ResumenObjetivo. comparar el comportamiento de tres genes diana 16S ADNr, polA, y TpN47, para la detección de T. pallidum subsp. Pallidum. Métodos. Se usaron técnicas moleculares como la reacción en cadena de la polimerasa en muestras de cordón umbilical . Mediante PCR convencional, PCR anidada y PCR en tiempo real se amplificaron blancos moleculares del microorganismo. Resultados. Se evidenció que con los tres genes por PCR convencional se obtienen similares resultados, pero por con PCR anidada y PCR en Tiempo Real, el gen TpN47 tiene mayor sensibilidad en comparación con los genes polA y 16S ADNr. Se concluye que el gen TpN47 se puede usar como blanco molecular para el diagnóstico oportuno de sífilis congénita por medio de PCR anidada y en tiempo real, ya que alcanzó la máxima sensibilidad y especificidad en este estudio.Palabras clave: Sífilis congénita, Treponema pallidum subsp. pallidum, PCR anidada, PCR en Tiempo Real. AbstractObjective. To compare the behavior of the three target genes (16S rDNA, polA, and TpN47) for the detection of T. pallidum subsp. Pallidum. Methods. Molecular techniques such as polymerase chain reaction were used on samples of an umbilical cord. Molecular targets of the microorganism were amplified by means of conventional PCR, nested PCR and real-time PCR. Results. Similar results for the three genes were obtained by conventional PCR; but in the case of nested PCR and real-time PCR, the gen TpN47 exhibited higher sensitivity in comparison to the genes polA and 16S rDNA. In conclusion, the gen TpN47 can be used as a molecular target for the prompt diagnosis of congenital syphilis through nested PCR and real-time PCR due to its high sensitivity and specificity shown in this study.
Introducción. La sífilis es una enfermedad producida por Treponema pallidum subespecie pallidum cuya incidencia mundial es de 12 millones de casos por año, aproximadamente; de estos, más de dos millones se presentan en mujeres gestantes, siendo la sífilis congénita la complicación más grave de esta infección en el embarazo.Objetivo. Detectar la presencia de T. pallidum subespecie pallidum en muestras clínicas para el diagnóstico de sífilis congénita mediante reacción en cadena de la polimerasa (PCR) anidada y determinar su concordancia con las pruebas serológicas.Materiales y métodos. Mediante PCR convencional y anidada, se amplificaron tres genes diana (polA, 16S ADNr y TpN47) y se confirmaron los productos de amplificación de los genes TpN47 y polA por secuenciación. Las pruebas serológicas empleadas fueron la VDRL (Venereal Disease Research Laboratory), la de reagina plasmática rápida (Rapid Plasma Reagin, RPR) y la de aglutinación de partículas para Treponema pallidum (Treponema pallidum Particle Agglutination Assay, TPPA).Resultados. La sensibilidad para la PCR convencional fue de 52 pg y, para la PCR anidada, de 0,52 pg. La especificidad con los iniciadores TpN47 y polA fue de 100 %; los resultados de la secuenciación mostraron una identidad de 97 % con T. pallidum. En 70 % de las muestras, los resultados de las pruebas serológicas y la PCR anidada concordaron.Conclusión. El gen TpN47 resultó ser el mejor blanco molecular para la identificación de T. pallidum. La PCR anidada se presenta como una alternativa de diagnóstico molecular promisoria para el diagnóstico de sífilis congénita.
Introduction: Congenital syphilis is a major public health problem, and early diagnosis and treatment are necessary to prevent it. Penicillin G benzathine is the treatment of choice in pregnant women; however, it may fail to prevent fetal infection, as in the present case. Case presentation: Male newborn, son of an HIV negative mother with gestational syphilis (venereal disease research laboratory (VDRL) 1:4 dilution, positive treponemal test) diagnosed at week 21 of gestation and treated with three doses of 2 400 000 IU of penicillin G benzathine. At delivery, the mother presented VDRL 1:1 dilution. The newborn was diagnosed with congenital syphilis due to VDRL 1:4 dilution, positive treponemal test, elevated aspartate aminotransferases, hyposthenuria, proteinuria, hematuria, and leukocyturia that resolved after treatment with crystalline penicillin for 10 days. The molecular testing in blood showed a high treponemal load. The VDRL test at 3 months was non-reactive. Conclusions: Preventing congenital syphilis with the recommended treatment for gestational syphilis may fail. Moreover, diagnosing this condition in an asymptomatic newborn is difficult. Therefore, clinical and serological tests are recommended to confirm whether maternal treatment was effective in the fetus.
Se emplearon quistes liofilizados de Cisticerco cellullosae para preparar un extracto antigénico, el cual se utilizó en la identificación de antígenos de Taenia soliumreconocidos por inmunoglobulinas G de pacientes con cisticercosis, por medio de la prueba de inmunoelectrotransferencia (EITB). Se estudiaron 193 muestras (105 líquidos cefalorraquídeos - LCR y 88 sueros) de individuos con diagnóstico de cisticercosis por hallazgos clínicos, epidemiológicos y serológicos mediante el inmunoensayo enzimático (ELISA). El análisis de las proteínas antigénicas por electroforesis, permitió identificar polipéptidos de 29, 45, 66, 96 y 116 kDa principalmente, así como también algunos menores de 29 kDa. Los de mayor frecuencia de aparición en muestras de suero y LCR fueron los de 29, 45 y 66 kDa. Por lo tanto, estos antígenos inmunodominantes, podrían emplearse con predilección en estudios epidemiológicos y clínicos de cisticercosis por western blot.
ResumenEl siguiente estudio estableció un inmunoensayo enzimático ELISA, usando partículas virales fraccionadas de una cepa de campo del virus IBR, este mostró una alta sensibilidad y especificidad y una baja tasa de falsos negativos y positivos. Los sueros bovinos positivos y negativos, con los cuales se normalizó la técnica de ELISA, fueron analizados por seroneutralización para determinar anticuerpos anti IBR; se recolectaron sueros negativos y positivos con títulos 1:4 a 1:128 y se consideró la dilución del suero bovino 1:50, como la dilución óptima para la técnica de ELISA. Por otra parte, se utilizó el coeficiente de variación BR, para analizar el grado de repetitibilidad de las absorbancias obtenidas de los sueros positivos y negativos, se obtuvo resultados menores del 20%, lo que indico una adecuada repetitibilidad y un bajo coeficiente de variación. La sensibilidad y especificidad encontrada fue de 96 y 98% respectivamente, teniendo como prueba de referencia la seroneutralización. Los resultados obtenidos, determinan que la prueba de ELISA con partículas virales fraccionadas de una cepa de campo, detecta anticuerpos anti IBR con alta sensibilidad y especificidad, siendo una herramienta para el mejoramiento diagnóstico de la enfermedad.Palabras Claves: ELISA, seroneutralización, rinotraqueitis bovina infecciosa, anticuerpos, mejoramiento diagnóstico. Abstract Evaluation of ELISA test use of field strain (antigen) to detect antibodies against InfectiousBovine Rhinotracheitis virus.The following study set up an enzymatic immune test (ELISA) using viral particles broke up of field strain of IBR virus; it showed a high sensibility and specificity and low rate of false negatives and positives. Positive and negative bovine serum which ELISA technique was normalized, were analyzed by seroneutralization to determine anti IBR antibodies; It was collected positive and negative serum with titles 1:4 to 1:128 and was considered dilution of bovine serum 1:50 as optimal dilution to ELISA technique.On the other hand, coefficient of variation BR was used to analyze the rehearsing of absorbance obtained by positive and negative serum. <p>La biopelícula como un mecanismo de virulencia en Staphylococcus involucrada en infecciones intrahospitalarias es regulada por un represor negativo, responsable de la transcripción completa del operón. La búsqueda de dominios funcionales por modulación computacional de permitió hallar las secuencias peptídicas con actividad biológica análoga a la proteína <em>ica</em>R. Mediante biología computacional se diseñaron péptidos empleando el programa de predicción AntiBP (http: //www. imtech.res.in/ raghava/antibp/); la síntesis química se hizo por N<sup>α</sup>-Fmoc y se caracterizaron y purificaron tres moléculas por RP-HPLC y MALDI-TOF. Se evaluó su seguridad biológica mediante ensayo de actividad citotóxica realizada sobre macrófagos murinos de la línea J774 y la actividad hemolítica se determinó mediante el uso de glóbulos rojos. Los tres péptidos caracterizados IR1, IR2 e IR3, presentaron estructura secundaria predominantemente alfa helicoidal, alto grado de pureza y alto score antimicrobiano; además, mostraron baja toxicidad, evidenciada por la actividad citotóxica y hemolítica en las concentraciones ensayadas y en comparación con los controles usados, que permitiría su potencial uso como moléculas candidatas o principios activos con actividad análoga al represor nativo, frente a la biopelícula de los Staphylococcus.</p>
Fungal infections have increased in recent decades with considerable morbidity and mortality, mainly in immunosuppressed or admitted-to-the-ICU patients. The fungal resistance to conventional antifungal treatments has become a public health problem, especially with Candida that presents resistance to several antifungals. Therefore, generating new alternatives of antifungal therapy is fundamental. One of these possibilities is the use of antimicrobial peptides, such as LL-37, which acts on the disruption of the microorganism membrane and promotes immunomodulatory effects in the host. In this study, we evaluated the in vitro antifungal activity of the LL-37 analogue peptides (AC-1, LL37-1, AC-2, and D) against different Candida spp. and clinical isolates obtained from patients with vulvovaginal candidiasis. Our results suggest that the peptides with the best ranges of MICs were LL37-1 and AC-2 (0.07 µM) against the strains studied. This inhibitory effect was confirmed by analyzing the yeast growth curves that evidenced a significant decrease in the fungal growth after exposure to LL-37 peptides. By the XTT technique we observed a significant reduction in the biofilm formation process when compared to yeasts untreated with the analogue peptides. In conclusion, we suggest that LL-37 analogue peptides may play an important antimicrobial role against Candida spp.
