1. Der histochemische Aktivitätsnachweis Pyridinnukleotid (PN)-spezifischer Dehydrogenasen mit der üblichen Tetrazolium-Technik ist unvollständig. Möglichkeiten der Artefaktbildung ergeben sich aus dem Mechanismus der zweistufigen Nachweisreaktion: Der von der PN-spezifischen Dehydrogenase katalysierten Oxydation des Substrats ist die von einer PN-spezifischen Tetrazolium-Reduktase katalysierte Übertragung des Wasserstoffs vom reduzierten Pyridinnukleotid auf den oxydierten Farbstoff (Tetrazoliumsalz) nachgeschaltet. Die Abscheidung des reduzierten Farbstoffs (Formazan) erfolgt zwangsläufig am Ort der Tetrazolium-Reduktase («Dia- phorase»). Eine exakte Ortung Pyridinnukleotid-spezifischer Dehydrogenasen ist auf diesem Wege so gut wie ausgeschlossen. Ein weiterer Nachteil der Methode beruht auf Diffusionsartefakten : Die Diffusion löslicher Dehydrogenasen führt zur Verlagerung der Enzymorte. 2. Ein neu entwickeltes «Gelschicht-Verfahren» ermöglicht den histochemischen Aktivitätsnachweis PN-spezifischer Dehydrogenasen direkt am Ort ihrer Wirkung. Der Aktivitätsnachweis ist unabhängig von der Wirkung der gewebseigenen Tetrazolium-Reduktasen, da als Redox-Mediator Phenazin-Methosulfat verwendet wird. Phenazin-Methosulfat, oxydiertes Pyridinnukleotid, reduziertes Substrat der nachzuweisenden Dehydrogenase und das Tetrazoliumsalz sind homogen in einer dünnen und planen Schicht von Agargel verteilt, die im Kontakt mit dem Gewebs- schnitt inkubiert wird. 3. Die an der Rattenleber durchgeführten Untersuchungen dienten dem Studium der intralobulären Aktivitätsverteilung von Oxydoreduktasen. Die Ergebnisse führen zur Unterscheidung bestimmter Verteilungstypen: a) Glyceraldehydphosphat-Dehydrogenase,Glycerin-l-phosphat-Dehydrogenase, Malat-Dehydrogenase, Glucose-6-phosphat-Dehydrogenase, «malie enzyme» und Glycerin-l-phosphat-Oxydase zeigen eine gleichmäßige Verteilung ihrer Aktivitäten im Bereich des Leberläppchens. b) Glutamat-Dehydrogenase und die TPN-spezifische Isocitrat-Dehydrogenase weisen höhere Aktivitäten im perizentralen als im periportalen Feld des Leberläppchens auf.
In a first paper, the structural and functional relationship of granulosa and theca of follicles during early development stages was reported. On the special question of whether and from which moment these two tissue formations are involved in the steroidbiosynthesis, our electronmicroscopic examinations have given a further insight to this. Neither the granulosa nor the theca folliculi of primordial, primary and secondary follicles show definite morphologic submicroscopic criteria of the steriodbiosynthesis. In the resting tertiary follicle, the electronmicroscopy defines the theca cells as steroidbiosynthetic active cells, whereas the granulosa cells of this stages of follicle demonstrate the morphologic characteristics of protein-synthetic active cells. Our, under physiological conditions, systematically conducted light and electronmicroscopic studies of preovulatory and freshly ruptured follicles showed remarkable structural changes in the granulosa, as well as in the theca folliculi. For the follicle granulosa cells of the preovulatory follicle, the studies could demonstrate a structural transformation process of proteinsynthetic active to steroidbiosynthetic active cells. This transformation process can be seen in the nucleus. Especially recognizable is the continuous change from rough to smooth endoplasmatic reticulum in the cytoplasm of most of the follicle granulosa cells, which apart from that, often displayed whorle-like formations. Furthermore, we noticed striking changes of the paraplasmatic structures, especially of the fat, present in various stages in the cytoplasm of the follicle granulosa cells. We also noticed large mitochondria, which showed a lot of vesicular cristae. Numerous existing Golgi-fields contained many little homogenous fat-like droplets, which were surrounded by a thin osmiophilic membrane. This recognizable transformation process of the proteinsynthetic active granulosa cells to the steroid cells is quite completed in freshly ruptured follicles, according to our examinations. Although the theca cells have already been defined submicroscopically in the resting tertiary follicle as steroidbiosynthetic active cells, we see in this cellgroup in the preovulatory, as well as in the freshly ruptured follicle a remarkable conspicuous size-increase of the mitochondria. Apart from that, there are wide areas which are solely occupied by smooth endoplasmatic reticulum, whereas other structures, for example fat, are scarely seen. As our examinations have shown, there is a close relationship between the transformation process in the granulosa and the theca of preovulatory follicles and the theca of preovulatory follicles that, the increasing concentration of progesteron in serum, which begins prior to the ovulation can be regarded as a product of the follicle granulosa cells being transformed to steroid cells.
Auf histochemischem Wege wurden Oxydoreduktasen des energieliefemden Stoffwechsels in quergestreiften Muskeln, insbesondere von Locusta migratoria, lokalisiert. Die Aktivitäten Pyridinnukleotid-spezifischer Dehydrogenasen wurden mit Hilfe des «Gelschicht-Verfahrens» geortet. Hinsichtlich der extramitochondrialen Dehydrogenasen lassen sich 2 verschiedene Lokalisationstypen unterscheiden: Laktat-Dehydrogenase und Glyceraldehyd-phosphat-Dehydrogenase können in verschiedenen Insektenmuskeln, aber ebenso auch in quergestreifter Kaninchenmuskulatur mit dem Hauptteil ihrer Aktivitäten im Bereich der isotropen Zonen der Myofibrillen nachgewiesen werden. Glycerin-1-phosphat-Dehydrogenase und «malie enzyme» sind dagegen im epi- bzw. perimitochondrialen Raum lokalisiert. Der gleiche Lokalisationstyp trifft auch für die extramitochondrialen Aktivitäten der Malat-Dehydrogenase und der TPN-spezifischen Isocitrat-Dehydrogenase zu. Offensichtlich ist die perimitochondriale Anordnung charakteristisch für Enzyme, die dem intramitochondrialen Enzymapparat funktionell verbunden sind. Die Lokalisation der 2 Dehydrogenasen des glykolytischen Stoffwechsels (Gly-ceraldehydphosphat-Dehydrogenase und Laktat-Dehydrogenase) innerhalb der in- terfilamentären Räume der isotropen Zonen läßt vermuten, daß auch andere, möglicherweise die Gesamtheit der Enzyme des Embden-Meyerhof-Wegs am gleichen Ort vertreten sind. Diese Lokalisation ist nicht nur von Interesse für die Bereitstellung energiereichen Phosphats über die Substratkettenphosphorylierung der Glykolyse im Bereich der isotropen Zonen, sondern hat möglicherweise auch Bedeutung für die Mechanik der Muskelkontraktion.
