A novel LDL-associated phospholipase A2 (LDL-PLA2) has been purified to homogeneity from human LDL obtained from plasma apheresis. This enzyme has activity toward both oxidized phosphatidylcholine and platelet activating factor (PAF). A simple purification procedure involving detergent solubilization and affinity and ion exchange chromatography has been devised. Vmax and Km for the purified enzyme are 170 micromol.min-1.mg-1 and 12 micromol/L, respectively. Extensive peptide sequence from LDL-PLA2 facilitated identification of an expressed sequence tag partial cDNA. This has led to cloning and expression of active protein in baculovirus. A lipase motif is also evident from sequence information, indicating that the enzyme is serine dependent. Inhibition by diethyl p-nitrophenyl phosphate and 3,4-dichloroisocoumarin and insensitivity to EDTA, Ca2+, and sulfhydryl reagents confirm that the enzyme is indeed a serine-dependent hydrolase. The protein is extensively glycosylated, and the glycosylation site has been identified. Antibodies to this LDL-PLA2 have been raised and used to show that this enzyme is responsible for >95% of the phospholipase activity associated with LDL. Inhibition of LDL-PLA2 before oxidation of LDL reduces both lysophosphatidylcholine content and monocyte chemoattractant ability of the resulting oxidized LDL. Lysophosphatidylcholine production and monocyte chemoattractant ability can be restored by addition of physiological quantities of pure LDL-PLA2.
In target-directed aiming, afferent information is used to adjust limb trajectories during movement execution (i.e. online) and to enhance the programming of subsequent trials (i.e. offline). The objective of the present study was to determine the influence of state anxiety on both online and offline afferent information processing for the first time. Participants practiced either a directional aiming task (Experiment 1) or an amplitude aiming task (Experiment 2) without anxiety before being transferred to a high anxiety condition. In both experiments, results revealed that anxiety resulted in a decrement in performance. Furthermore, use of afferent information to adjust movement trajectories online was disrupted when movements were performed with anxiety, whereas there were no differences in the offline processing of afferent information between the low anxiety and high anxiety conditions.
Increases in reaction time (RT) as a function of response complexity have been shown to differ between simple and choice RT tasks. Of interest in the present study was whether the influence of response complexity on RT depends on the extent to which movements are programmed in advance of movement initiation versus during execution (i.e., online). The task consisted of manual aiming movements to one or two targets (one- vs. two-element responses) under simple and choice RT conditions. The probe RT technique was employed to assess attention demands during RT and movement execution. Simple RT was greater for the two- than for the single-target responses but choice RT was not influenced by the number of elements. In both RT tasks, reaction times to the probe increased as a function of number of elements when the probe occurred during movement execution. The presence of the probe also caused an increase in aiming errors in the simple but not choice RT task. These findings indicated that online programming was occurring in both RT tasks. In the simple RT task, increased executive control mediated the integration between response elements through the utilization of visual feedback to facilitate the implementation of the second element.
The relative age effect (RAE) describes an overrepresentation of players born early (Q1) in a selection year and is highly prevalent within youth sport pathways. However, a dearth of research has investigated the RAE at the "super-elite" level. The present research assessed the presence of RAE in super-elite performers. Study 1 investigated RAEs in the world's best international Test cricketers (N = 262) over a 20-year period according to a robust and stringent "super-elite" criteria. Results revealed the RAE (Q1) when all disciplines were combined. Upon closer examination, this effect was also observed for the batting and spin bowling disciplines, whereas no RAE was found for the pace bowling discipline. Study 2 investigated RAEs in super-elite rugby union players (N = 691) over a 20 year period. Results revealed the RAE for backs (Q1) and a reversal of the traditional RAE (Q4) for forwards, and when all rugby union positions were combined. These findings provide new evidence of RAEs at the super-elite level and present both inter and intra sports differences. Potential explanations for these findings are explored, owing to the survival and evolution of the fittest concepts, and the implications for future research and applied practice are presented.
