Os métodos de avaliação da aptidão agrícola tem como objetivo orientar o uso adequado das terras, porém seus resultados apresentam elevado grau de subjetividade devido a serem dependentes da experiência do usuário. Neste trabalho objetivou-se avaliar o uso de um algoritmo para interpretação de propriedades do solo e características da paisagem e compará-lo com a interpretação obtida por um grupo controle, constituído por seis especialistas. Foram utilizadas amostras de solo coletadas por tradagem ao lado dos perfis pedológicos da IX Reunião Brasileira de Correlação e Classificação de Solos, de modo a obter os dados de entrada do algoritmo, enquanto os avaliadores utilizaram os dados físicos, químicos e morfológicos dos perfis pedológicos. A avaliação do grupo controle não identificou as potencialidades para o uso dos solos na Amazônia de forma consistente, enquanto, o algoritmo mostrou-se promissor na avaliação da aptidão para Latossolo, Argissolos, Vertissolos e Luvissolo, no entanto, ainda requer revisões em algumas regras de interpretação para uma avaliação mais coerente da aptidão, principalmente, para o Espodossolo. ABSTRACT:The objective of this study was to test the efficiency of preservation and maceration methods for Euterpe precatoria leaflet tissue to obtain genomic DNA for molecular studies. The leaflets of E. precatoria were collected in an experimental field at Embrapa Acre, Brazil. The study was conducted in a completely randomized design with 10 replicates, in a 12 × 2 factorial structure, with 12 storage treatments (fresh; lyophiliser 3 days; refrigerator 3, 5, and 7 days; silica gel 7, 10, 20, and 30 days; and transport buffer 3, 5, and 7 days) and two leaf tissue maceration methods (liquid nitrogen and the TissueLyser®). Statistically significant differences in the obtained DNA concentration were found between the maceration and storage treatments. The TissueLyser® macerator produced higher DNA concentrations when compared to liquid nitrogen. For the storage treatments, five groups were formed based on DNA concentration when macerated with the TissueLyser® and two groups when macerated with liquid nitrogen. The DNA concentrations ranged from 285.00 ng/µL (7 days in transport buffer) to 702.00 ng/µL (30 days in silica gel) when the leaflets were macerated with liquid nitrogen, and from 572.73 ng/µL (30 days in silica gel) to 2,850.00 ng/µL (3 days in lyophiliser) using the TissueLyser® macerator. The DNA purity (A260/A280 nm) varied from 1.30 to 1.70 when the leaflets were macerated with liquid nitrogen and from 1.30 to 1.90 with the TissueLyser® macerator. Despite the variations in leaf tissue preservation and DNA concentration, all treatments were effective for DNA isolation and it was possible to amplify genomic regions of microsatellite markers by PCR. It was concluded that leaflets of E. precatoria stored in a lyophiliser and processed with an automatic macerator resulted in satisfactory DNA for molecular studies.
RESUMO. Diversas técnicas analíticas podem ser usadas na determinação carbono do solo, predominando no Brasil métodos baseados na oxidação da matéria orgânica na presença de dicromato de potássio em meio ácido ou na análise elementar. O objetivo deste trabalho foi avaliar a viabilidade da estimativa do teor de carbono em solos da bacia do Acre por meio de espectroscopia NIR combinada com calibração por análise multivariada. Foram utilizadas 190 amostras de solos, coletadas em diversas localidades da bacia do Acre, para testar a espectroscopia NIR na determinação de teor de carbono no solo, comparativamente aos métodos de oxidação por dicromato em meio ácido e a análise elementar. Tomando-se como referência o método do analisador elementar, verificase que o método da oxidação recuperou, em média, 63,8 % do carbono determinado pelo método de referência. Sugere-se que para determinação do teor total de carbono solo a partir do método da oxidação seja adotado o coeficiente de 1,55 para corrigir os valores para o total de carbono do solo. Quanto ao uso da espectroscopia NIR, o modelo desenvolvido para análise de carbono de solos da 2 bacia do Acre por espectroscopia NIR apresentou classificação boa segundo os valores de R e classificação excelente segundo os valores de RMSEC < RMSEP e RPD, tendo como referência o carbono determinado pelo método da análise elementar.
