Modelo do estudo: Estudo quantitativo, de natureza descritiva, com delineamento transversal, realizado no ano de 2013. Objetivo: Realizar uma avaliação das interações medicamentosas potenciais (IMP), em prescrições médicas contendo antimicrobianos de uso restrito, de pacientes internados em um hospital no interior do estado da Bahia. Metodologia: A coleta de dados foi realizada no período de janeiro a maio, empregando a análise documental, com acompanhamento prospectivo de prescrições contidas nos prontuários, para possibilitar a detecção das IMP. A ocorrência e a classificação das IMP foram verificadas através da base de dados MICROMEDEX® DrugReax System. Resultados: 50 pacientes participaram do estudo, sendo analisadas 797 prescrições médicas. Dentre o grupo dos antimicrobianos de uso restrito, a associação piperacilina + tazobactam foi a mais prescrita, estando presente em 33,0% das prescrições. Das prescrições utilizadas para análise das IMP, 357 (46,5%) apresentaram pelo menos uma IMP, sendo que destas, 7 (5,8%) envolviam antimicrobianos de uso restrito. Foram encontrados como fármacos envolvidos nas IMP: amitripitilina + linezolida (46,0%), piperacilina + warfarina (26,0%), entre outros. Do total de interações encontradas, 467 (31,9%) foram classificadas como sendo de gravidade moderada e documentação boa, 808 (53,5%) como interações de mecanismo de ação farmacocinético e 743 (50,8%) como interações de início de reação demorado. Conclusão: Foi realizada uma avaliação das IMP, em prescrições médicas contendo antimicrobianos de uso restrito, e a partir delas uma tabela de auxílio foi construída, criando subsídio para o uso racional de alguns antimicrobianos potencialmente envolvidos em interações medicamentosas.
The enzyme glucanase from Moniliophthora perniciosa was produced in liquid medium and purified from the culture supernatant. A multivariate statistical approach (Response Surface Methodology -RSM) was employed to evaluate the effect of variables, including inducer (yeast extract) and fermentation time, on secreted glucanase activities M. perniciosa detected in the culture medium. The crude enzyme present in the supernatant was purified in two steps: precipitation with ammonium sulfate (70%) and gel filtration chromatography on Sephacryl S-200. The best inducer and fermentation time for glucanase activities were 5.9 g L −1 and 13 days, respectively. The results revealed three different isoforms (GLUI, GLUII and GLUIII) with purification factors of 4.33, 1.86 and 3.03, respectively. The partially purified enzymatic extract showed an optimum pH of 5.0 and an optimum temperature of 40 • C. The enzymatic activity increased in the presence of KCl at all concentrations studied. The glucanase activity was highest in the presence of 0.2 M NaCl. The enzyme showed high thermal stability, losing only 10.20% of its specific activity after 40 minutes of incubation at 90 • C. A purified enzyme with relatively good thermostability that is stable at low pH might be used in future industrial applications.

Lytic enzymes such as chitinases, proteases and β-1,3-glucanases are usually necessary. Mathematical models have been increasingly used to help explain responses of biochemical reactions. This technique is often used for optimization and/or verification of the influence of medium components and cultivation parameters for enzyme production (FLEURI; SATO, 2008).The production and optimization of the parameters that affect the enzymatic synthesis should always be investigated because the optimal conditions vary among different organisms and different enzymes (BRAVO et al., 2000). In many cases, the interaction of parameters that influence fermentation processes can be evaluated with a reduced number of tests using an experimental design (THÉODORE; PANDA, 1995). Several experimental design models could be used to reduce the number of experiments under different conditions. Among them are the Plackett-Burman, Doehlert, Central Composite, and Box-Behnken (LI et al., 2007).The choice of Doehlert design is justified by a number of advantages such as (1) its spherical experimental domain with an uniformity in space filling, (2) its ability to explore the whole of the domain, and (3) its potential for sequentially where the experiments can be reused when the boundaries have not been well chosen at first (BENSALAH et al., 2010). Doehlert designs are easily applied to optimized variables and offer advantages over the Central Composite (CCD) and Box-Behnken designs, used in response surface analysis. Objetivo: Descrever a prevalência de interações medicamentosas potenciais (IMP) em hipertensos e as suas possíveis consequências clínicas. Métodos: Estudo transversal, descritivo-analítico, realizado com uma amostra de hipertensos no município de Jequié-BA. Os dados foram coletados entre fevereiro de 2014 e fevereiro de 2015, por meio de amostra aleatória simples, a partir de um questionário constituído de dados sociodemográficos, clínicos e dos medicamentos em uso. Resultados: Dos 235 usuários entrevistados, 74,90% eram do sexo feminino e 57,45% apresentavam pressão arterial ≥121/81mmHg. Foram prescritos 868 medicamentos com média de 3,69±2,01 por paciente. Do total, 61,61% pacientes apresentaram pelo menos uma IMP. Encontrou-se 358 IMP com média de 2,75±2,12 por paciente, sendo que, 12,47% eram graves e 87,43% moderadas. As IMP mais prevalentes foram entre hidroclorotiazida e glibenclamida (11,45%) e hidroclorotiazida e metformina (10,89%), ambas consideradas de gravidade moderada com risco aumentado da hiperglicemia. Constatou-se associação estatística entre a polifarmácia e número de IMP (p0,001), aumentando a chance de IMP em mais de duas vezes (OR=2,39). Já a presença de mais de uma doença crônica não-transmissível aumentou a chance de IMP em quase quatro vezes (OR=4,36). Conclusão: Esta avaliação demonstrou alta frequência de IMP com gravidade moderada em pacientes hipertensos associada a polifarmácia, sendo necessária intervenções em saúde capazes identificar tais problemas e garantir a segurança do paciente.
Amylases have the ability to catalyze the α-1,4 binding of starch by releasing glucose and dextrin, with prominence in several industrial fields. The research sought to characterize the enzyme amylase obtained from the fungus Rhizopus microsporus var. oligosporus, through the Doehlert methodology, evaluating the behavior of the enzyme in the face of variations in pH and temperature. The production of amylase occurred with the use of starch as an inducer, and the characterization was carried out using the response surface methodology, with pH analysis at 5 levels (3.0, 4.0, 5.0, 6.0 and 7.0) and temperature in 3 levels (30, 50 and 70°C). The evaluation of the amylase thermostability occurred at 60, 70 and 80ºC. The application of the experimental model indicated that the amylase obtained from R. microsporus var. oligosporus shows better catalytic development at temperatures between 40ºC and 55ºC and pH between 2.5 and 3.2. The evaluation of thermostability indicated that the increase in temperature negatively influences the catalytic activity of amylase. The experimental model led to the understanding of the conditions favorable to the production of amylases from R. microsporus var. oligosporus.
