Background:
Envenomation caused by Bothrops alternatus is common in Southern Brazil.
Acute Kidney Injury occurs after Bothrops snakebite and more information is necessaryrequired to
understand its mechanism.
Objective:
The objective was to evaluate the effect of Bothrops alternatus venom (BaV) on renal cells
and rat isolated kidney function.
Methods:
Wistar rats (n = 6, weighing 260-320 g) were perfused with a Krebs-Henseleit solution containing
6 g 100 mL-1 of bovine serum albumin. After 30 minutes, the kidneys were perfused with BaV
to a final concentration of 1 and 3 μgmL-1; and subsequently were evaluated for Perfusion Pressure
(PP), Renal Vascular Resistance (RVR), Urinary Flow (UF), Glomerular Filtration Rate (GFR), and
percentage of electrolyte tubular transport. Renal histological analysis, cytokine release, oxidative
stress and cytotoxicity in renal proximal tubular cells were assessed.
Results:
BaV reduced PP, RVR, GFR, UF, total and proximal sodium transport (%TNa+), and chloride
(%TCl-) in the isolated kidney perfusion model. Histological analysis of perfused kidneys disclosed the
presence of proteinaceous material in the glomeruli and renal tubules, vacuolar tubular epithelial cell
degeneration, Bowman's capsule degeneration, swelling of glomerular epithelial cells, glomerular atrophy
and degeneration, and the presence of intratubular protein. Cytokine release (TNF-α, IL-1β, IL-10)
and oxidative stress were increased in the kidneys. The viability of LLC-MK2 cells (IC50: 221.3
μg/mL) was decreased by BaV and necrosis was involved in cell death.
Conclusion:
These findings indicate that BaV modifies functional parameters in an isolated perfused
kidney model and has cytotoxic effects on renal lineage cells.
Neste estudo propõe-se avaliar os efeitos toxicológicos, pressóricos e renais da onco-A em modelos animais in vivo e ex vivo. A Oncocalyxona A (onco-A) isolada de Auxemma oncocalyx foi estudada nas doses 1, 3, e 10 µg/mL. Utilizou-se camundongos Swiss fêmeas (20-25g) para o teste de toxicidade oral aguda. Os dados foram comparados por teste t de Student, Two-Way ANOVA e pós-teste de Bonferroni (p<0,05). Foi estimado uma DL50 ˃ 2000 mg/kg, os tecidos pulmonar, renal e cardíaco demonstraram alterações histológicas. Sobre os níveis pressóricos de ratos, a onco-A ocasionou redução significativa da pressão arterial média, alterou os níveis de creatinina e, observou-se alterações histológicas nos tecidos cardíaco e renal. Em perfusão de rim isolado a onco-A causou aumento na pressão de perfusão, fluxo urinário e resistência vascular renal. O ritmo de filtração glomerular encontrou-se reduzido nas concentrações de 1 µg/mL e 10 µg/mL e aumentado para a dose de 3 µg/mL. As fotomicrografias do tecido renal após a perfusão demonstraram alterações histológicas significativas induzidas pela onco-A. Pode-se inferir que, embora a onco-A tenha apresentado baixa letalidade em todos os protocolos experimentais realizados, mostrou evidências de toxicidade pulmonar, renal e cardíaca.
Nos eritrócitos deficientes de glicose 6 fosfato desidrogenase (G6PD), a diminuição da redução de NADP em NADPH, leva a um baixo potencial redutor que interfere na capacidade metabólica oxidativa do organismo, ficando vulnerável a hemólise, com oxidação da membrana do glóbulo, podendo levar a crise hemolíticas. As manifestações da deficiência de G6PD são desencadeadas por fatores ambientais como drogas, fava, infecções. As manifestações clínicas consistem de crise hemolítica e icterícia neonatal. Estudos indicam que no Brasil a prevalência de G6PD está em torno de 1 a 10%, sendo os maiores índices em populações com ancestralidade africana. O objetivo foi a pesquisa da deficiência da G6PD em recém-nascidos numa maternidade pública na cidade de Campina Grande. Trata-se de um estudo transversal, onde foram coletados sangue do cordão umbilical de 147 recém-nascidos. O diagnóstico da deficiência da G6PD será realizado através do teste da redução da metahemoglobina (teste de Brewer). A cidade de Campina Grande, bem como todo o Compartimento da Borborema não apresentam nenhum dado relativo a prevalência da deficiência da G6PD. O Ministério da Saúde fortalece através do SUS a coleta de informações relativas a esta doença na perspectiva de implantação de um programa de atenção à saúde dos doentes detectados, promovendo melhoras na sua qualidade de vida.
