Olefine mit innenstandiger Doppelbindung, die an den olefinischen C-Atomen keine Verzweigungen aufweisen (Typ l), reagieren niit N-Carbonyl-sulfamidsaurechlorid (2) unter Cycloaddition zu a.P-disubstit. P-Lactam-N-sulfochloriden 3. Die Anlagerung erfolgt rnit hoher Stereoselektivitat als cis-Addition; ihre Richtung wird von den Substituenten an der C=C-Bindung beeinflufit. Cyclische Olefine reagieren gleichartig. -Die P-Lactam-N-sulfochloride 3 werden durch Hydrolyse in die freien P-Lactame 5 -11 ubergefuhrt.Cruf 1) hat gefunden, dafi bei der Umsetzung von N-Carbonyl-sulfamidsaurechlorid (2) mit zahlreichen Olefinen meist schon unter milden Bedingungen P-Lactam-N-sulfochloride [Azetidinon-(2)-N-suIfochloride, 31 gebildet werdenz), die sich durch selektive hydrolytische Spaltung in die freien P-Lactame uberfuhren lassen :;).In dieser Arbeit werden Umsetzungen von Olefinen mit ,,innerer" Doppelbindung [Buten-(2), Penten-(2), Hexen-(2), Hepten-(2) und Hepten-(3)] sowie von cyclischen Olefinen [Cyclopenten, Cyclohexen, Cycloocten u. a.] mit 2 beschrieben3). Die Untersuchungen wurden unternommen, um 1) die P-Lactam-Synthese nach Lit.*) zu erweitern und 2) AufschluB uber den sterischen Verlauf und die Anlagerungsrichtung der Cycloaddition zu erhalten. ReaktionsverlaufDa Olefine vom Typ 1 im Vergleich zu Olefinen des Vinyliden-Typs sehr langsam reagieren, fiihrt man die Umsetzung in konzentrierter Losung oder ohne Losungsmittel durch. In verdunnter Losung gehorcht ihre Kinetik dem Gesetz 2. Ordnung, in konzentrierter Losung wird die Reaktion durch das Reaktionsprodukt stark beschleunigt.Grofiere Ansatze lassen sich nur bei geeigneter Reaktionsfiihrung und wirksamer Warmeableitung unter Kontrolle halten.Die Reaktionsgeschwindigkeit ist stark von der Polaritat des Reaktionsmediums abhangig: In polaren Losungsmitteln (Methylenchlorid, Acetonitril) verlauft die Umsetzung mehrere Zehnerpotenzen schneller als in wenig polaren (Hexan, Diathylather).
Während aus Alkyl‐ und Arylisocyanaten mit Orthocarbonsäureestern labile, beim Erhitzen wieder in die Komponenten zerfallende 1 : 1‐Addukte entstehen, bilden Sulfonyl‐isocyanate unter Abspaltung von Carbonsäureestern und Umlagerung N‐Sulfonyl‐N‐alkyl‐carbamidsäureester. Formal entstehen dabei die – durch direkte Umsetzung nicht zugänglichen – Anlagerungsprodukte von Dialkyläthern an Sulfonyl‐isocyanate. Die Reaktion eröffnet einen ergiebigen und einfachen Zugang zu dieser Verbindungsklasse. Aus den mit Chlorsulfonyl‐isocyanat entstehenden N‐Chlorsulfonyl‐N‐alkyl‐carbamidsäureestern werden durch partielle Verseifung N‐Alkyl‐sulfamidsäuren bzw. N‐Alkyl‐carbamid‐säureester erhalten.
Aus dem bekanntens) 4-Nitro-nicotins&urs I-oxyd (I) wird nach der ,,Anhydrid"-Methode das Amid I1 erhalten. Aus I1 lPDt sich rnit Na-Benzylat das 4-Benzyloxy-amid 111, mit Methylat die 4-Methoxyverbindung VI, rnit Acetylchlorid die 4-Chlorverbindung VIII a, mit Acetylbromid die 4-Bromverbmdung VIII b und durch selektive Hydrierung die 4-Aminoverbindung VIIIc herstellen. Das 4-Mercapto-nicotinsiiureamid-I -oxyd (VIII d) wird durch Thiohydrogenolyse von VIII b erhalten. Katalytische Hydrierung iiber Pd greift vorwiegend den Substituenten in 4-Stellung an (Nitro-, Benzyloxy-Gruppe. Chlor, Brom) unter Erhaltung der N-Oxyd-Funktion. Diese ltiRt sich, auDer bei der Nitroverbindung 11, durch Eisen in Essigsaure, besser katalytisch iiber Raney-Nickel selektiv abreduzieren. Die so u. a. hergestellten 4-Halogen-nicotinsaureamide XI und XI1 enthalten reaktives Halogen und lassen sich durch SHe oder S03HG zum 4-Mercapto-nicotinsSureamid (XVIII) oder zur 4-Sulfons%ure XVII umsetzen.Die im Zusammenhang rnit der Biochemie der Wasserstoffiibertragenden Coenzyme interessierenden Pyridon-(4)-und 4-Methoxy-pyridincarbonsaure-(3)-amide sind im hiesigen Laboratorium vor k u m m auf zwei verschiedenen Wegen synthetisiert worden2J). Bei einer dieser Synthesen3) benutzten wir das nach E. c. TAYLOR und J. S. DRISCOLL~) leicht zugangliche 4-Nitro-nicotinsaurel-oxyd (I), in welchem nach der dort angegebenen Vorschrift die Nitrogruppe gut durch verschiedene nucleophile Reste, u. a. auch durch den Benzyloxyrest, substituierbar ist. Die 4-Benzyloxysaure V wurde damals3) verestert, aus dem Ester das Amid und aus diesem durch katalytische Hydrierung uber das N-Oxyd der CHydroxyverbindung das Pyridon IV erhalten. Eine weitere Vereinfachung hat sich nun dadurch ergeben, daI3 man die Einfuhrung des Amidstickstoffs mit Ammoniak schon auf der Stufe der Nitrosaure I nach der Methode der gemischten Anhydrides) erreichen kann. Im so gewonnenen 4-Nitro-nicotinslureamid-1-oxyd (11) ist die Nitrogruppe mindestens ebenso leicht wie in I nucleophil substituierbar, so daD z. B. mit Na-Benzylat das 4-Benzyloxynicotinsaureamid-1-oxyd (III) und aus ihm durch katalytische Hydrierung das Pyridon IV hergestellt werden kann.

Die beiden in 4-Stellung oxydierten Derivate I und 11 des Nicotinsilureamids sind vor kuner Zeit von TH. WIELAND, 01. FEST und G. PPLEIDERER~), ausgehend vom 3-Nitro-emethoxy-Schmp. (aus H20): 257' (Zers.). Es dllrfte sich um das N-Oxyd des 4-Hydroxy-pyridio-carbansaure(3)-amids (H statt Benyl in 1V) handeln. Ber. fur C6H6N203 (154.1) C 46.75, H 3.93, N 18.18; gef. C 46.90, H 4.16. N 17.90. Die in 3-Stellung unsubstituierte Verbindung, das N-Oxyd von 4-Hydroxy-pyridin, ist auch durch katalyt. Hydrierung der Benxyloxyverbindung erhalten worden (E. SHAW, J. h e r . chem. SOC. 71, 68 [1949]).
