The avermectins, milbemycins and spinosyns are collectively referred to as macrocyclic lactones (MLs) which comprise several classes of chemicals derived from cultures of soil micro-organisms. These compounds are extensively and increasingly used in veterinary medicine and agriculture. Due to their potential effects on non-target organisms, large amounts of information on their impact in the environment has been compiled in recent years, mainly caused by legal requirements related to their marketing authorization or registration. The main objective of this paper is to critically review the present knowledge about the acute and chronic ecotoxicological effects of MLs on organisms, mainly invertebrates, in the terrestrial and aquatic environment. Detailed information is presented on the mode-of-action as well as the ecotoxicity of the most important compounds representing the three groups of MLs. This information, based on more than 360 references, is mainly provided in nine tables, presenting the effects of abamectin, ivermectin, eprinomectin, doramectin, emamectin, moxidectin, and spinosad on individual species of terrestrial and aquatic invertebrates as well as plants and algae. Since dung dwelling organisms are particularly important non-targets, as they are exposed via dung from treated animals over their whole life-cycle, the information on the effects of MLs on dung communities is compiled in an additional table. The results of this review clearly demonstrate that regarding environmental impacts many macrocyclic lactones are substances of high concern particularly with larval instars of invertebrates. Recent studies have also shown that susceptibility varies with life cycle stage and impacts can be mitigated by using MLs when these stages are not present. However information on the environmental impact of the MLs is scattered across a wide range of specialised scientific journals with research focusing mainly on ivermectin and to a lesser extent on abamectin doramectin and moxidectin. By comparison, information on compounds such as eprinomectin, emamectin and selamectin is still relatively scarce.

β-Adrenergic receptor blockers (β-blockers) are applied to treat high blood pressure, ischemic heart disease, and heart rhythm disturbances. Due to their widespread use and limited human metabolism, β-blockers are widely detected in sewage effluents and surface waters. β-Adrenergic receptors have been characterized in fish and other aquatic animals, so it can be expected that physiological processes regulated by these receptors in wild animals may be affected by the presence of β-blockers. Because ecotoxicological data on β-blockers are scarce, it was decided to choose the β-blocker atenolol as a case study pharmaceutical within the project ERAPharm. A starting point for the assessment of potential environmental risks was the European guideline on the environmental risk assessment of medicinal products for human use. In Phase I of the risk assessment, the initial predicted environmental concentration (PEC) of atenolol in surface water (500 ng L−1) exceeded the action limit of 10 ng L−1. Thus, a Phase II risk assessment was conducted showing acceptable risks for surface water, for groundwater, and for aquatic microorganisms. Furthermore, atenolol showed a low potential for bioaccumulation as indicated by its low lipophilicity (log KOW = 0.16), a low potential for exposure of the terrestrial compartment via sludge (log KOC = 2.17), and a low affinity for sorption to the sediment. Thus, the risk assessment according to Phase II-Tier A did not reveal any unacceptable risk for atenolol. Beyond the requirements of the guideline, additional data on effects and fate were generated within ERAPharm. A 2-generation reproduction test with the waterflea Daphnia magna resulted in the most sensitive no-observed-effect concentration (NOEC) of 1.8 mg L−1. However, even with this NOEC, a risk quotient of 0.003 was calculated, which is still well below the risk threshold limit of 1. Additional studies confirm the outcome of the environmental risk assessment according to EMEA/CHMP (2006). However, atenolol should not be considered as representative for other β-blockers, such as metoprolol, oxprenolol, and propranolol, some of which show significantly different physicochemical characteristics and varying toxicological profiles in mammalian studies.

Cattle treated with the veterinary parasiticide ivermectin fecally excrete residues. The authors report the exposition and dissipation characteristics of these residues in dung of ivermectin-treated cattle and in soil beneath this dung on pastures in Canada, France, Switzerland, and The Netherlands. Residues were quantified for dung collected from cattle after 3 d, 7 d, 14 d, and 28 d posttreatment and subsequently exposed in the field for up to 13 mo. The authors optimized a high-performance liquid chromatography-fluorescence detection method to detect ivermectin residues in dung and soil matrices. They showed that a solid phase extraction and purification step generally can be eliminated to reduce the time and cost of these analyses. They also found that the addition of water to relatively dry samples improves the extraction efficiency of residues. They then analyzed the field samples to document differences in ivermectin dissipation in cattle dung among sites, with 50% dissipation times of up to 32 d and 90% dissipation times >396 d. They further showed that the dissipation characteristics of residues are comparable between dung of ivermectin-treated cattle and dung to which ivermectin has been added directly. Lastly, they report the first use of a desorption electrospray ionization-high-resolution-mass spectrometric method to detect residues of metabolites in a dung matrix. Possible mechanisms to explain endocrine effects on reproduction and sex differentiation are presented for selected pharmaceuticals, agrochemicals, industrial chemicals and plant sterols which are known to be present in the aquatic environment. Disruptions of the hormonal coordination can be induced by xenobiotics on various levels of the hierachically organised endocrine system of vertebrates. Phthalate plasticisers, for example, may disrupt the pituitary control of gonadal functions; prenatal/larval exposure to synthetic estrogen impairs sex differentiation and neuroendocrine sexual determination of the central nervous system; phenylurea herbicides block the androgen receptor; the biotransformation of weakly estrogenic plant sterol components of paper mill wastewater (e.g. beta-sitosterol) may lead to androgenic compounds. The effect of hypolipidemic drugs on lipid homeostasis (peroxysom proliferation) is transmitted via a receptor protein that seems to be closely related to the endocrine system both functionally as well as phylogenetically; possible interferences with the neuroendocrine control of sex differentiation are also discussed. In invertebrates, tributyltin is known to effect the biosynthesis of steroidal sexual hormones. PCBs are suspected to be competitive inhibitors of the steroid catabolism. In order to identify potential risks caused by chemicals to the reproductive capacities of aquatic animals and to the quality of drinking water, methods should be established to detect endocrine disrupters at the various levels of the endocrine system.

In Europe, the decision whether waste is hazardous or not is based on 15 properties, among them the HP 14 property ("ecotoxic": waste that presents or may present immediate or delayed risks for 1 or more sectors of the environment). This document describes a strategy for assessing the HP 14 property, based on a combination of 2 approaches: the summation of classified compounds in the waste carried out according to the regulation on Classification, Labeling and Packaging (CLP) of substances and mixtures and the usage of the results of biotests performed on waste eluates and solid wastes. The proposal is based mainly on recommendations of a European ring test carried out in 2007, the work performed in the CEN/TC 292/WG 7 standardization working group, and the results of various research projects regarding the ecotoxicological characterization of waste carried out mainly in France and Germany. Examples are provided showing that, using this approach, a distinction between hazardous and nonhazardous wastes is possible, independent of which type of threshold values is used (currently, both effect concentrations [EC] or lowest ineffective dilutions [LID] values have been used successfully). Furthermore, a battery of tests (3 using waste eluates and 3 using solid waste samples, plus, under certain conditions, a genotoxicity test) is recommended for the ecotoxicological testing of wastes. We propose to consider this combined approach when defining the legal requirements for the ecotoxicological classification of wastes.

By degrading the dung of livestock that graze on pastures, coprophilous arthropods accelerate the cycling of nutrients to maintain pasture quality. Many veterinary medicinal products, such as ivermectin, are excreted unchanged in the dung of treated livestock. These residues can be insecticidal and may reduce the function (i.e., dung-degradation) of the coprophilous community. In the present study, we used a standard method to monitor the degradation of dung from cattle treated with ivermectin. The present study was performed during a 1-yr period on pastures in Canada, France, The Netherlands, and Switzerland. Large effects of residue were detected on the coprophilous community, but degradation of dung was not significantly hampered. The results emphasize that failure to detect an effect of veterinary medicinal product residues on dung-degradation does not mean that the residues do not affect the coprophilous community. Rather, insect activity is only one of many factors that affect degradation, and these other factors may mask the nontarget effect of residues. Environ Toxicol Chem 2016;35:1953-1958. © 2015 SETAC.
