oil and acibenzolar-S-methyl in the control of brown eye spot of coffee treeAbstract -The objective of this work was to assess the effect of the coffee berry husk extract, thyme essential oil and acibenzolar-S-methyl on the germination and micelial growth and on in vivo development of Cercospora coffeicola, and to characterize their effi ciency as resistance inducers in coffee plants, and to determine the peroxidase activity and lignin accumulation in tissues of coffee tree. The coffee berry husk extract presented no toxic effect on germination; however, it inhibited the mycelial growth proportionally to the increase of the concentrations. The thyme essential oil inhibited the germination and the mycelial growth with the increase of concentrations. Both coffee berry husk extract and acibenzolar-S-methyl neither affected the germination nor the development of germinative tubes, differently of thyme essential oil. Seedlings treated with acibenzolar-S-methyl, coffee berry husk extract and thyme essential oil presented peaks of peroxidase activity at 2 nd and 11 th , 7 th and 11 th and, 2 nd and 9 th days after treatment, respectively. The treatments did not differ in lignin content. Coffee berry husk extract and acibenzolar-S-methyl induced resistance in coffee seedlings against C. coffeicola, and thyme essential oil presented toxic effect on the pathogen.
RESUMONos estudos envolvendo a interação Colletotrichum gloeosporioides-cafeeiro (Coffea arabica L.), poucas são as informações a respeito do modo de penetração e colonização deste patógeno. Estudou-se por meio da microscopia eletrônica de varredura (MEV), os eventos de pré-penetração e penetração de C. gloeosporioides em hipocótilos de cafeeiros e a colonização natural de ramos e frutos provenientes de plantas com mancha manteigosa. Realizaram-se três ensaios. No primeiro, fez-se a observação dos eventos de pré-penetração de C. gloeosporioides inoculados em hipocótilos de plântulas da cv. Acaiá Cerrado com e sem ferimentos; no segundo, observou-se a colonização de ramos enfermos em que havia murcha drástica e necrose local e, no terceiro, a colonização de frutos enfermos. A germinação dos conídios nos hipocótilos feridos ocorreu 6 horas após a inoculação, com a formação de um ou dois tubos germinativos terminais e a adesão dos conídios nas depressões dos hipocótilos. Apressórios globosos a subglobosos e de contorno regular surgiram 12 horas após a inoculação. Até 72 horas, não foi possível observar a formação de acérvulos sobre os tecidos submetidos à inoculação. Nos ramos observavase colonização nos vasos floema e parênquima cortical. Nos frutos em diferentes estádios de maturação foi observada a colonização do tecido próximo à epiderme e colapso dos estômatos na área lesionada.Palavras-chave: Glomerella cingulata, Coffea arabica, mancha manteigosa, microscopia eletrônica de varredura. ABSTRACT HISTOPATHOLOGICAL STUDIES OF THE INTERACTION COLLETOTRICHUM GLOEOSPORIOIDES -COFFEE TREESIn studies involving the interaction Colletotrichum gloeosporioides -coffee trees (Coffea arabica L.) there is little information regarding the pre-penetration, penetration and colonization pathways of this pathogen. The objectives of this work were: 1. to study through scanning electron microscopy (SEM) the pre-penetration and penetration events of C. gloeosporioides in hypocotyls of coffee plantlets; 2. to observe the colonization of Colletotrichum spp. in branches and fruits of coffee trees with blister spot symptoms. The study took place in three trials. In the first trial, it was observed and reported the pre-penetration events of C. gloeosporioides inoculated on hypocotyls of coffee cv. Acaiá Cerrado with and without wounds. The second trial was performed to investigate the colonization of diseased coffee branches that presented drastic wilt and local necrosis was observed. Finally the third trial investigated the colonization of this pathogen in diseased coffee fruits. When C. gloeosporioides was inoculated, germination could be observed in wounded hypocotyls 6 hours after inoculation. Better adhesion of conidia was evident at surface depressions of hypocotyls and germination started by emission of one or two terminal germ tubes. Round or sub-round shaped apressoria appeared 12 hours after inoculation. Up to 72 hours, acervula formation was not observed on inoculated tissues. RESUMOObjetivou-se estudar a dispersão espacial da mancha manteigosa em cafeeiro, por meio de arranjos espaciais e da análise da dinâmica e estruturas de focos da doença durante três anos consecutivos. Nesse período, não houve progresso da doença no campo. Sob plantas doentes, surgiam mudas com sintomas da doença, um indício da transmissibilidade via semente. Constatou-se um total de 10 focos, com média de 2,5 plantas/focos e com tendência desses focos (maior número de plantas sintomáticas) na direção das linhas de plantios, tendo disposição de forma elíptica. Observou-se número elevado de focos unitários, correspondendo a 52% das plantas doentes. Pelas análises de seqüências ordinárias runs e doublet verificou-se padrão espacial aleatório. Tal fato indica que a mancha manteigosa ocorre a partir de plantas isoladas (focos unitários) e que a principal via de transmissão é a semente (semente/planta/ semente). ABSTRACTThe aim of this study was to analyze the spatial dispersion pattern of the blister spot on coffee trees, through spatial arrangements and the analysis of the dynamics and foci structure of the disease during three consecutive years. No disease progress was observed in this area during the period of study. Seedlings showing disease symptoms were found under the canopy of diseased trees, strongly indicating that the disease may be transmitted through the seeds. Ten foci with 2.5 trees per focus were observed in the studied area. The foci were distributed following the plant rows with elliptical shape. Unitary foci predominated in the area in 52% of the diseased trees. Analyses of runs and doublets showed a random spatial pattern of the disease. These results evidenced that the blister spot occurs from isolated plants (unitary foci) and this characteristic may possibly be explained if the seeds are the main disease transmission agent (seed-plant-seed).
A Ferrugem Asiática (Phakopsora pachyrhizi H. Sydow & P. Sydow), relatada em diversas regiões do globo terrestre de climas tropicais e subtropicais, causa redução significativa na produtividade da soja [Glycine max (L.) Merr.]. Fatores bióticos como interação patógeno-hospedeiro e abióticos influenciam o progresso da doença. Objetivou-se neste trabalho estudar os efeitos da temperatura e de períodos de molhamento foliar no progresso da Ferrugem Asiática nas cultivares Conquista, Savana e Suprema. O experimento foi conduzido no Departamento de Fitopatologia da Universidade Federal de Lavras, em junho de 2004, em câmaras de crescimento vegetal nas temperaturas de 15, 20, 25 e 30 °C e períodos de molhamento foliar de 0, 6, 12, 18 e 24 horas. A inoculação foi realizada pulverizando-se as plantas com suspensão de 10(4) uredósporos de P. pachyrhizi.mL-1 de água. Dados da incidência e da severidade foram utilizados para avaliar o progresso da doença e integrados por meio da área abaixo da curva de progresso da incidência (AACPI) e da severidade (AACPS). Modelos de regressão não-linear foram ajustados para a AACPI e AACPS. Foi calculado o volume abaixo da superfície de resposta para incidência (VASRI) e severidade (VASRS) em relação à temperatura e molhamentos foliares com o objetivo de detectar diferenças entre cultivares. Molhamentos foliares acima de 15 horas e temperaturas próximas a 20 ºC, nas 3 cultivares avaliadas, determinaram maior intensidade da Ferrugem Asiática. Temperaturas próximas a 30 e 15 ºC ocasionaram menor intensidade da doença. Períodos de molhamento foliar abaixo de 6 horas reduziram a intensidade da doença. Todas as cultivares testadas foram suscetíveis à doença, entretanto, a cultivar Conquista apresentou maior VASRI e VASRS da Ferrugem Asiática comparada às cultivares Savana e Suprema, as quais não diferiram estatisticamente. Houve diferença entre as cultivares para AACPI em cada temperatura e molhamento foliar.
RESUMOEspécies de Colletotrichum estão presentes em todos os órgãos do cafeeiro. A importância desses fungos para a cultura do cafeeiro ainda é muito discutida, pois tratam-se de populações de espécies de Colletotrichum ocasionando diversos sintomas ou colonizando as plantas de Termos para indexação: Colletotrichum gloeosporioides, cafeeiros, colonização, frutos. ABSTRACTColletotrichum species are present in all organs of coffee trees and the importance of them is still controversial, since several populations can infect plant and cause disease or colonize plants in an endophytic form. This study was carried out with the objective to verify the presence of Colletotrichum spp. in different stages of development of coffee fruits and in the exocarp plus mesocarp, endocarp and endosperm of ripe fruits. In Fruits from the cultivars Catuaí Amarelo and Vermelho, Icatu, Topázio, Katipó, Rubi, Acaiá Cerrado and Mundo Novo came from the experimental farm the of Federal University of Lavras, Brazil. In the developing young fruits Colletotrichum spp. occurred at the average incidence of 86.6%. The highest incidence, 94.4% was observed in fruits from Topázio and Rubi cultivars, and the lowest incidences were 72.8% and 78.4%, in fruits from Icatu and Mundo Novo cultivars, respectively. Incidence of Colletotrichum spp. in the tissues of coffee berries varied according to the cultivar and the type of infected tissue. Based on exocarp plus mesocarp infection, the cultivars Topázio, Rubi, Acaiá Cerrado and Icatu were colonized with 84.72%, 79.16%, 77.77% and 48.6%, respectively. In relation to endocarp the maximum colonization was of 9.72% in cultivar Rubi and lower percentage of colonization was observed in Acaiá Cerrado with 1.39%. In relation to endosperm, the maximum colonization was observed in Rubi cultivar with 8.33% and lower percentage of colonization was observed in Mundo Novo and Katipó cultivars, both with 1.39%. Colletotrichum spp. was present in the seeds of all cultivars studied.
Honey from stingless bees (Melipona sp.) is a nutritious and medicinal product economically valued in the informal Brazilian market, driven by the growing demand for natural products; however, its physicochemical characteristics are still unknown. Thus, the aim of this study is to evaluate the physicochemical profile of the stingless bee honey produced in Cruzeiro do Sul, Acre. Honey samples were analyzed following the methodology recommended by Ministério da Agricultura, Pecuária e Abastecimento; the following parameters were established: moisture, total sugars, reducing sugars, apparent sucrose, ash, crude protein, diastase activity, Brix degrees, free acidity, lactonic acidity, total acidity, pH, hydroxymethylfurfural, electrical conductivity, color and Lugol, Lund and Fiehe reactions. Results show that these parameters are not suitable for all samples, being incompatible with established standards, indicating that the current legislation on Apis mellifera is not suitable for all characters analyzed, mainly moisture content, corroborating the need to standardize guidelines for honey from stingless bees. ResumenLa miel de abejas sin aguijón (Melipona sp.) es considerada un producto nutritivo, medicinal, económico y valorizado en el mercado informal del Brasil, impulsado por la creciente demanda de productos naturales. Sin embargo, las características fisicoquímicas del producto aún son poco conocidas. Por lo tanto, el objetivo de este estudio fue evaluar las características fisicoquímicas de la miel de abejas sin aguijón producidas en Cruzeiro do Sul, Acre, Brasil. Las mieles se analizaron de acuerdo a la metodología recomendada por el Ministério da Agricultura, Pecuária e Abastecimento, y los siguientes parámetros se establecieron: humedad, azúcares totales, azúcares reductores, sacarosa aparente, ceniza, proteína cruda, actividad de la diastasa, grados Brix, acidez libre, acidez lactónica, acidez total, pH, hidroximetilfurfural, conductividad eléctrica, color y reacciones de Lugol, Lund y Fiehe. Los resultados de este trabajo demuestran que estos parámetros no son adecuados para todas las muestras, siendo incompatibles con las normas establecidas, indicando que la legislación vigente para Apis mellifera no es adecuada para todas las variables analizadas, principalmente el contenido de humedad, lo que corrobora la necesidad de una estandarización propia para la miel de abejas sin aguijón. RESUMOObjetivou-se, neste trabalho, verificar o efeito da temperatura e do período de molhamento foliar no progresso da severidade do oídio Microsphaera diffusa Cke. & Pk., nas cultivares de soja Conquista e Suprema [Glycine max (L.) Merr.]. As plantas foram inoculadas em casa-de-vegetação por agitação de plantas doentes sobre plantas sadias no estádio vegetativo V 3 . Em seguida foram colocadas, ao acaso, em câmaras de crescimento vegetal, nas temperaturas de 15, 20, 25 e 30°C e períodos de molhamento foliar de 0, 6, 12, 18 e 24 h. Dados da severidade foram utilizados para avaliar o progresso da doença e integrados ao longo do tempo por meio da área abaixo da curva de progresso da severidade (AACPS). Modelos de regressão não-linear foram ajustados para AACPS. O progresso da intensidade do oídio (AACPS) nas cultivares Conquista e Suprema foi favorecido por temperaturas próximas a 23ºC e 24ºC, respectivamente. Períodos de molhamento foliar de 8h proporcionaram o progresso máximo da severidade da doença na cultivar Conquista sob temperatura de 23ºC. ABSTRACTIn this study the effects of temperature and leaf wetness period in the severity progress of Powdery Mildew Cke. & Pk. in soybean cultivars Conquista and Suprema [Glycine max (L.) Merr.] were evaluated. Plants at the V3 stage were inoculated in greenhouse. Subsequently, they were conditioned into growth chambers at temperatures of 15, 20, 25 and 30°C and leaf wetness periods of 0, 6, 12, 18 and 24 hours. Severity data were integrated in time by the disease progress curve for severity (AUDPCS). Nonlinear regression models were adjusted for the disease intensity. Temperatures near 23ºC and 24ºC favored the powdery mildew intensity progress (AUDPCS) in Conquista and Suprema cultivars, respectively. Leaf wetness period of 8 h allowed the maximum progress of the disease in Conquista at temperatures of 23ºC. Temperatures near 30ºC and 15ºC reduced powdery mildew intensity.
RESUMOCom o objetivo de avaliar a eficiência de alguns fungicidas sobre Colletotrichum gloeosporioides, agente etiológico da mancha manteigosa do cafeeiro (Coffea arabica L.), testes in vitro foram conduzidos no Laboratório de Diagnose e Controle/UFLA. Utilizouse o método de incorporação de fungicidas ao meio de cultura MEA 2% para a avaliação da inibição do crescimento micelial e em lâmina escavada contendo água com fungicida para a germinação de conídios. Os fungicidas, tetraconazol, triadimenol, chlorotalonil e mancozeb foram testados quanto à inibição do crescimento do micelial (nas concentrações de 1, 5, 10, 25, 50, 100, 500 e 1.000 mg L -1 ) e quanto à inibição da germinação de conídios (nas concentrações de 1, 5, 10, 25, 50 e 100 mg L -1 ). Os fungicidas tetraconazol e triadimenol apresentaram alta eficiência na inibição do crescimento micelial. Os fungicidas chlorotalonil e mancozeb mostraram baixa eficiência e ineficiência, respectivamente. Quanto à germinação dos conídios, os fungicidas que demonstraram maior eficiência em baixas concentrações foram o chlorotalonil e o tetraconazol. ABSTRACTWith the aim of assessing the effect of selected fungicides on Colletotrichum gloeosporioides, the cause of coffee blister spot, in vitro tests were carried out in the Laboratory of Diagnosis and Control/UFLA, Federal University of Lavras, Brazil. In the in vitro experiments the fungicides were incorporated into malt extract medium (MEA 2%) to evaluate the effect on the fungus growth rate, and concavity slides containing water plus fungicide to assess the conidia germination. The fungicides tetraconazol, triadimenol, chlorothalonil and mancozeb were tested on the mycelial growth inhibition (in the concentrations of 1, 5, 10, 25, 50, 100, 500 and 1.000 mg L -1 ) and on the inhibition conidia germination (in the concentrations of 1, 5, 10, 25, 50 e 100 mg L -1 ). The fungicides tetraconazol and triadimenol showed high efficiency on the mycelial growth inhibition. Chlorotalonil and mancozeb showed low efficiency and inefficiency, respectively. As to the germination of conidia, chlorothalonil and tetraconazol demonstrated to be more efficient in low concentrations.
