Brazilian Santa Inês (SI) sheep are very well-adapted to the tropical conditions of Brazil and are an important source of animal protein. A high rate of twin births was reported in some SI flocks. Growth and Differentiation Factor 9 (GDF9) and Bone Morphogenetic Protein 15 (BMP15) are the first two genes expressed by the oocyte to be associated with an increased ovulation rate in sheep. All GDF9 and BMP15 variants characterized, until now, present the same phenotype: the heterozygote ewes have an increased ovulation rate and the mutated homozygotes are sterile. In this study, we have found a new allele of GDF9, named FecG(E) (Embrapa), which leads to a substitution of a phenylalanine with a cysteine in a conservative position of the mature peptide. Homozygote ewes presenting the FecG(E) allele have shown an increase in their ovulation rate (82%) and prolificacy (58%). This new phenotype can be very useful in better understanding the genetic control of follicular development; the mechanisms involved in the control of ovulation rate in mammals; and for the improvement of sheep production.
We performed a meta-analysis of over 30 case-control studies of association between schizophrenia and a bi-allelic, Bal I polymorphism in exon 1 of the dopamine D3 receptor gene. We observed a significant excess of both forms of homozygote in patients (P = 0.0009, odds ratio (OR) = 1.21, 95% Confidence Interval (CI) = 1.07-1.35) in the combined sample of 5351 individuals. No significant heterogeneity was detected between samples and the effects did not appear to be the product of publishing bias. In addition we undertook an independent, familybased association study of this polymorphism in 57 parent/proband trios, taken from unrelated European multiplex families segregating schizophrenia. A transmission disequilibrium test (TDT) showed a significant excess of homozygotes in schizophrenic patients (P = 0.004, odds ratio (OR) = 2.7, 95% CI = 1.35-5.86). Although no significant allelic association was observed, a significant association was detected with the 1-1 genotype alone (P = 0.02, OR = 2.32, 95% CI = 1.13-4.99). In addition when the results of the family-based association study were included in the meta-analysis, the homozygosity effect increased in significance (P = 0.0002, OR = 1.23, 95% CI = 1.09-1.38). The results of the meta-analysis and family-based association study provide independent support for a relationship between schizophrenia and homozygosity at the Bal I polymorphism of the D3 receptor gene, or between a locus in linkage disequilibrium with it.
Three sheep farms were used in the Distrito Federal, Central Brazil, to study the occurrence of parasites in the feces. A total of 1798 collections were taken over the period of a year. A total of 1205 were taken in Santa Inês breed (SI) in all three farms, 323 in Bergamasca (Berg), 54 in Ile de France, 49 in IlexSI, 103 in Morada Nova (MN) and 64 in TexelxSI, these last five groups being on a single farm. The animals were drenched soon after weaning and feces collected every 3 weeks to calculate fecal egg count (FEC), at least on two occasions on each animal. In some cases, blood was collected to determine packed cell volume (PCV) at fecal collection. Fixed effects included farm, breed/genetic group within farm, animal age (months), birth type (simple, twin) and sex. (Co)variance components were estimated for Santa Inês sheep using restricted maximum likelihood under an animal model. FECs were affected by month and farm showing that climate and management are important sources of variation for the parasites studied. While age and birth type of the lambs did not affect infection level, their genetic group was important, showing that breeding strategies can help control these parasites. Heritabilities for infection level in the sheep varied between 0.09 for Strongyloides and 0.31 for Moniezia expansa. Genetic selection strategies for sheep aimed at reducing these infections should result in more resistant animals.
In the hatchery-bred tambaqui Colossoma macropomum, spontaneous semen release does not occur, and hand-stripping produces reduced semen volume. The goal of this work is to evaluate the effects of hormonal induction with carp pituitary extract (CPE) on both qualitative (visual aspect, pH, motility, viability and morphological abnormalities) and quantitative (volume, concentration and number of spermatozoa per ejaculate) traits of tambaqui semen. Eleven males were treated with CPE (induced), and 11 were left untreated as a control (non-induced). All analysed parameters except motility and percentage of viable spermatozoa presented significant differences (p < 0.05) between the induced and non-induced treatments. CPE induction resulted in a 25-fold increase in semen volume and a 10-fold increase in the number of spermatozoa collected. However, both sperm concentration and the frequency of sperm with morphological abnormalities (commonly detached heads or bent tails) were significantly lower in CPE-induced fish. The hormonal induction of tambaqui males with CPE is efficient and positively influences some qualitative and quantitative properties of semen. Additionally, semen collection via gentle abdominal massage occurs more readily in CPE-induced fish.
Seminal features of tambaqui were evaluated after hormonal induction of spermiation with common carp pituitary extract. Seventeen adult (6.1 ± 0.9 kg, 62 ± 6 cm) males were collected from earthen ponds and transported to indoor concrete tanks. Semen was evaluated according to volume, pH, osmolality, motility, concentration, viability, sperm morphometry and morphological abnormalities. The semen of tambaqui was white and milky. The volume was 10.2 ± 5.1 mL at pH 8.0 ± 0.1, and yielded a concentration of 9.1 · 10 9 spermatozoa mL )1 while seminal plasma osmolality was 260 ± 7.3 mOsm kg )1 . The percentage of viable sperm cells was determined with 97.0 ± 2.0% and 87.0 ± 8.0% using an eosin-nigrosin staining and fluorescent live-dead kit (propidium iodide and SYBR-14), respectively. A negative correlation ()0.65) between semen pH and sperm motility was observed suggesting this feature might influence the tambaqui sperm kinetics. Morphometrically the spermatozoa were on average 35.48 ± 1.55 lm long with a roundish head (mean length: 2.73 ± 0.21 lm; mean width: 2.58 ± 0.18 lm; n = 250 spermatozoa), without an acrosome, and presented a long midpiece (2.90 ± 0.52 lm) and flagellum (29.84 ± 1.63 lm). About 15.8% of the spermatozoa carried morphological abnormalities, with bent tail being the most frequent defect (7.81 ± 3.12%). The characterization of tambaqui semen favors the development of more precise and efficient procedures for its analysis and utilization in controlled breeding.
