INTRODUÇÃO: Dengue é um importante problema de saúde pública, em vários países, e tem como principal vetor o Aedes aegypti, mosquito mais adaptado às áreas urbanizadas. Apresenta-se, pela primeira vez, as alterações ultraestruturais em larvas de 3º estádio, desse mosquito, causadas pelos larvicidas naturais, um diterpeno labdano, extraído de Copaifera reticulata, e uma fração rica em taninos catéquicos, extraída de Magonia pubescens, evidenciando o mecanismo de ação dessas substâncias. MÉTODOS: Os experimentos foram realizados com larvas de 3º estádio em solução de 0,9ppm, do diterpeno (3-β-acetoxylabdan-8(17)-13-dien-15-óico) e de 3,7ppm, da fração majoritária de tanino catéquico de massa molecular 864Da. Obtiveram-se as substâncias através de fracionamentos cromatográficos sucessivos, identificadas por ressonância magnética nuclear de hidrogênio e espectrometria de massas. As larvas que atingiram estado letárgico foram coletadas e dissecadas e seus tubos digestórios fixados, desidratados, emblocados e polimerizados. Cortes ultrafinos foram feitos e contrastados com acetato de uranila 3% e citrato de chumbo, posteriormente, levados ao microscópio eletrônico. RESULTADOS: As principais alterações ultraestruturais provocadas pelos diterpeno e tanino sobre larvas de Aedes aegypti foram vacuolização citoplasmática, desorganização e degeneração celular, mudança estrutural dos microvilos e deslocamento das células da lâmina basal. CONCLUSÕES: O diterpeno e a fração rica em taninos catéquicos provocaram a morte das larvas de Aedes aegypti através da destruição celular no intestino médio.
Mechanisms of the larvicidal effect of diflubenzuron on Aedes aegypti larvae, as evidenced by ultrastructural changesDengue is a viral disease transmitted by Aedes aegypti in over 100 countries in the intertropical band in the world and the main forms of control are antivectorial actions. In this paper, ultrastructural changes caused by diflubenzuron (DFB) are presented. The experiments were performed with 3 rd instar larvae of Ae. aegypti with DFB at concentrations of 0.1 and 1 µg/mL. After 24h of exposure, larvae were collected, fixed, dehydrated, embedded, cut, contrasted with uranyl acetate and 3% lead citrate and examined under the electron microscope. Ultrastructural alterations caused by the diflubenzuron inhibitory activity were observed in the cuticle and in the midgut of Ae. aegypti larvae treated with 0.1 and 1 µg/mL, for 24 hours. The alterations observed by scanning microscopy included higher number of bristles, which were thinner and longer than the controls and exhibited a coiling pattern in the intersegmental grooves. Transmission electron microscopy analysis revealed that the epicuticle almost came entirely off the new epicuticle and had no points of reinforcements commonly found in the control. The midgut cells of the larvae treated with diflubenzuron presented a spongy skeleton and in the ultrathin sections, the cells showed intense vacuolation and damage, however with secretion vesicles present and mitochondria preserved. These studies indicate that the diflubenzuron can also act in the midgut cells, being a promising product for use in Ae. aegypti control. This study showed that the DFB interferes with the process of ecdysis, prevents the release of the old cuticle that accumulates in the intersegmental spaces and strangle the segmental portions in a successive and cumulative process, blocked the changes, causing the death of the larva. It also acts destroying the midgut cells.
As políticas públicas e demais ações de promoção da saúde, relativas às DST (doenças sexualmente transmissíveis), devem atentar para quais informações ou o que as pessoas conhecem sobre DST. Dados do Boletim Epidemiológico-AIDS/DST do Ministério da saúde apresentam um total de 608.230 casos de AIDS notificados de 1980 a junho de 2011 no Brasil. E apesar das instituições de ensino serem um excelente campo de conscientização sobre as DSTs, no cotidiano das escolas há uma grande dificuldade dos professores em abordar assuntos relacionados com a sexualidade não só com adolescentes, mas também com jovens e adultos membros da EJA (Educação de jovens e adultos). O presente artigo apresenta resultados do trabalho desenvolvido com alunos da EJA pelo Projeto Sexualidade-Mitos e Verdades da UFG, que aborda temas que envolvem a sexualidade e DSTs. Foi possível verificar que a maior parte dos alunos sabe identificar pelo menos um sintoma que costuma ser relacionado às DSTs, além disso, as mulheres apresentaram maior conhecimento sobre as formas de prevenção da AIDS do que os homens, mas também foi possível verificar a falta de conhecimento dos alunos sobre informações básicas referentes a prevenção de DSTs. Estes resultados expõem a necessidade da atuação de projetos e políticas públicas relacionadas à sexualidade não só entre adolescentes, mas também entre jovens e adultos, tendo como campo aberto e carente, os estudantes da EJA. Analysis of knowledge of students and the education of young adults (EJA) about diseases sexually transmitted ABSTRACT: Public politics and other actions to promote health, relating to STD (sexually transmitted diseases), should pay attention to what information or what people know about STDs. Data from the Epidemiological Bulletin-AIDS / STD Ministry of Health show a total of 608,230 AIDS cases were reported from 1980 to June 2011 in Brazil. And while educational institutions are excellent field awareness about STDs, in daily school there is a great difficulty for teachers in teach subjects related to sexuality not only to teens but also to young and adults members of the EYA (education of youth and adults). This article presents results of the work with the EYA students by Sexuality Project at UFG, which subjects involving sexuality and STDs. We noticed that most students can identify at least one symptom that is often related to STDs, moreover, women had greater knowledge about ways of preventing AIDS than men, but it was also possible to verify the absence of students' knowledge of basic information regarding the prevention of STDs. These results expose the need of projects and public politics related to sexuality not only among adolescents but also among youth and adults, it having the open field and needy, the students of the EYA.
O principal mecanismo de controle do dengue tem sido por meio da eliminação do Aedes aegypti, seu transmissor mais competente. Este mecanismo consiste, principalmente, na destruição dos criadouros potenciais, na educação sanitária e no uso de inseticidas sintéticos. Estes inseticidas apresentam eficiência, mas são altamente tóxicos aos vertebrados e ao meio ambiente. Com o aparecimento da resistência dos mosquitos a essas substâncias, abre-se uma oportunidade aos inseticidas de origem botânica. Neste estudo, apresentam-se os mecanismos da atividade larvicida do ácido 3-β-acetoxylabdan-8(17)-13-dien-15-oico, extraído da planta medicinal Copaifera reticulata Ducke (Leguminosae), sobre larvas de terceiro estádio de Ae. aegypti. As larvas foram submetidas a uma solução do ácido a 9ppm, por até 24 horas. Aquelas que atingiram estado letárgico foram coletadas e fixadas em paraformaldeído a 4%, em tampão cacodilato de sódio 0.1M pH 7.2, incluídas em resina, e as lâminas, coradas pela técnica de hematoxilina-eosina e analisadas por microscopia de luz. O ácido causou a morte das larvas pela destruição celular do intestino médio, por meio de vacuolização citoplasmática, hipertrofia celular e nuclear, degeneração do bordo em escova, formação vesicular apical com liberação de conteúdo citoplasmático, estratificação do epitélio e dobras na matriz peritrófica.
