The immune responses to Haemonchus contortus were compared in studies in resistant Barbados Black Belly (BBB) and susceptible INRA 401 (INRA) breeds of lambs. The cytokine patterns indicated a Th2-biased response in both breeds. A more persistent and elevated Th2 cytokine mRNA transcription and blood eosinophilia were noted in the BBB lambs. However, at days 4 and 30 post-infection, abomasal recruitment of eosinophils and mast cells were similar between the two breeds. Following primary infections, the BBB demonstrated a substantially lower faecal egg count compared to the INRA lambs. Furthermore, worm counts at 4 and 30 days post-infection, and adult female worm size and in utero egg counts 30 days after the first infection were significantly lower in the BBB than in the INRA breed. In the INRA breed, re-infection caused a significant reduction in most parasitological parameters compared with those observed after the primary infection. A similar response was not observed in the BBB sheep. In conclusion, while the major driving force in the response to H. contortus infection is a Th2-biased immunity in which the BBB showed its maximal performance during the primary infection, the INRA breed performed better after re-infection compared to its response to first exposure.

BackgroundTrypanosomosis, via causing anaemia, emaciation, production loss and death, is arguably the most important constraint to livestock development in Sub-Saharan countries, including Ethiopia and its impact in Baro-Akobo and Gojeb river basins (endemic areas for tsetse flies) is unknown. This study was carried out from November 2011 to April 2012 to assess farmers’ perception on the presence, impact, management and the need of intervention programs of bovine trypanosomosis and tsetse fly in selected districts located in Baro-Akobo and Gojeb river basins, Southwestern Ethiopia. A standardized questionnaire survey was employed to collect the relevant information from the farmers.ResultsThe result of this study showed that 94.1% of the respondents considered bovine trypanosomosis as an economically important cattle disease which accounted for 64.6% of the total annual deaths in the year 2011/2012. Estimated mean annual financial loss via mortality due to trypanosomosis was reported to be 3501 Ethiopian Birr (US$200)/household. The reported trypanosomosis suggestive signs were consistent with published reports and farmers strongly associated the occurrence of the disease with biting flies (particularly, tsetse fly). Respondents also explained that the seasonality of the disease and its vectors, i.e. May and June are peak risk months of the year. Chemotherapy was reported the major method to combating the problem, mean frequency of treatment being 5.7 times per animal per year. Because of the economic burden of the disease, farmers expressed their strong interest and support for the establishment of intervention program in their area.ConclusionThe study revealed that livestock keepers are familiar with bovine trypanosomosis and its vectors as well as its impacts. Thus, trypanosomosis and tsetse control strategies should be integrated with the local communities’ participation to minimize the impacts of the disease and its vectors in the area.

BackgroundTrypanocidal drugs have been used to control African animal trypanosomosis for several decades. In Ethiopia, these drugs are available from both authorized (legal) and unauthorized (illegal) sources but documentation on utilization practices and quality of circulating products is scanty. This study looked at the practices of trypanocidal drug utilization by farmers and the integrity of active ingredient in trypanocides sold in Gurage zone, south western Ethiopia. The surveys were based on a structured questionnaire and drug quality determination of commonly used brands originating from European and Asian companies and sold at both authorized and unauthorized markets. One hundred farmers were interviewed and 50 drug samples were collected in 2013 (Diminazene aceturate = 33 and Isometamidium chloride = 17; 25 from authorized and 25 from unauthorized sources). Samples were tested at the OIE-certified Veterinary Drug Control Laboratory (LACOMEV) in Dakar, Senegal, by using galenic standards and high performance liquid chromatography.ResultsTrypanosomosis was found to be a major threat according to all interviewed livestock keepers in the study area. Diminazene aceturate and isometamidium chloride were preferred by 79% and 21% of the respondents respectively, and 85% of them indicated that an animal receives more than six treatments per year. About 60% of these treatments were reported to be administered by untrained farmers. Trypanocidal drug sources included both unauthorized outlets (56%) and authorized government and private sources (44%). A wide availability and usage of substandard quality drugs was revealed. Twenty eight percent of trypanocidal drugs tested failed to comply with quality requirements. There was no significant difference in the frequency of non-compliance between diminazene-based and isometamidium chloride products (P = 0.87) irrespective of the marketing channel (official and unofficial). However, higher rates of non-compliant trypanocides were detected for drugs originating from Asia than from Europe (P = 0.029).ConclusionThe findings revealed the presence of risk factors for the development of drug resistance, i.e. wide distribution of poor quality drugs as well as substandard administration practices. Cryptosporidium, a protozoan parasite in the phylum Apicomplexa, is the etiological agent of cryptosporidiosis, an intestinal infection characterized by profuse watery diarrhea. Over 30 species of Cryptosporidium are recognized, some host specific whereas others infect a broader host range. Cryptosporidium hominis and Cryptosporidium parvum are the species most commonly associated with human infection; C. hominis is largely associated only with human infections, but C. parvum is also associated with infection in animals, especially young ruminants. In some regions, cryptosporidiosis is a serious veterinary problem, particularly for calves, and lambs. Many outbreaks of human cryptosporidiosis have been associated with zoonotic transmission following contact with infected animals. In Africa, where cryptosporidiosis is a major contributor to pediatric morbidity and mortality, evidence suggests transmission is principally anthroponotic. Given the frequent close contact between humans and animals in Africa, the apparent predominance of human-to-human transmission is both interesting and puzzling. In this article, after a brief "text book" introduction to the parasite, we consider in separate sections the different aspects of relevance to Cryptosporidium transmission in African countries, describing different aspects of the various species and subtypes in human and animal infections, considering livestock management practices in different African countries, and looking for any characteristic "hot spots" where zoonotic transmission has apparently occurred. Studies where transmission networks have been investigated are particularly relevant. Finally, in a separate section, we try to gather these different strands of evidence together in order to assess the reasons behind the apparent predominance of anthroponotic transmission in Africa. Reviewing the available evidence provides an opportunity to rethink transmission pathways, not only in Africa but also elsewhere, and also to pose questions. Does the predominance of human-to-human transmission in Africa reflect a relative absence of zoonotic C. parvum in African livestock? Are Africans less susceptible to zoonotic Robertson et al. Zoonotic Cryptosporidium Transmission in Africa Cryptosporidium infection, perhaps resulting from early immunostimulation by C. hominis or due to inherent genetic traits? Is the African environment-in all its variety-simply more detrimental to oocyst survival? Will the so-called hypertransmissible subtypes, currently relatively rare in Africa, be introduced from Europe or elsewhere, and, if so, will they fade out or establish and spread? Our intention with this manuscript is not only to summarize and consolidate diverse data, thereby providing an overview of data gaps, but also to provide food for thought regarding transmission of a parasite that continues to have a considerable impact on both human and animal health.

BackgroundToxoplasmosis is a major public health concern in many countries of the world. A cross-sectional and follow up experimental study designs were used for seroepidemiological and bioassay studies, respectively from November 2012 to April 2013. The objectives were to estimate the seroprevalence of T. gondii infection, to assess risk factors and to isolate the parasite from camels in the Fentale district, Ethiopia. A direct agglutination test (DAT) and indirect enzyme linked immunosorbent assay (ELISA) kits were used to test camel sera. Hearts and tongues (each 25 g) from 31 seropositive camels were bioassayed in mice. Associations between seroprevalence and potential risk factors (collected using a questionnaire survey) were analyzed using logistic regression.ResultsAn overall T. gondii prevalence of 49.62% (220/455) by DAT and 40.49% (179/451) by indirect ELISA test were detected. Herd level seroprevalence of 96.77% (30/31) (95% CI: 83.30– 99.92) by DAT was recorded and it was significantly higher in areas where wild felids are present (P = 0.038). Multivariable logistic regression showed that the likelihood of acquiring T. gondii infection was significantly higher in camels in the Ilala pastoral association [PA] (82.26%) (Adjusted Odds ratio [aOR] = 10.8; P < 0.001) than camels in the Galcha PA (31.43%), in camels of ≥ 8 years old (56.52%; aOR = 1.88; P = 0,033) than camels of ≤ 4 years old (34.26%) and in areas where domestic cats are present (aOR = 4.16; P = 0.006). All camel owners were uneducated, handle aborted fetus with bare hands, and drink raw camel milk. DAT and ELISA tests had moderate agreement (Kappa = 0.41). Viable T. gondii were isolated from 16.13% (5/31) of DAT positive camels. One DAT positive but ELISA negative camel sample gave a cyst positive result.ConclusionsT. gondii infection of camels in the study district is widespread. Age, presence of domestic cats and study PA are independent predictors of T. gondii seropositivity. BackgroundAnimal trypanosomosis is one of the major disease problems affecting agricultural productivity in Ethiopia. The impact of the disease is believed to vary with season and agro-ecologies in line with fly vector distribution. A cross-sectional study on bovine trypanosomosis was conducted from November 2015 to June 2016, in seven selected villages of Gidami district, Oromia Regional State, western Ethiopia. A total of 930 blood samples were collected and subjected to parasitological and hematological analysis.ResultThe overall prevalence of bovine trypanosomosis was 14.1%. The seasonal prevalence shows 9.06% in early dry and 18.4% in early rainy seasons. Three trypanosome species, Trypanosoma congolense, Trypanosoma vivax and Trypanosoma brucei were identified in the examined animals. T. congolense followed by T. vivax were the predominant species (respectively 59.0 and 35.9% in early dry season and 62.0 and 22.8% in early rainy season). The prevalence of T. vivax remained similar in both early dry and early rainy seasons in both lowland and midland agroecologies whereas T. congolense was more dominant in the lowland area in both seasons compared to mid land study sites. The disease was more prevalent in lowland (23.9%) compared to midland (11.1%) during early rainy season (P < 0.001) whereas no significant difference was observed between the two agroecologies during early dry season (P = 0.165). Packed cell volume (PCV) was much lower in parasitemic animals than in aparasitemic cattle whereas the mean PCV value for parasitemic animals (20.36%; 95% CI 19.56 to 21.16) in early dry season was similar to values in early rainy season (20.46%, 95% CI 18.84 to 21.08%). A similar situation was noticed for animals in both low land and mid land study sites.ConclusionOverall, the detection of trypanosomes in blood was significantly affected by agro-ecology, season and body condition of the animals. Special emphasis should be given to integrated trypanosomosis management in early rainy months where fly population is believed to start increasing.
