Subacute effects of pulsed copper, zinc, or ammonia exposures were examined, including a range of pulse concentrations, durations, frequencies, and recovery times between pulses, using short-term chronic Pimephales promelas and 21-d Daphnia magna tests. Sublethal effects were rarely observed independent of mortality. Effects were observed only at concentrations near the species continuous exposure 48 h LC50 for each chemical. Daphnia often rebounded from temporary reproduction effects, meeting or exceeding control responses by the end of the test. Effects of 24 h ammonia or copper pulses were diminished soon after the pulse was removed, while 24 h zinc pulses caused continued effects for several days following removal of the pulse, indicating a slower uptake and/or depuration rate for zinc. D. magna exhibited less mortality as copper pulses were spaced further apart, while fish were equally or more affected with longer recovery times between copper pulses, indicative of different adaptation mechanisms between the two species. Responses were not predictable based on either average concentration or a combination of duration and concentration. Chronic water quality criteria and effluent permit limits, expressed as a 4- or 30-d average concentration, respectively, may not be appropriate for protecting against effects of pulsed exposures, depending on the frequency, magnitude, and duration of pulses, as well as the recovery period between events.

The U.S. Environmental Protection Agency (U.S. EPA) and state agencies implement the Clean Water Act, in part, by evaluating the toxicity of effluent and surface water samples. A common goal for both regulatory authorities and permittees is confidence in an individual test result (e.g., no-observed-effect concentration [NOEC], pass/fail, 25% effective concentration [EC25]), which is used to make regulatory decisions, such as reasonable potential determinations, permit compliance, and watershed assessments. This paper discusses an additional statistical approach (test of significant toxicity [TST]), based on bioequivalence hypothesis testing, or, more appropriately, test of noninferiority, which examines whether there is a nontoxic effect at a single concentration of concern compared with a control. Unlike the traditional hypothesis testing approach in whole effluent toxicity (WET) testing, TST is designed to incorporate explicitly both α and β error rates at levels of toxicity that are unacceptable and acceptable, given routine laboratory test performance for a given test method. Regulatory management decisions are used to identify unacceptable toxicity levels for acute and chronic tests, and the null hypothesis is constructed such that test power is associated with the ability to declare correctly a truly nontoxic sample as acceptable. This approach provides a positive incentive to generate high-quality WET data to make informed decisions regarding regulatory decisions. This paper illustrates how α and β error rates were established for specific test method designs and tests the TST approach using both simulation analyses and actual WET data. In general, those WET test endpoints having higher routine (e.g., 50th percentile) within-test control variation, on average, have higher method-specific α values (type I error rate), to maintain a desired type II error rate. This paper delineates the technical underpinnings of this approach and demonstrates the benefits to both regulatory authorities and permitted entities.

Semiempirical models are useful for interpreting the response of aquatic organisms to toxicants as a function of exposure concentration and duration. Most applications predict cumulative mortality at the end of the test for constant exposure concentrations. Summary measures, such as the median lethal concentration, are then estimated as a function of concentration. Real-world exposures are not constant. Effects may depend on pulse timing, and cumulative analysis based only on integrated exposure concentration is not sufficient to interpret results. We undertook a series of pulsed-exposure experiments using standard toxicological protocols and interpreted the results (mortality, biomass, and reproduction) using a dynamic generalization of a Mancini/Breck--type model that includes two compartments, one for internal concentration as a function of exposure and one for site-of-action concentration or accumulated damage as a function of the internal dose. At exposure concentrations near the effects level, the model explained approximately 50% of the variability in the observed time history of survival, 43% of the change in biomass, and 83% of the variability in net reproduction. Unexplained variability may result from differences in organism susceptibility, amplified by the effects of small sample sizes in standard tests. The results suggest that response is sensitive to prior conditions and that constant-exposure experiments can underestimate the risk from intermittent exposures to the same concentration. For pulsed exposures, neither the average nor the maximum concentration alone is an adequate index of risk, which depends on both the magnitude, duration, and timing of exposure pulses. Better understanding about the impacts of pulsed exposures will require use of experimental protocols with significantly greater numbers of replicates.

A framework is presented that is intended to facilitate the evaluation of potential aquatic ecological risks resulting from discharges of down-the-drain chemicals. A scenario is presented using representatives of many of the types of chemicals that are treated domestically. Predicted environmental chemical concentrations are based on reported loading rates and routine removal rates for 3 types of treatment: trickling filter, activated sludge secondary treatment, and activated sludge plus advanced oxidation process as well as instream effluent dilution. In tier I, predicted effluent concentrations were compared with the lowest predicted-no-effect concentration (PNEC) obtained from the literature using safety factors as needed. A cumulative risk characterization ratio (cumRCR) < 1.0 indicates that risk is unlikely and no further action is needed. Otherwise, a tier 2 assessment is used, in which PNECs are based on trophic level. If tier 2 indicates a possible risk, then a retrospective assessment is recommended. In tier 1, the cumRCR was > 1.0 for all 3 treatment types in our scenario, even though no chemical exceeded a hazard quotient of 1.0 in activated sludge or advanced oxidation process. In tier 2, activated sludge yielded a lower cumRCR than trickling filter because of higher removal rates, and the cumRCR in the advanced oxidation process was << 1.0. Based on the maximum cumulative risk ratio (MCR), more than one-third of the predicted risk was accounted for by one chemical, and at least 90% was accounted for by 3 chemicals, indicating that few chemicals influenced the mixture risk in our scenario. We show how a retrospective assessment can test whether certain chemicals hypothesized as potential drivers in the prospective assessment could have, or are having, deleterious effects on aquatic life. The free-flowing Clinch and Powell watershed in Virginia, USA, harbors a high number of endemic mussel and fish species but they are declining or going extinct at an alarming rate. To prioritize resource management strategies with respect to these fauna, a geographical information system was developed and various statistical approaches were used to relate human land uses with available fish, macroinvertebrate, and native mussel assemblage data. Both the Ephemeroptera, Plecoptera, Trichoptera (EPT) family-level index, and the fish index of biotic integrity (IBI) were lowest in a subwatershed with the greatest coal mining activity (analysis of variance [ANOVA], p < 0.05). Limited analyses in two other subwatersheds suggested that urban and agricultural land uses within a specified riparian corridor were more related to mussel species richness and fish IBI than land uses in entire catchments. Based on land uses within a riparian corridor of 200 m x 2 km for each biological site in the watershed, fish IBI was inversely related to percent cropland and urban area and positively related to pasture area (stepwise multiple regression, R2 = 0.55, p < 0.05). Sites less than 2 km downstream of urban areas, major highways, or coal mine activities had a significantly lower mean IBI value than those more than 2 km away (ANOVA, p < .05). Land use effects included poorer instream cover and higher substrate embeddedness (t test, p < 0.05). Weaker land use relationships were observed for EPT and mussel species richness. Episodic spills of toxic materials, originating from transportation corridors, mines, and industrial facilities, also have resulted in local extirpations of native species. particularly mussels. The number of co-occurring human activities was directly related to stream elevation in the Clinch River, with more human land uses in headwater areas. Approximately 60% of known U.S. Fish and Wildlife mussel concentration sites in the watershed are located within 2 km of at least two land use sources identified as potentially stressful in our analyses. Our results indicate that a number of land uses and stressors are probably responsible for the decline in native species. However, protection of naturally vegetated riparian corridors may help mitigate some of these effects.
