Multi-platform molecular profiling identifies subgroups of TNBC with different biomarker profiles, suggesting numerous potentially targetable alterations in TNBC. TNBC is further characterized by different gene mutations and proliferative activity relative to AR expression, highlighting a need for comprehensive pathologic examination with potential to develop different, individualized treatment options.
Ein Biomarker (biologischer Marker) ist definiert als ein charakteristisches Merkmal, das nach der Validierung, der objektiven Messung normaler biologischer und pathogenetischer Prozesse oder der pharmakologischen Ansprechbarkeit auf eine therapeutische Intervention dient. Man setzt große Hoffnung in die Entwicklung von Bio− markern, da man mit deren Hilfe eine wesentli− che Verbesserung der Diagnose von Krankheiten erreichen könnte. Bei der Parkinsonkrankheit er− hofft man vor allem eine sichere Differenzialdi− agnose und prämotorische Diagnose zu ermögli− chen. Weiterhin könnte ein Biomarker als Surro− gatmarker für klinische Studien zum Nachweis der Wirksamkeit von neuartigen therapeuti− schen Interventionen, wie z. B. neuroprotektiven Strategien, und zur Beobachtung der Krankheits− progression Verwendung finden. Schließlich er− wartet man, dass die Anwendung von Biomar− kern zu einer besseren Klassifizierung der Er− krankung führt; nach dem gegenwärtigen Kennt− nisstand wird unter dem Begriff Parkinsonkrank− heit eine Gruppe von neurodegenerativen Er− krankungen subsumiert, die eine unterschiedli− che Pathogenese und Symptomatik aufweisen. In diesem Beitrag werden wir zunächst kurz die Eigenschaften nennen, die ein Biomarker der Par− kinsonkrankheit haben soll. Dann werden wir die Kriterien besprechen, die zur diagnostischen Eva− luierung von Labortests der Parkinsonkrankheit verwendet werden. Schließlich werden wir die unserer Meinung nach wichtigsten Biomarker− Kandidaten der Parkinsonkrankheit vorstellen und deren Eignung als Biomarker anhand der Kri− terien zur diagnostischen Evaluierung bewerten.
