This article presents the results of pathomorphological research on the spleen and mesenteric lymph nodes of 23 dead cats aged from 3 months to 7 years, which in their lifetimes (according to anamnesis, clinical signs, laboratory-instrumental methods of examination and VetExpert FCoV Ab express test) had been diagnosed with infectious peritonitis. All the animals were domestic. Blood was drawn from the subcutaneous vein of the forelimb of the diseased cats. We determined ESR, morphological parameters of blood and content of hemoglobin. For histological and histochemical examinations, we selected samples of spleen and mesenteric lymph nodes, which were fixated in 10% aqueous solution of neutral formalin, Carnoy’s and Bouin’s solutions. The prepared histological sections were stained using haematoxylin and eosin, Van Gieson’s stain, methyl green-pironin stain (Brashe), PAS-reaction (McManus), alcian blue and Congo red. Hematological parameters during 3 weeks of clinical progression of the disease among the cats demonstrated a decrease in the hemoglobin content and in the number of erythrocytes and leukocytes. Possible decrease in the number of lymphocytes indicated the development of an immune-deficiency state. Also, during the development of disease, the animals had possible increase in ESR, which indicated the development of an inflammatory process in the organism and decrease in the number of thrombocytes, which conditioned development of disseminated intravascular coagulation. The anatomical pathology autopsy showed that in most animals the spleen was diminished in size, the surface of the organ was tuberous, the capsule was wrinkled and mat, the edges were sharpened. The histostructural change was accompanied by a sharp depletion of the lymph nodes and reduction in the number of micro- and macrophages, which indicated the reduction of white pulp, rapid inhibition of the activity of T- and B-lymphocytes, plasmacytic and macrophage reaction and manifested in development of immune-deficient condition of the organism. In this process, the reticular carcass of the lymph nodes saturated with PAS-positive and eosinophilic masses was clearly manifested, which indicated formation of fibrinoid. In the spleen of 5 individuals, during staining using Congo red, we found deposition of amyloid masses both in the intima of the blood vessels and along the reticulary fibers of the lymph nodes. In the cytoplasm of macrophages, we found pyroninophilic formations. In two cases, we observed blood accumulation of red pulp and bleeding following the reduction of white pulp, and in one case fibrinogenous perisplenitis. In the mesenteric lymph nodes of most of the cats which had suffered from infectious peritonitis, we determined that edema, exposure of the reticular soft skeleton (stroma) of adrenal and paracortical zones, dilation of the border and central sinuses and thrombosis of vessels were followed by steep decrease in the number of T- and B-lymphocytes, plasma cells, micro- and macrophages, which indicated the development of atrophic processes of lympoid tissue and immune-suppression. In three cases, in mesenteric lymph nodes of cats, we determined development of sinus histiocytosis. The changes determined in the spleen and lymph nodes of the cats which had suffered from FIP indicate immune-suppressed condition and steep decrease in the functional ability of the organs and organism in general.
In the development of effective methods of prevention and treatment of poultry today, importance is given to probiotics based on microbial cultures, in particular, lactobacilli, bifidobacteria, yeast, etc. In contrast to antibiotics, the use of probiotics stimulates the immune response of animals, restores the microflora of the gastrointestinal tract, and ensures its optimal ratio. At the same time, livestock products remain safe for the consumer. The purpose of our work was to conduct comparative clinical trials of the efficiency of the probiotic feed additive Probion-forte in terms of productivity, blood indices, and histological structure of immune organs of chicken broilers during fattening. The clinical studies were carried out with broiler chickens of “Cobb-500” cross at the age of 2 days, which were formed into four groups (300 units in each one). In order to determine the efficiency of application, probiotic feed additives were given to chickens in different concentrations during the growing period, together with the main diet. The first group (1st) was given probiotic Probion-forte in the dose of 1 g/kg of feed, the second group (2nd) – Probion-forte in a dose of 0.5 g/kg, the third (3rd) one – a probiotic-analog “Bio plus 2B” in a dose of 0.4 g/kg; the forth (4th) group was a control group – chickens received an essential diet, without adding any feed additives. The compound feed was provided according to norms recommended for the cross “Cobb-500”, considering age. On the 15th, 30th, and 43rd day of the test, 20 units were selected for hematological, pathomorphological and microbiological tests. The productivity of birds of all studied groups was evaluated by the average daily gains, safety, feed conversion, and slaughter output. The stabilized with EDTA chicken blood was used for morphological studies, and blood serum – for biochemical and immunological studies. Comparative clinical studies have shown that adding to the main diet of broiler chickens probiotic feed additive Probion-forte at a dose of 0.5 and 1.0 g/kg throughout the growing period increases the safety and improves feed digestibility, and increases the bird’s European Efficiency Index. Based on the obtained hematological, biochemical, and immunological parameters of blood, it can be argued about the activating effect on the body of broiler chickens probiotic feed additives, namely the intensification of reanimation of free amino acids, which increases the content of the studied metabolites of lipid metabolism, which chickens use as energy and plastic material. Stimulating cellular and humoral protection, nonspecific resistance is established, confirmed by a high level of lysozyme activity in blood serum and significantly higher content of T- and B-lymphocytes, NK-cells, and γ-globulins. The macro- and microscopic structure of the studied immune organs is preserved in all groups of broiler chickens. Compared with the control group, morphometric examination in the immune organs of broiler chickens of the first and second groups revealed an increase in the area and density of lymphoid elements, an increase in the number of plasmablasts and plasma cells, much higher structural and functional capacity of cells, which was reflected at the ultrastructural level. The most significant difference was found in the 1st group of chickens fed with the feed additive Probion-forte at a dose of 1.0 g/kg of feed.
The article presents results of histological, histochemical investigations of duodenal mucosa of broilers, which were fed 36 days in a row with were fed 36 days in a row with fodder mixed of probiotic supplements «Probion forte» in doses of 100 g/kg. At 36 day of the experiment 10 chickens from each group were taken out from the experiment, was performed autopsy and selected models of duodenum, which are fixed in 10% of solution neutral formalin, Karnua liquid, followed by pouring into paraffin. It was produced histological sections, were stained with hematoxylin-eosin, by Stidmen, McManus. Prolonged feeding of poultry with probiotic supplements «Probion forte» in excessive dose of 100 g/kg has caused structural and functional changes in the mucosa of the duodenum, expressed the formation of ugly bifurcated broad villi and reflected a decrease in productive indexes of poultry.
The article is dedicated to the study of the effect of a class of microorganisms and substances of microbial and other origin that are used in feeding pigs to achieve maximum growth in animal mass, as well as therapeutic purposes. Study of the effect of biologically active additives on the state of the pig's digestive system. The article considers the rational nutrition that increase the productivity and reproductive capacity of animals, and also prevent the emergence of profound disturbances of all metabolic processes which leads to a decrease in resistance to productivity, a clinically pronounced disease of adult animals and young animals. The authors pay attention to the morphometric parameters, ultrastructure and content of nucleic acids in the wall of the duodenum of pigs by forage feeding with the addition of probiotic fodder additive «Probion-forte» in dose of 1 g/kg of fodder. Research were conducted on 28 day pigs, breed «Large White». It was formed two groups of piglets per 30 heads; piglets were fed with standard mixed fodders; piglets from the first group were received standard mixed fodder with the addition of probiotic fodder additive «Probion-forte» in dose 1 g/kg of fodder for 42 days. After the slaughter of piglets, pieces of duodenum for histological, histochemical and ultrastructural studies were selected. Statistical significance of differences was determined by Student's t test , assuming 5% estimate error. It was shown that feeding with forage within 42 days from the addition of probiotic fodder additive «Probion-forte» at a dose of 1 g/kg, villus height is increase, crypt depth and a number of plasma cells in the lamina propria of mucosa of the duodenum, which helps the digestive process and increase the area of nutrient absorption in the intestines. The number of plasma cells are increased in the lamina propria of mucosa and testify immunomodulatory effect of fodder additives. Ultra structural alteration of microvilli and changes in the nuclei of duodenal enterocytes of piglets of the first group indicates a more pronounced acfunctional activity of enterocytes and thereby increases the activity of parietal digestion in the intestine. As a conclusion, the task by definition performed of the effective dose of probiotic in feed for pig it can be considered completed and we can state the probiotics can be incorporated, as a alternative to antibiotics and increase the weight gain of animals and also to increase the level of immune resistance of the organism.
The article presents the results of histological studies of the frontal area of the cerebral cortexб taken from 10 corpses of cats aged from 3 months to 5.5 years, in which infectious peritonitis (based on anamnesis, clinical signs, morphological and biochemical blood analysis , ultrasound diagnostics, Rivalt test and express diagnosis) was diagnosed during their life. A pathoanatomical study was conducted; samples of the frontal area of the cerebral cortex of cats, which were fixed in a 12% aqueous solution of neutral formalin, a solution of Carnoua and 96° ethyl alcohol, were selected. The histocuts were made, which were stained with hematoxylin and eosin, thionine for Nisslem. In order to detect the astrocytic glory, a fraction of the cerebral cortex was fixed by the Golgi-Clatzo method with a fresh mixture of chloral hydrate-formalin-bichromate and produced histocuts on a freezing microtome. For histological examination of the cerebral cortex of cats, changes in the non-inflammatory character were observed, which were characterized by disorders of hemomycrocycle circulation, edema and degenerative processes of the neuroglial complex and neurons. Violations of the structural organization of endothelial cells, basal membranes of capillaries and venules should be considered as an important indicator of functional changes in tone, permeability of the microcirculatory channel, which contributed to the development of perivascular and pericellular edema, which in its turn led to regressive changes of glial elements; acute swelling of neurons with the formation of microcavities in nuclei; a sharp decrease in the content of the chromatophilic substance in pyramidal and stellate cells; the development of gidropic dystrophy in star cells and creatures; rise of shadow cells. Circulatory disorders caused the violation of the trophic transport systems, which caused hypoxia, accumulation of acidic metabolism products, and formed the basis for the pathogenesis of the development of dyscirculatory dystrophy of neuroglial cells and neurons, i.e. neurotrophic disorders of the cerebral cortex of cats for FIR.
