RESUMONos estudos envolvendo a interação Colletotrichum gloeosporioides-cafeeiro (Coffea arabica L.), poucas são as informações a respeito do modo de penetração e colonização deste patógeno. Estudou-se por meio da microscopia eletrônica de varredura (MEV), os eventos de pré-penetração e penetração de C. gloeosporioides em hipocótilos de cafeeiros e a colonização natural de ramos e frutos provenientes de plantas com mancha manteigosa. Realizaram-se três ensaios. No primeiro, fez-se a observação dos eventos de pré-penetração de C. gloeosporioides inoculados em hipocótilos de plântulas da cv. Acaiá Cerrado com e sem ferimentos; no segundo, observou-se a colonização de ramos enfermos em que havia murcha drástica e necrose local e, no terceiro, a colonização de frutos enfermos. A germinação dos conídios nos hipocótilos feridos ocorreu 6 horas após a inoculação, com a formação de um ou dois tubos germinativos terminais e a adesão dos conídios nas depressões dos hipocótilos. Apressórios globosos a subglobosos e de contorno regular surgiram 12 horas após a inoculação. Até 72 horas, não foi possível observar a formação de acérvulos sobre os tecidos submetidos à inoculação. Nos ramos observavase colonização nos vasos floema e parênquima cortical. Nos frutos em diferentes estádios de maturação foi observada a colonização do tecido próximo à epiderme e colapso dos estômatos na área lesionada.Palavras-chave: Glomerella cingulata, Coffea arabica, mancha manteigosa, microscopia eletrônica de varredura. ABSTRACT HISTOPATHOLOGICAL STUDIES OF THE INTERACTION COLLETOTRICHUM GLOEOSPORIOIDES -COFFEE TREESIn studies involving the interaction Colletotrichum gloeosporioides -coffee trees (Coffea arabica L.) there is little information regarding the pre-penetration, penetration and colonization pathways of this pathogen. The objectives of this work were: 1. to study through scanning electron microscopy (SEM) the pre-penetration and penetration events of C. gloeosporioides in hypocotyls of coffee plantlets; 2. to observe the colonization of Colletotrichum spp. in branches and fruits of coffee trees with blister spot symptoms. The study took place in three trials. In the first trial, it was observed and reported the pre-penetration events of C. gloeosporioides inoculated on hypocotyls of coffee cv. Acaiá Cerrado with and without wounds. The second trial was performed to investigate the colonization of diseased coffee branches that presented drastic wilt and local necrosis was observed. Finally the third trial investigated the colonization of this pathogen in diseased coffee fruits. When C. gloeosporioides was inoculated, germination could be observed in wounded hypocotyls 6 hours after inoculation. Better adhesion of conidia was evident at surface depressions of hypocotyls and germination started by emission of one or two terminal germ tubes. RESUMOObjetivou-se estudar a dispersão espacial da mancha manteigosa em cafeeiro, por meio de arranjos espaciais e da análise da dinâmica e estruturas de focos da doença durante três anos consecutivos. Nesse período, não houve progresso da doença no campo. Sob plantas doentes, surgiam mudas com sintomas da doença, um indício da transmissibilidade via semente. Constatou-se um total de 10 focos, com média de 2,5 plantas/focos e com tendência desses focos (maior número de plantas sintomáticas) na direção das linhas de plantios, tendo disposição de forma elíptica. Observou-se número elevado de focos unitários, correspondendo a 52% das plantas doentes. Pelas análises de seqüências ordinárias runs e doublet verificou-se padrão espacial aleatório. Tal fato indica que a mancha manteigosa ocorre a partir de plantas isoladas (focos unitários) e que a principal via de transmissão é a semente (semente/planta/ semente). ABSTRACTThe aim of this study was to analyze the spatial dispersion pattern of the blister spot on coffee trees, through spatial arrangements and the analysis of the dynamics and foci structure of the disease during three consecutive years. No disease progress was observed in this area during the period of study. Seedlings showing disease symptoms were found under the canopy of diseased trees, strongly indicating that the disease may be transmitted through the seeds. Ten foci with 2.5 trees per focus were observed in the studied area. The foci were distributed following the plant rows with elliptical shape. Unitary foci predominated in the area in 52% of the diseased trees. Analyses of runs and doublets showed a random spatial pattern of the disease. These results evidenced that the blister spot occurs from isolated plants (unitary foci) and this characteristic may possibly be explained if the seeds are the main disease transmission agent (seed-plant-seed).
RESUMOEspécies de Colletotrichum estão presentes em todos os órgãos do cafeeiro. A importância desses fungos para a cultura do cafeeiro ainda é muito discutida, pois tratam-se de populações de espécies de Colletotrichum ocasionando diversos sintomas ou colonizando as plantas de Termos para indexação: Colletotrichum gloeosporioides, cafeeiros, colonização, frutos. ABSTRACTColletotrichum species are present in all organs of coffee trees and the importance of them is still controversial, since several populations can infect plant and cause disease or colonize plants in an endophytic form. This study was carried out with the objective to verify the presence of Colletotrichum spp. in different stages of development of coffee fruits and in the exocarp plus mesocarp, endocarp and endosperm of ripe fruits. In Fruits from the cultivars Catuaí Amarelo and Vermelho, Icatu, Topázio, Katipó, Rubi, Acaiá Cerrado and Mundo Novo came from the experimental farm the of Federal University of Lavras, Brazil. In the developing young fruits Colletotrichum spp. occurred at the average incidence of 86.6%. The highest incidence, 94.4% was observed in fruits from Topázio and Rubi cultivars, and the lowest incidences were 72.8% and 78.4%, in fruits from Icatu and Mundo Novo cultivars, respectively. Incidence of Colletotrichum spp. in the tissues of coffee berries varied according to the cultivar and the type of infected tissue. Based on exocarp plus mesocarp infection, the cultivars Topázio, Rubi, Acaiá Cerrado and Icatu were colonized with 84.72%, 79.16%, 77.77% and 48.6%, respectively. In relation to endocarp the maximum colonization was of 9.72% in cultivar Rubi and lower percentage of colonization was observed in Acaiá Cerrado with 1.39%. In relation to endosperm, the maximum colonization was observed in Rubi cultivar with 8.33% and lower percentage of colonization was observed in Mundo Novo and Katipó cultivars, both with 1.39%. Colletotrichum spp. was present in the seeds of all cultivars studied.
Considerando-se a importância das mudas na formação de lavoura cafeeira (Coffea arabica L.), objetivou-se avaliar características agronômicas e índices de qualidade de mudas do cafeeiro cv. Topázio MG 1190, produzidos em tubetes com a utilização de diferentes proporções de materiais orgânicos no substrato comercial. O experimento foi em delineamento experimental em blocos casualizados, dispostos em esquema fatorial 4 x 4, sendo quatro fontes de esterco: bovino, cama de frango, coelho e suíno em quatro proporções (0, 20, 40 e 60%) no substrato comercial Bioplant® com quatro repetições. As parcelas foram formadas por sete plantas dispostas em tubetes com volume de 180 mL. Foram avaliadas as características agronômicas: altura da planta, número de par de folhas, área foliar, diâmetro de caule, massa seca da parte aérea, massa seca da raiz, relação altura da planta e diâmetro de caule, relação massa seca de parte aérea e massa seca da raiz e índice de qualidade de Dickson (IQD). Os resultados obtidos permitem concluir que o esterco de coelho até a dose de 32,7% no substrato proporcionou maior desenvolvimento das mudas de café representado pelo IQD calculado em 1,31. A adição de cama de frango, esterco suíno e esterco bovino podem ser utilizados até as doses máximas de 25%, 5,2% e 17,4% respectivamente. A partir dessas doses pode ocorrer uma rápida diminuição no IQD. Na dose de 60% de esterco suíno e esterco bovino foi observado morte de mudas.
RESUMOCom o objetivo de avaliar a eficiência de alguns fungicidas sobre Colletotrichum gloeosporioides, agente etiológico da mancha manteigosa do cafeeiro (Coffea arabica L.), testes in vitro foram conduzidos no Laboratório de Diagnose e Controle/UFLA. Utilizouse o método de incorporação de fungicidas ao meio de cultura MEA 2% para a avaliação da inibição do crescimento micelial e em lâmina escavada contendo água com fungicida para a germinação de conídios. Os fungicidas, tetraconazol, triadimenol, chlorotalonil e mancozeb foram testados quanto à inibição do crescimento do micelial (nas concentrações de 1, 5, 10, 25, 50, 100, 500 e 1.000 mg L -1 ) e quanto à inibição da germinação de conídios (nas concentrações de 1, 5, 10, 25, 50 e 100 mg L -1 ). Os fungicidas tetraconazol e triadimenol apresentaram alta eficiência na inibição do crescimento micelial. Os fungicidas chlorotalonil e mancozeb mostraram baixa eficiência e ineficiência, respectivamente. Quanto à germinação dos conídios, os fungicidas que demonstraram maior eficiência em baixas concentrações foram o chlorotalonil e o tetraconazol. ABSTRACTWith the aim of assessing the effect of selected fungicides on Colletotrichum gloeosporioides, the cause of coffee blister spot, in vitro tests were carried out in the Laboratory of Diagnosis and Control/UFLA, Federal University of Lavras, Brazil. In the in vitro experiments the fungicides were incorporated into malt extract medium (MEA 2%) to evaluate the effect on the fungus growth rate, and concavity slides containing water plus fungicide to assess the conidia germination. The fungicides tetraconazol, triadimenol, chlorothalonil and mancozeb were tested on the mycelial growth inhibition (in the concentrations of 1, 5, 10, 25, 50, 100, 500 and 1.000 mg L -1 ) and on the inhibition conidia germination (in the concentrations of 1, 5, 10, 25, 50 e 100 mg L -1 ). The fungicides tetraconazol and triadimenol showed high efficiency on the mycelial growth inhibition. Chlorotalonil and mancozeb showed low efficiency and inefficiency, respectively. As to the germination of conidia, chlorothalonil and tetraconazol demonstrated to be more efficient in low concentrations.
The present study was carried out with the objective to provide information on the pre-penetration, penetration and colonization events of isolates of Colletotrichum gloeosporioides from mango and coffee, when inoculated in coffee leaves. The cultivar Catucaí Vermelho was used in all experiments. Coffee leaves were selected, standardized and washed in sterile water. After that, circular areas of 0.5 cm in diameter were marked on the undersurface of the leaves. In the center of each circular area, 20 µL of spore suspension was deposited. RESUMONeste trabalho, os objetivos foram avaliar a transmissibilidade de C. gloeosporioides mancha manteigosa (MM) em sementes e estudar o efeito do tratamento por fungicidas no controle do referido patógeno. Colheram-se frutos cerejas em plantas doentes e sadias, procedeu-se o despolpamento manual secando as sementes à sombra, retirou-se o endocarpo e, em seguida, realizaram-se os devidos tratamentos (fungicida) semeando-as em areia esterilizada. Avaliaram-se os sintomas de necrose e morte das plântulas e a incidência de C. gloeosporioides, por meio do teste de incubação em substrato de papel (blotter test), em sementes de cafeeiro tratadas e não tratadas com mistura comercial de benzimidazol+dimetilditiocarbamato (dose i.a. de 45 g+105 g/100 kg sementes). Com o desenvolvimento das plântulas, observaram-se em sementes de plantas com MM , altos índices de morte de plântulas, tendo apenas 5,2% de sobrevivência, porém, com a abertura dos primeiros folíolos observaram-se, nestas plântulas, sintomas típicos da doença, com posterior morte das mesmas, enquanto que, a partir de sementes sadias, as plântulas apresentam-se vigorosas e em pleno crescimento. Já, com o uso do tratamento fungicida, observou-se efeito significativo, principalmente em sementes de plantas doentes (MM), nas quais verificou-se uma menor área abaixo da curva de progresso da doença e, consequentemente, menor incidência e severidade da doença. Pelo bloter test, em sementes tratadas, não houve crescimento de nenhum agente fúngico tanto para sementes de plantas com MM como para sementes de plantas sadias. Em sementes não tratadas, observou-se incidência de C. gloeosporioides em 29,8% nas sementes de plantas com MM e 1,2% de incidência em sementes de plantas sadias. Com base nestes resultados, fica evidenciado que neste patossistema, mancha manteigosa -cafeeiro, a principal via de transmissão de C. gloeosporioides é a semente.Termos para indexação: Sementes, fungicidas, Colletotrichum gloeosporioides e Coffea arabica. RESUMO Neste trabalho, objetivou-se avaliar a eficiência de alguns fungicidas no controle da mancha manteigosa (Colletotrichum gloeosporioides) do cafeeiro (Coffea arabica L.) em campo, e verificar os efeitos climáticos no progresso da doença. Foram conduzidos ensaios sobre cafeeiros da cultivar Catucaí Vermelho, com aproximadamente 8 anos de idade. Os fungicidas testados foram: tetraconazol, triadimenol, chlorotalonil e mancozeb, nas dosagens de 1,0L/ha, 12Kg/ha, 3Kg/ha e 2Kg/ha, respectivamente. Coletaram-se dados climáticos diariamente, no período de outubro de 2004 a maio de 2006. Os fungicidas chlorotalonil e triadimenol foram os que proporcionaram os melhores resultados de eficiência de controle do progresso de morte de ramos e frutos, destacando a produção de frutos cereja. Entre os meses de outubro a janeiro, verificou-se elevado progresso de mortes de ramos influenciado pelas altas precipitações pluviais. Os fungicidas tetraconazol e o mancozeb mostraram-se ineficientes. Plantas com mancha manteigosa, mesmo quando pulverizadas com fungicidas, tiveram significativa queda de frutos, enquanto plantas sadias apresentaram produções 95% superiores àquelas. ABSTRACT The aim of this work was to assess the effect of selected fungicides on the control of coffee blister spot in the field and to verify the climatic effects on the progress of the disease. Trials were carried out with coffee plants of cultivar Catucaí Vermelho, approximately 8 years old. The fungicides tested were tetraconazol (1L/ha), triadimenol (12Kg/ha), chlorothalonil (3Kg/ha), and mancozeb (2Kg/ha). Climatic data were collected daily from October 2004 to May 2006. The fungicides chlorotalonil and triadimenol provided the best efficiency results on the control of the progress of die-back and the number of fruits, with an increase in the cherry production. The increase of die-back during the period from October to January was possibly induced by the rainfall. Tetraconazol and mancozeb were not efficient. Coffee trees affected by blister spot, even if treated with fungicides, had significant decrease in the fruit production and the untreated healthy trees had their yield 95% higher than of those treated.
