Delivery of microbial products into the mammalian cell cytosol by bacterial secretion systems is a strong stimulus for triggering pro-inflammatory host responses. Here we show that Salmonella enterica serovar Typhi (S. Typhi), the causative agent of typhoid fever, tightly regulates expression of the invasion-associated type III secretion system (T3SS-1) and thus fails to activate these innate immune signaling pathways. The S. Typhi regulatory protein TviA rapidly repressed T3SS-1 expression, thereby preventing RAC1-dependent, RIP2-dependent activation of NF-κB in epithelial cells. Heterologous expression of TviA in S. enterica serovar Typhimurium (S. Typhimurium) suppressed T3SS-1-dependent inflammatory responses generated early after infection in animal models of gastroenteritis. These results suggest that S. Typhi reduces intestinal inflammation by limiting the induction of pathogen-induced processes through regulation of virulence gene expression.
The purpose of this study was to investigate histological changes in dairy cows' hooves with or without injuries from naturally acquired laminitis. Cull cows with no clinical signs of hoof abnormalities (G1, n=9) and those with macroscopic lesions associated with laminitis without (G2, n=23) or with lameness (G3, n=7) were used in the study. After slaughter, samples of dermo-epidermal junctions of sole, axial and dorsal regions of the hoof were obtained and histologically processed using HE and PAS staining. Congestion, hemorrhage and inflammatory infiltrate in the dermis of sole, axial and dorsal regions were blindly and semiquantitatively evaluated by the same researcher. Inflammatory infiltrate was evaluated in the dermal laminae of axial and dorsal regions. The morphology of epidermal cells and the presence of irregularities in three regions of the basement membrane (BM) length were examined using PAS staining. Scores of lesions in different regions of the hoof in the same group and in different groups for each region of the hoof were compared using non-parametric analyses. Inflammatory infiltrate in the dermis of all regions of the hoof was detected in all groups with no significant statistical difference. Cows with no clinical signs of hoof abnormalities secondary to laminitis (G1) have inflammation scores and epidermal cell changes similar to those of groups with laminitis injuries, suggesting the existence of a prodromal phase for this disease in bovines. BM had irregularities with a variable intensity along its length, however, with no difference among groups. The pattern of BM irregularities found has not been reported so far and does not resemble the BM collapse described in horses and cattle with induced acute laminitis. Is it concluded that even in the absence of macroscopic hoof signs associated to laminitis, dairy cows have histological injuries compatible with inflammation of the dermo-epidermal junction as in affected animals. Basement membrane of cows with or without laminitis associated lesions had irregularities with an irregular distribution along its length which need to be further studied.
RESUMOAvaliou-se o perfil eletrocardiográfico de 60 eqüinos da raça Mangalarga Marchador, clinicamente sadios, com idades variando de um a 28 anos, sendo 39 fêmeas e 21 machos. A freqüência cardíaca tendeu a diminuir com a idade e a amplitude da onda P aumentou ao longo do crescimento do animal. Os parâmetros de duração de QRS e amplitude de S foram maiores em machos que em fêmeas.Palavras-chave: eqüino, eletrocardiograma, Mangalarga Marchador ABSTRACT For the electrocardiographic evaluation of Mangalarga Marchador breed horses, 60 electrocardiograms were recorded from clinically healthy equines aging from one to 28-year-old, being 39 females and 21 males. The heart rate tended to decrease with age and the amplitude of P wave increased with the growth of the animals. The duration of the QRS complex and the amplitude of S wave were greater in males than in females.
Phenylbutazone (PBZ) is widely used in equine medicine, and its side effects on the gastrointestinal tract are well known. The inhibition of prostaglandins and the oxidative stress induced by nonsteroidal anti-inflammatory drugs (NSAIDs) are described as mechanisms of gastric mucosal injury in humans. In horses, only the secondary effect of changes in cyclooxygenases is related to gastric mucosal injury. The objective of this study was to evaluate the effect of PBZ on certain antioxidative/oxidative parameters of the gastric mucosa. The concentrations of antioxidants and oxidants (superoxide dismutase, SOD; catalase, CAT; nitric oxide, NO; total glutathione, GSH; myeloperoxidase, MPO; and malondialdehyde, MDA), PGE2 levels, and the ulcerative lesions score were assessed. The results demonstrated decreased levels of antioxidant variables, increased levels of oxidant variables, and alterations in the prostaglandin E2 (PGE2 ), myeloperoxidase (MPO), and glutathione (GSH) levels. In conclusion, PBZ induces oxidative stress in the gastric glandular mucosa of horses by changing the antioxidant-oxidant balance of this surface, which might be regarded as another mechanism of injury in the horse stomach.
O equilíbrio ácido-base foi estudado em eqüinos com compactação experimental do cólon maior após o uso de solução isotônica poliônica enteral, sene mais hidratação intravenosa e hidratação intravenosa. As amostras sangüíneas foram colhidas de 20 animais, quatro grupos (E8, RL, SE e C) de cinco animais cada. Os animais dos grupos tratados eram portadores de compactação no cólon maior induzida experimentalmente. Eqüinos no grupo E8 receberam solução isotônica poliônica enteral (8mL kg-1 h-1 48h-1); no grupo SE, sene foram administrados na dose de 20mg kg-1, duas doses de 24/24h, mais Ringer lactato intravenoso, 10mL kg-1h(-1)12h-1, durante dois dias; sendo que o grupo RL recebeu Ringer lactato intravenoso (16mL kg-1h(-1)12h-1) durante dois dias. O grupo C, controle, não foi tratado. Entre os tratamentos testados, a fluidoterapia intravenosa com solução de Ringer lactato (RL) foi o tratamento mais eficiente para a correção das variáveis hemogasométricas em eqüinos com compactação do cólon maior. A solução isotônica poliônica enteral (E8) e sene associada com fluidoterapia intravenosa (SE) foram eficientes, porém em menor grau.
RESUMO.-O objetivo deste trabalho foi estudar o efeito do plasma rico em plaquetas (PRP) no tratamento da tendinite induzida no tendão do músculo flexor digital superficial (TFDS) de eqüinos mediante avaliação ultrasonográfica. Para isso foram utilizados seis eqüinos hígidos machos castrados, com idade de 8-15 anos (x=12 anos). A tendinite do TFDS foi provocada em ambos os membros torácicos, mediante a administração intratendínea de 2,5mg de colagenase (2,5mg.μL -1 ), sendo esse procedimento considerado o início da fase experimental. Doze dias após a indução da tendinite, os animais foram submetidos a dois tratamentos: sendo que na lesão efetuada no TFDS direito (grupo tratado, GT), foram administrados 2,5mL de PRP ativado com cloreto de cálcio a 0,0125mol.L -1 , contendo concentrações variando de 320.000 a 500.000 plaquetas.μL -1 ; na tendinite do TFDS esquerdo (grupo controle, GC), foram injetados 2,5mL de solução salina a 0,9%. Após cinco dias, os The objective of the study was to evaluate the effect of platelet-rich plasma (PRP) in the treatment of induced tendinitis in the superficial digital flexor tendon (SDFT) of horses through ultrasonographic evaluation. Thus, six 8 to15-year-old healthy gelding horses ( x =12 years) were used. Tendinitis of the SDFT was provoked in both forelimbs via intratendineous administration of 2.5mg of collagenase (2.5mg.mL ), what was considered as beginning of the experimental phase. Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L -1 , at concentrations from 320,000 to 500,000 platelets.μL -1 , were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon crosssectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. The result revealed reduction (P<0.05) in the L-CSA and in the degree of lesion echogenicity in function of time, but with difference (P<0.05) between groups only for L-CSA, with smaller values in TG. The treatment with PRP promotes greater reduction in the area of the lesion measured by ultrasound.
