Dendrobium spp. are precious medicinal plants, used in China for thousands of years as health foods and nutrients. Polysaccharides are the main effective ingredients in Dendrobium plants. In this study, the chemical characteristics and the effects of crude polysaccharides (CPs) from five species of Dendrobium on macrophage function were investigated and compared in vitro for the first time. Chemical characteristic studies showed that CPs from different species of Dendrobium were diverse, displaying widely varied Mw distributions and molar ratios of monosaccharides. Their effects on macrophage functions, such as promoting phagocytosis, release of NO and cytokines IL-1α, IL-6, IL-10 and TNF-α, were also different. Moreover, CPs from D.
officinale, especially collected from Yunnan Province, exerted the strongest immunomodulatory activities and could be explored as a novel potential functional food. The diverse chemical characteristics of CPs from different species of Dendrobium might contribute to their varied effects on macrophage functions, which should be further investigated.
The sesquiterpenoids are one of major groups of antioxidants in Curcuma besides curcuminoids. However, the real substances contributing to the antioxidant activity are still unknown. In this paper, the antioxidant activity of sesquiterpenoids in four species and two essential oils from Curcuma genus was determined and compared based on TLC separation and DPPH bioautography assay. Their antioxidant capacities were quantitatively evaluated using densitometry with detection at 530 nm (λreference = 800 nm) using vitamin C as reference. The results showed that Curcuma longa rhizomes had the highest antioxidant capacity while C. phaeocaulis presented the lowest one among the four species of Curcuma. Moreover, essential oil of C. wenyujin showed higher antioxidant potential than that of C. longa. The main TLC bands with antioxidant activity of the four species of Curcuma were collected and characterized using GC-MS, and thus curzerene, furanodiene, α-turmerone, β-turmerone and β-sesquiphellandrene were determined as major sesquiterpenoids with antioxidant activity in Curcuma.
Fructooligosaccharides (FOS), which are regarded as functional ingredients, are commonly classified as dietary fibers in many countries. However, few analytical methods for separation and analysis of individual FOS in plants, crops, and food products have been developed. In this study, a simple, rapid, and sensitive high performance liquid chromatography with charged aerosol detector (HPLC-CAD) method was developed for simultaneous determination of 11 inulin-type FOS with degree of polymerization (DP) 3-13 in different samples. The separation was performed on a Waters XBridge Amide column (4.6 × 250 mm i.d., 3.5 μm) with gradient elution. All calibration curves for investigated analytes showed good linear regression (R(2) > 0.9962). Their limits of detection (LOD) and quantification (LOQ) were in the ranges 0.4-0.6 μg/mL and 1.4-2.3 μg/mL, respectively. The recoveries ranged from 94.0% to 114.4%. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was applied to qualitative analysis of FOS in different samples. The developed method was successfully applied to analysis of 11 FOS in different samples of plants from Compositae, Campanulaceae, and Rubiaceae families. The developed HPLC-CAD nethod with microwave-assisted extraction can be used for quantitative analysis of FOS and is helpful for quality control of plants containing FOS.
