RESUMOO potencial da cultura do amendoim forrageiro associada aos fungos micorrízicos arbusculares (FMA) tem sido objeto de alguns estudos, porém a influência do genótipo sobre essa associação é pouco relatada. O objetivo deste trabalho foi determinar a densidade de esporos, riqueza de espécies, a frequência e ocorrência relativa de FMAs associados a genótipos de amendoim forrageiro. Foram coletadas amostras simples de solo de 45 genótipos pertencentes ao Banco Ativo de Germoplasma na Embrapa Acre. As amostras de solo foram coletadas a 5 cm de profundidade, com três repetições em delineamento inteiramente casualizado. As amostras de solo foram levadas para o Laboratório de Micorrizas da Embrapa Agrobiologia, para determinação da densidade de esporos e identificação das espécies de FMAs. Foi realizada análise de variância e teste de Scott-Knott. Destacaram-se quatro genótipos de A. pintoi e dois híbridos interespecíficos, que apresentaram maior densidade de esporos. Foi verificada a ocorrência de 21 espécies de FMAs nas amostras de solo. A riqueza variou entre três e dez espécies. Três espécies de FMAs apresentaram elevada frequência relativa: Glomus macrocarpum (100,0%), Acaulospora tuberculata (97,8%) e Racocetra verrucosa (88,99%). Conclui-se, assim que: (i) Existe variabilidade genética entre os genótipos de amendoim forrageiro quanto à promoção da esporulação e riqueza de espécies de FMAs nas suas rizosferas; (ii) As espécies de FMAs Glomus macrocarpum, Acaulospora tuberculata, Racocetra verrucosa possuem alta presença na rizosfera dos genótipos de amendoim forrageiro, devendo serem estudadas visando sua introdução na cultura. ABSTRACT:The objective of this study was to test the efficiency of preservation and maceration methods for Euterpe precatoria leaflet tissue to obtain genomic DNA for molecular studies. The leaflets of E. precatoria were collected in an experimental field at Embrapa Acre, Brazil. The study was conducted in a completely randomized design with 10 replicates, in a 12 × 2 factorial structure, with 12 storage treatments (fresh; lyophiliser 3 days; refrigerator 3, 5, and 7 days; silica gel 7, 10, 20, and 30 days; and transport buffer 3, 5, and 7 days) and two leaf tissue maceration methods (liquid nitrogen and the TissueLyser®). Statistically significant differences in the obtained DNA concentration were found between the maceration and storage treatments. The TissueLyser® macerator produced higher DNA concentrations when compared to liquid nitrogen. For the storage treatments, five groups were formed based on DNA concentration when macerated with the TissueLyser® and two groups when macerated with liquid nitrogen. The DNA concentrations ranged from 285.00 ng/µL (7 days in transport buffer) to 702.00 ng/µL (30 days in silica gel) when the leaflets were macerated with liquid nitrogen, and from 572.73 ng/µL (30 days in silica gel) to 2,850.00 ng/µL (3 days in lyophiliser) using the TissueLyser® macerator. The DNA purity (A260/A280 nm) varied from 1.30 to 1.70 when the leaflets were macerated with liquid nitrogen and from 1.30 to 1.90 with the TissueLyser® macerator. Despite the variations in leaf tissue preservation and DNA concentration, all treatments were effective for DNA isolation and it was possible to amplify genomic regions of microsatellite markers by PCR. It was concluded that leaflets of E. precatoria stored in a lyophiliser and processed with an automatic macerator resulted in satisfactory DNA for molecular studies.

É perceptível a relevância da iniciação científica na formação dos estudantes, uma vez que além de aproximarem os alunos da pesquisa, proporciona ricas experiências acadêmicas e de vida. O objetivo deste trabalho foi investigar a perspectiva de alunos, docentes e gestores sobre os impactos da iniciação científica na formação dos alunos dos cursos técnicos de nível médio do Instituto Federal do Acre – Campus Cruzeiro do Sul. A pesquisa classifica-se como explicativa, sendo utilizado o método quali-quantitativo, em que foram aplicados questionários semiestruturados a seis egressos que foram bolsistas do Programa Institucional de Bolsa de Iniciação Científica na modalidade Júnior (PIBIC Jr), a 14 docentes e três integrantes da equipe gestora. Os resultados evidenciam que os alunos compreendem a importância da iniciação científica para a sua formação e que, além de aproximá-los da ciência, habilidades como escrita, leitura, trabalho em equipe e expressão oral são melhoradas. A maioria dos docentes também ratifica essa importância da iniciação científica nos cursos técnicos, no entanto relatam que há limitações às atividades de pesquisa, sendo a infraestrutura inadequada e a falta de tempo, apontadas pela maioria dos entrevistados. A equipe gestora também declara ser importante a iniciação científica, concorda haver fatores limitantes à pesquisa no campus e citaram seus esforços na busca da minimização e/ou resolução destas problemáticas.
