Львів, 79034, Україна Досліджували вплив цитрату ванадію на вміст продуктів пероксидного окиснення ліпідів, відновленого глутатіону, активність ензимів антиоксидантної системи та глюкозо-6-фосфатдегідрогенази у скелетних м'язах стегна щурів з алоксан-індукованим діабетом. Тварини були поділені на п'ять груп: I група -контрольна, II, III, IV та V -дослідні. Щурам І та ІІ груп давали пити чисту воду, III, IV та V групам впродовж місяця до питної води додавали розчин цитрату ванадію у концентраціях 0,125, 0,5 і 2,0 мкг V/мл води. У тварин усіх дослідних (II-V) груп експериментально викликали цукровий діабет внутрішньоочеревинним введенням алоксану (150 мг/кг маси тіла). У м'язах тварин ІІ групи з експериментальним діабетом спостерігалось зростання вмісту гідропероксидів і ТБК-активних продуктів та зниження активності ензимів антиоксидантного захисту (супероксиддисмутази, каталази і глутатіонредуктази), глюкозо-6-фосфатдегідрогенази та вмісту відновленого глутатіону, тоді як активність глутатіонпероксидази підвищувалася порівняно з тваринами контрольної групи. За випоювання щурам цитрату ванадію у різних концентраціях спостерігалося зниження вмісту, тоді як активність глутатіонпероксидази знижувалася (в ІІІ, IV та V групах) порівняно з показниками у м'язах тварин ІІ групи з експериментальним діабетом. Цитрат ванадію у досліджуваних концентраціях здійснює дозозалежний стабілізуючий вплив на стан прооксидантно-антиоксидантної системи у скелетних м'язах щурів з експериментальним діабетом. Animals were divided into five groups: I group -control, II, III, IV and V -experimental. The rats of I and II groups were given pure water to drink, whereas a solution of vanadium citrate at concentrations of 0.125, 0.5 and 2.0 μg V/ml of water was added to drinking water of III, IV and V groups within a month. The animals of all experimental (II-V) groups were induced diabetes by intraperitoneal injection of alloxan (150 mg/kg body weight).In muscles of the II group experimentally diabetic animals, the increase in the content of hydroperoxides and TBA-active products and the decrease in the activity of antioxidant protection enzymes (superoxide dismutase, catalase and glutathione reductase), glucose-6-phosphate dehydrogenase and the content of reduced glutathione were observed, while the activity of glutathione peroxidase increased as compared to control animals. Dose-dependent changes in protein metabolism in the blood and hematological parameters of pregnant rats under the effect of vanadium citrate are presented in the article. The animals were divided into five groups: group I – non-pregnant females, II – pregnant females consuming pure water without additives, III, IV, V – females which during the mating and pregnancy period received the solution of vanadium citrate at concentrations of 0.03, 0.125 and 0.50 μg V/mL water. The research findings show that in pregnant animals of group II, the level of urea and alkaline phosphatase activity increased, meanwhile aspartate aminotransferase activity decreased, as compared to the non-pregnant females of group І. The levels of total protein and albumin decreased; however, the content of β-globulins increased in the pregnant animals of group II, as compared with that in group I. Also, in the rats of group II, there was a decrease in hemolysis time, total content of erythrocytes and hemoglobin, the content of old and mature erythrocytes, while the content of young erythrocytes increased, as compared to group I. The platelet content and thrombocrit in rats of group II increased in comparison with group I. The content of leukocytes and lymphocytes in pregnant animals of group II decreased, while the content of granulocytes increased, in contrast to non-pregnant rats. Under the effect of vanadium citrate at concentrations of 0.03–0.50 μg V/mL, there was a significant increase in the maximum number of prohemolized erythrocytes, the time of maximum hemolysis was delayed by 0.4–0.6 min, as compared with the pregnant rats of group II. This did not affect the time of total hemolysis in rats of groups III and V, as compared with the pregnant animals in group II. Under the effect of vanadium citrate, an increase in the content of young erythrocytes was observed, as compared with group II. The hemoglobin content decreased at the concentration of 0.125 μg V/mL, while at the concentration of 0.50 μg V/mL it increased, as compared to the pregnant animals of group II. Also, under the effect of vanadium citrate there was a decrease in the mean hemoglobin concentration in the erythrocyte. In pregnant animals fed with vanadium citrate solutions, the platelet content and thrombocrit, the relative width of platelet distribution by volume decreased, as compared with the pregnant rats of group II. The content of leukocytes, lymphocytes and granulocytes under the effect of vanadium citrate increased, as compared with the pregnant animals in group II. Under the effect of vanadium citrate at the concentration of 0.03 μg V/mL, the level of albumin, creatinine and aspartate aminotransferase activity increased in blood plasma in comparison with group II. Meanwhile, at the concentration of 0.125 μg V/mL, the relative content of γ-globulins and aspartate aminotransferase activity increased, alkaline phosphatase activity and urea level decreased in comparison with group II. However at the concentration of 0.50 μg V/mL, the relative α- and γ-globulins content and aspartate aminotransferase activity increased, at the same time, the relative β-globulins content and urea level decreased in comparison with group II. Therefore, vanadium citrate normalizes the indicators of protein metabolism during pregnancy, thus it can be considered as a potential dietary drug for the pregnant.
The article presents a historical development path of the Laboratory of biochemistry of adaptation and ontogenesis of animals which began with the founding of the Ukrainian Research Institute of Physiology and Biochemistry of Farm Animals in 1960. At the beginning of its existence laboratory was called “The laboratory of higher nervous activity” and was headed by associate professor Dr. I. A. Medyanyk. Since 1962 it was called “The laboratory of neurohumoral regulation” and was headed by Dr. of science, Professor E. M. Berkovych. Since 1963 the laboratory was headed by Dr. of science, Professor Z. P. Skorodynsky. Since 1972 the laboratory was headed by Dr. M. D Hanin, since 1980 — Professor V. M. Holovach, since 1988 — academician of NAAS, Professor V. V. Snitynsky. In 1994 the laboratory was renamed into the laboratory of endocrine regulation. Since 2011 until now the laboratory is called “The laboratory of biochemistry of adaptation and ontogenesis of animals”. From 2011 to 2013 duties of the laboratory head were performed by Dr. O. M. Buchko. From 2013 until now the head of the laboratory is Dr. of science, senior researcher R. Ya. Iskra.

Тварини були розділені на п'ять груп: І група -контрольна, ІІ, ІІІ, ІV і V -дослідні. У тварин усіх чотирьох дослі-дних груп індукували експериментальний цукровий діабет. Щурам І та ІІ-ї груп давали пити чисту воду без добавок, а тваринам ІІІ, ІV і V груп протягом місяця до питної води додавали розчин цитрату ванадію в кількостях 0,125, 0,5 і 2,0 мкг V / мл води. У результаті проведених досліджень встановлено зростання рівня сечовини, активності лужної фос-фатази та аспартатамінотрансферази у плазмі крові тварин ІІ групи відносно контрольної. Вміст загального білка та α-глобулінів у ІІ групі знижував-ся, в той час як концентрація β-глобулінів -підви-щувалась, відносно І групи. За випоювання твари-нам цитрату ванадію спостерігалося вірогідне зни-ження сечовини, креатиніну, активності амінотран-сфераз та лужної фосфатази, порівняно з активніс-тю у тварин ІІ групи. Таким чином, за дії цитрату ванадію відбувається нормалізація білкового мета-болізму, порушення якого виникали за експеримен-тального цукрового діабету.Ключові слова: цитрат ванадію; діабет; біохі-мічні параметри; загальний білок; фракції білка.Зв'язок роботи з науковими програмами, планами, темами. Діабет характеризується надмірним рівнем циркулюючої глюкози внаслідок абсолютної чи відносної недостатності інсуліну. На сьогодні, люди страждають не тільки від діабету, а й від по-в'язаних з ним ускладнень [9]. В організмі хворих на цукровий діабет, крім порушення обміну вугле-водів і ліпідів, інтенсифікується білковий обмін, причому процес розпаду білка відбувається швид-ше, ніж синтезу, що призводить до його втрат [10].Сполуки ванадію мають здатність активувати інсуліновий сигнальний каскад через фосфорилю-вання ключових білків цього шляху в клітинах орга-нізму [11]. Ванадій розглядається як інсулінопідси-люючий агент, у зв'язку із конкурентним гальмуван-ням регуляторних фосфатаз, зокрема фосфатази 1B (pp1B) за діабету. Ці фосфатази є першими ен-зимами в інсулін-регуляторному каскаді і є чутливи-ми до пригнічення ванадієвмісними сполуками [4].Метою даної роботи було дослідити особли-вості білкового обміну в організмі щурів з експери-ментальним діабетом та визначити профілактичну дію цитрату ванадію на це захворювання.Матеріали та методи досліджень. Експери-менти проводилися згідно «Загальних етичних принципів експериментів на тваринах», ухвалених Першим національним конгресом з біоетики (Київ, 2001). У період вагітності організм зазнає фізіологічних, ендокринних та метаболічних змін, спостерігається дисбаланс мікроелементів, зокрема Ванадію. Для компенсації втрат цього мікроелементу та покращення метаболічних процесів у вагітних ми використовували органічну сполуку-цитрат ванадію. Метою нашого дослідження було вивчити вплив цитрату ванадію на глюкозо-6-фосфатдегідрогеназну та лактатдегідрогеназну активність у підшлунковій залозі та скелетному м'язі вагітних самок щурів. Дослідження проведені на самках білих лабораторних щурів, масою тіла 140-160 г, які були розділені на п'ять груп: І група-невагітні тварини, ІІ-вагітні самки, які споживали чисту воду без добавок, щури ІІІ, ІV, V груп-у період спаровування і вагітності отримували розчин цитрату ванадію у концентраціях, відповідно 3,75, 15,62 та 62,5 мкг V/кг маси тіла. Матеріалом для досліджень були гомогенати підшлункової залози та скелетного м'язу стегна вагітних самок щурів, у яких визначали глюкозо-6-фосфатдегідрогеназну та лактатдегідрогеназну активність. У підшлунковій залозі та скелетному м'язі вагітних самок щурів знижувалась глюкозо-6-фосфатдегідрогеназна активність, порівняно із невагітними. Це є наслідком пригнічення окиснення глюкози у пентозофосфатному шляху. Лактатдегідрогеназна активність у цих же тканинах у вагітних тварин зростала, порівняно із І групою. Зростання активності лактатдегідрогенази у вагітних вказує на активацію гліколізу та інтенсивне використання глюкози в енергетичному метаболізмі. За умов випоювання щурам цитрату ванадію у підшлунковій залозі глюкозо-6-фосфатдегідрогеназна активність вірогідно зростала у IV групі, тоді як лактатдегідрогеназна активність знижувалась-у V-й групі, порівняно з вагітними тваринами ІІ групи, які не вживали Ванадію. У м'язах вагітних тварин, яким випоювали Ванадій, глюкозо-6-фосфатдегідрогеназна активність вірогідно знижувалась у ІІІ та ІV групах, проте зростала у V групі, тоді як лактатдегідрогеназна активність знижувалась у ІІІ групі та зростала у V-й, порівняно із вагітними тваринами у ІІ групі. Ванадій дозозалежно сприяє наближенню глюкозо-6-фосфатдегідрогеназної та лактатдегідрогеназної активності у підшлунковій залозі та скелетних м'язах вагітних тварин до значень їх активності у невагітних тварин. Він зумовлює нормалізуючий вплив на лактатдегідрогеназну активність та сприяє окисненню глюкози у пентозофосфатному шляху. Це дозволяє розглядати цей мікроелемент як потенційний дієтич-БІОЛОГІЯ Г. В. The aim of the work was to study the effect of vanadium citrate on the activity of glucose-6-phosphate dehydrogenase, lactate dehydrogenase, estradiol content and to establish correlations between the activity of the studied enzymes and the estradiol content in pregnant female rats under the effect of vanadium compound. The study was carried out on 25 female Wistar rats, divided into five groups: group I - non-pregnant, group II - pregnant consuming pure water without additives, groups III, IV, V - females which during the mating and pregnancy periods received the solution of vanadium citrate at the concentrations of 3.75, 15.63 and 62.5 μgV/kg of body weight, respectively. Glucose-6-phosphate dehydrogenase, lactate dehydrogenase and estradiol content increase in pregnant females of group II compared to group I of non-pregnant animals. In pregnant females, the effect of vanadium citrate resulted in the decrease in the activity of glucose-6-phosphate dehydrogenase and lactate dehydrogenase in the blood of animals in groups IV and V, and the content of estradiol in the animals of all three experimental groups (III, IV, V), compared to group II of pregnant females. The values of glucose-6-phosphate dehydrogenase activity in the animals of group IV, which received the solution of vanadium citrate at the concentration of 15.63 μgV/kg of body weight, was similar to that of the animals in group I. This may indicate the normalizing effect of vanadium citrate on the activity of glucose-6-phosphate dehydrogenase at the indicated concentration, which is probably due to the ability of the vanadium microelement to inhibit the activity of this enzyme. Vanadium citrate at the concentration of 15.63 μgV/kg of body weight caused the recovery of the enzyme activity to the values of the activity in group І.
