Abstract. Tumor-infiltrating lymphocytes (TILs) are a heterogeneous cell family which plays an important role in tumorassociated immune response. Of these, T regulatory (Treg) cells have also been shown to inhibit anti-tumor response. We aimed to evaluate the expression of T regulatory cell markers (CD4, CD25, CTLA-4 and FoxP3) in samples of oral cavity squamous cell carcinoma (OCSCC) and lip SCC (LSCC) by immunohistochemistry. The relationship of Treg markers with survival data and the proliferative index were also evaluated. We observed similar numbers of CD4-, CD25-and FoxP3+ cells in OCSCC and LSCC. On the other hand, numbers of CTLA-4 + cells were significantly lower in OCSCC than in LSCC. OCSCC samples with high numbers of CD4 exhibited a high proliferative index, while samples with high CTLA-4 counts demonstrated a low tumoral proliferative index. A log-rank test showed that patients with OCSCC that presented high counts of CD4 showed a significantly decreased survival compared with patients with low cell counts. In contrast, high CD25 + cell counts were associated with increased survival. Our results suggest an association of CD4 with poor prognosis, while CD25 expression is related with favorable prognosis. These findings result from the heterogeneity of TIL subsets that display an antagonistic role in tumor immune cell response.
The present study investigated the effect of birthweight on testicular development and spermatogenesis in boars. Twenty-four pairs of littermate boars were selected: one piglet with the highest birthweight (HW) and the other with the lowest birthweight (LW) within the litter. Two subsets of 12 pairs of male littermates from each birthweight group were obtained after selection: one subset was orchiectomised at 8 days and the other at 8 months of age. HW boars had higher body and testicular weights at both ages (P<0.05). Testosterone concentrations and the relative expression of 17α-hydroxylase in the testis were similar between birthweight groups. Birthweight affected somatic and germ cell numbers in the neonatal testis, which were higher in HW boars (P<0.05). Moreover, a significant reduction in the number of pachytene spermatocytes and round spermatids was observed in LW boars (P<0.05) at 8 months of age, which caused a decrease in the total number of elongated spermatids and daily sperm production (P<0.05). Hence, HW boars have the potential to produce more spermatozoa and consequently more semen doses per ejaculate, and would be very valuable to an industry that relies on AI.
AbstractBackground: The diagnosis of wide variety of lesions occurring in the oral cavity is critical to the practice of dentistry. It is common for dentists to find oral lesions of doubtful diagnosis. In these situations, the dentist should consider the indication of biopsy as a complement or even conclusive final diagnosis of these oral lesions. The evaluation of the agreement between the clinical and histopathological diagnosis of oral lesions allows the accuracy and ability analysis of the professional or his/her dentistry academic level to perform clinical diagnosis correctly. Objective: This study aimed to evaluate the correlation between clinical and histopathological diagnosis of oral lesions diagnosed in a university dental clinic. Method: This is an analytical, retrospective study conducted with patients biopsied and who had pathologic reports issued between February 2010 and July 2012. We evaluate the Reynolds-averaged Navier-Stokes equations available as commercial computational fluid dynamics code for the simulation of a neutral atmospheric boundary layer and attempt to define a proper numerical simulation procedure. Four turbulence models, including two-equation and Reynolds stress models, were evaluated together with two near-wall models. Mesh and map digitization sensitivity tests were also performed. The simulations were compared to experimental field data from the Askervein Hill in Scotland. The results show that the simulations performed with ANSYS CFX 12.1 on a proper mesh and topological map with a Reynolds stress turbulence model provided the best wind-speed predictions when compared to the experimental results.
Genetic diversity in sugar apple (Annona squamosa L.) by using RAPD markers PalavrasGenetic diversity in a collection of 64 sugar apple accessions collected from different municipalities in northern Minas Gerais was assessed by RAPD analysis. Using 20 selected RAPD primers 167 fragments were generated, of which 48 were polymorphic (28.7%) producing an average of 2.4 polymorphic fragments per primer. Low percentage of polymorphism (< 29%) was observed by using the set of primers indicating low level of genetic variation among the 64 accessions evaluated. Genetic relationships were estimated using Jaccard's coefficient of similarity. Accessions from different municipalities clustered together indicating no correlation between molecular grouping and geographical origin. The dendrogram revealed five clusters. The first cluster grouped C19 and G29 accessions collected from the municipalities of Verdelândia and Monte Azul, respectively. The second cluster grouped G16 and B11 accessions collected from the municipalities of Monte Azul and Coração de Jesus, respectively. The remaining accessions were grouped in three clusters, with 8, 15 and 37 accessions, respectively. In summary, RAPD showed a low percentage of polymorphism in the germplasm collection.
