An ultra performance liquid chromatography (UPLC)-tandem mass spectrometry (MS/MS) method was developed for the determination of the Alternaria toxins tenuazonic acid, alternariol, alternariol monomethyl ether, altenuene, altertoxin I and tentoxin. Owing to its instability, altenusin could not be determined. The sample preparation includes an acidic acetonitrile/water/methanol extraction, followed by SPE clean-up step, before injection into the UPLC-MS/MS system. The separation was made on an Acquity UPLC column using a water/acetonitrile gradient with ammonium hydrogen carbonate as a modifier. Matrix compounds of real samples led to enhancement as well as suppression of the target compounds, depending on analyte and matrix. The recoveries were between 58 and 109% at a level of 10 μg/kg. Eighty-five tomato products, consisting of peeled and minced tomatoes, soup and sauces, tomato purées and concentrates, ketchup as well as dried and fresh tomatoes, were taken from the Swiss market in 2010. Tenuazonic acid was found most frequently (81 out of 85 samples) and in the highest levels of up to 790 μg/kg. Alternariol and alternariol monomethyl ether were found in lower concentrations, ranging from <1 to 33 μg/kg for alternariol and <5 to 9 μg/kg for alternariol monomethyl ether. Only a few samples were positive for altenuene and tentoxin. Altertoxin I was never detected.
Die gesundheitsfördernde Wirkung polyphenolreicher Pflanzenextrakte ist Thema vieler wissenschaftlicher Veröffentlichungen. So wird berichtet, dass das Spülen mit polyphenolhaltigen Getränken, wie grünem oder schwarzem Tee, Traubensaft oder Wein, die Bakterienadhäsion an die Zahnhartsubstanz reduziert. Am effektivsten erwies sich dabei Cistus-incanus-Tee [1], ein Tee, bereitet aus den oberirdischen Teilen der mediterranen Zistrose, die schon seit der Antike als Heilpflanze bekannt ist.Neben Cistus incanus L. gibt es eine Vielzahl weiterer Cistus-Arten, deren Polyphenolzusammensetzung bisher allerdings nur lückenhaft beschrieben worden ist [2]. Im vorgestellten Projekt wurden daher neben 4 Cistus-incanus-Tees des Handels 23 verschiedene Cistus-Arten, Subspezies und Hybriden mit HPLC-DAD-MS / MS untersucht. 43 phenolische Verbindungen, überwiegend Ellagitannine und Flavonol-Glykoside, wurden identifiziert, 33 dieser Substanzen auch quantifiziert. Die Polyphenolprofile der untersuchten Cistus-Pflanzen unterschieden sich deutlich, so dass die Cistus-Proben in drei Gruppen eingeteilt werden konnten: Gruppe 1 fiel durch hohe Gehalte an Ellagitanninen auf, in Gruppe 2 dominierten Flavonol-Glykoside und in Gruppe 3 waren sowohl hohe Ellagitannin-als auch Flavonol-Glykosid-Gehalte vorhanden [3].Extrakte der jeweiligen Cistus-Proben sowie einzelne isolierte Polyphenolfraktionen wurden in vitro auf ihre antibakterielle Wirkung gegen Streptococcus mutans, den Leitkeim der Kariogenese, geprüft. OriginalarbeitBestimmung von 11 β-Lactamen in Fleisch mittels dispersiver SPE und UPLC-MS/MS Methoden Dispersive SPE [3]3 g homogenisierte Fleisch-/Leberprobe wurden mit 27 mL eines Gemisches aus Acetonitril/Wasser (4 : 1, v / v) auf dem Vortex bei 3000 U/min. intensiv und danach auf der Schüttelma-schine 15 Min. bei 300 U/min. geschüttelt und extrahiert. Danach wurde die Mischung zentrifugiert und 20 mL Überstand in einem PP-Tube mit 1 g C 18 -Sorbens Bakerbond (Octadecyl 40 µm) von J. T. Baker für ca. 30 s bei 1800 U/min geschüttelt. Nach erneutem Zentrifugieren wurden 10 mL überstehende Lö-sung mit Stickstoff schonend auf etwa 0.7 mL eingedampft und das Konzentrat mit Reinstwasser auf 1 mL ergänzt. Dieser Extrakt wurde über einen 0.2 µm PP-Spritzenfilter für die UPLC-MS/MS-Bestimmung filtriert.Konventionelle SPE nach [6] 3 g homogenisierte Fleisch/Leberprobe werden mit 27 mL eines Gemisches aus Acetonitril/Wasser (4 : 1, v / v) auf dem Vortex bei 3000 U/min. intensiv und danach auf der Schüttelmaschi-ne 15 Min. bei 300 U/min. extrahiert. Nach dem Zentrifugieren der Mischung wurden 25 mL Überstand mit 12.5 mL Hexan versetzt und 30 s auf dem Vortex bei 1800 U/min. geschwenkt. Nach erneutem Zentrifugieren wurde die obere Hexanphase verworfen und 20 mL der unteren Phase wurden zur Trockene eingedampft. In 20 mL 0.05 M NaH 2 PO 4 -Lösung (pH = 8.5) wurde der Rückstand gelöst, der pH-Wert der Lösung kontrolliert und nötigenfalls mit 0.2 M NaOH auf pH = 8.5 eingestellt. Über eine vorkonditionierte Oasis HLB-SPE-Kartusche (500 mg, 6cc) von Waters wurde die ganze Lösung filtriert. Die Elution der Analyten erfolgte mit 3 mL Acetontril/Wasser (1 :1 , v / v). Das Eluat wurde bei 40 °C mit Stickstoff auf 1,5 mL eingedampft und mit Reinstwasser auf 2.0 mL ergänzt. Nach der Filtration über einen 0.2 µm PP-Spritzenfilter erfolgte die LC-MS/MS-Bestimmung.
