Microsatellites or SSRs (simple sequence repeats) are ubiquitous short tandem duplications occurring in eukaryotic organisms. These sequences are among the best marker technologies applied in plant genetics and breeding. The abundant genomic, BAC, and EST sequences available in databases allow the survey regarding presence and location of SSR loci. Additional information concerning primer sequences is also the target of plant geneticists and breeders. In this paper, we describe a utility that integrates SSR searches, frequency of occurrence of motifs and arrangements, primer design, and PCR simulation against other databases. This simulation allows the performance of global alignments and identity and homology searches between different amplified sequences, that is, amplicons. In order to validate the tool functions, SSR discovery searches were performed in a database containing 28 469 nonredundant rice cDNA sequences.
Molecular and morphological data analyses matrices are very informative tools for the estimation of genetic distances. We used AFLP markers, morphological traits and combined analysis to estimate the genetic distances between wheat genotypes and ascertain any associations between the two techneques. Nineteen wheat (Triticum aestivum L.) genotypes were analyzed using amplified fragment length polymorphism (AFLP) markers and field evaluated for two years. The matrices obtained by morphological and molecular marker data analyses revealed a significant but moderate correlation (r = 0.47), indicating that such techniques sample distinct genome regions. The combined analysis was found to be highly correlated with AFLP (r = 0.97) and moderately correlated with morphological (r = 0.59) markers. A possible explanation for such results is a bias caused by the much higher number of AFLP (229) than morphological (17) markers. Thus, it is evident that the combined analysis is not efficient when a very dissimilar number of markers are used in each isolated technique. Therefore, to obtain a better knowledge of the degree of divergence among genotypes it is necessary to consider each analysis separately.
Resumo -O objetivo deste trabalho foi determinar o padrão de desenvolvimento de afilhos em genótipos de trigo contrastantes para esse caráter, em diferentes densidades de semeadura, bem como seus efeitos sobre os componentes do rendimento de grãos. O experimento foi conduzido no Município de Capão do Leão, RS, em 2006. Dez cultivares de trigo, selecionadas com base na capacidade de afilhamento, foram utilizadas em delineamento de parcelas divididas, com a parcela composta pelo fator cultivar, e as subparcelas pelas densidades de semeadura, com 50, 200, 350, 500 e 650 sementes aptas por metro quadrado. Observou-se que a senescência de afilhos esteve diretamente relacionada ao potencial de afilhamento dos genótipos. Os genótipos com elevada capacidade de afilhamento apresentaram efeito mais pronunciado da senescência, com o aumento da densidade de semeadura. O melhor ajuste dos componentes do rendimento foi obtido por meio da adequada densidade de semeadura, que deve ser realizada com base no potencial de afilhamento dos genótipos. Além disto, o efeito compensatório de genótipos com reduzido potencial de afilhamento é resultante de maior massa de grãos por espiga, em detrimento do número de espigas por unidade de área.Termos para indexação: Triticum aestivum, estádio de desenvolvimento, potencial de afilhamento, senescência, sobrevivência de afilhos. Tiller development and yield components in wheat genotypes under different seeding densitiesAbstract -The objective of this work was to determine the developmental pattern of tillers in wheat genotypes showing contrasting number of tillers. The genotypes were tested under different seeding densities, in order to evaluate their effect on grain yield components. The experiment was performed in Capão do Leão County, Rio Grande do Sul State, Brazil, in 2006. Ten wheat cultivars, selected by their tillering ability, were tested in a split plot design, where plots were formed by the genotype (cultivar), and subplots by different seeding densities: 50, 200, 350, 500 and 650 seeds per square meter. Results indicated that tiller senescence is directly correlated to tillering potential of the genotypes, and genotypes with higher tillering ability presented higher senescence values, when subjected to higher seeding densities. The best adjustment of yield components is obtained from the adequate seeding density, which can be achieved by matching the density and tillering potential. Also, higher yield in genotypes with reduced tillering potential is achieved by increasing grain mass per ear, to compensate for their lower number of ears per area unit.
Multicolinearity consequence on path analysis in canola RESUMO
Rice is a model genome for cereal research, providing important information about genome structure and evolution. Retrotransposons are common components of grass genomes, showing activity at transcription, translation and integration levels. Their abundance and ability to transpose make them good potential markers. In this study, we used 2 multilocus PCR-based techniques that detect retrotransposon integration events in the genome: IRAP (inter-retrotransposon amplified polymorphism) and REMAP (retrotransposon-microsatellite amplified polymorphism). Markers derived from Tos17, a copia-like endogenous retrotransposon of rice, were used to identify genetic similarity among 51 rice cultivars (Oryza sativa L.). Genetic similarity analysis was performed by means of the Dice coefficient, and dendrograms were developed by using the average linkage distance method. A cophenetic correlation coefficient was also calculated. The clustering techniques revealed a good adjustment between matrices, with correlation coefficients of 0.74 and 0.80, or lower (0.21) but still significant, between IRAP and REMAP-based techniques. Consistent clusters were found for Japanese genotypes, while a subgroup clustered the irrigated Brazilian genotypes.

Foram utilizados sete genótipos, obtidos através de avanços de gerações de 1999 a 2001, utilizando duas épocas de semeadura por ano, na estação de verão e inverno, sob condições de campo e telado, na Universidade Federal de Pelotas, RS. O experimento foi desenvolvido em delineamento de blocos casualizados com cinco repetições. A análise de trilha permite identificar o componente primário MMG, como o de maior potencial para seleção de constituições genéticas superiores para rendimento de grãos.Palavras-chave: produtividade, enchimento de sementes, variabilidade. ABSTRACT PATH ANALYSIS FOR THE YIELD COMPONENTS OF SEEDS IN WHEATThe purpose of this work was to estimate the effect of the ear number per plant, number of seeds per ear, seed weight, size of the ear and spikiest number per ear in relation to the grain yield through its correlation coefficients and its decomposition by the path analysis. The genotypes were obtained through the advances generation in the years from 1999 to 2001, using two sowing dates per year, in the warm and cold season, in the field and in green-house conditions, at the Federal University of Pelotas, RS. The experiment was conducted as a randomized complete block design with five replications. The characters spikiest number for spike and hectoliter weight were identified by the path analysis, as being the one of high potentiality for selection of superior genetic constitutions for seed yield seeds.
ResumoA toxicidade do alumínio é um dos principais fatores limitantes do desenvolvimento das plantas em solos ácidos. Pelo fato da utilização de corretivos da acidez do solo não ser a estratégia mais viável em muitas situações com solos ácidos (por razões técnicas e econômicas), o desenvolvimento de genótipos tolerantes ao Al tem sido o caminho mais focado, assim a investigação dos mecanismos de tolerância bem como as bases genéticas da tolerância ao Al têm merecido atenção especial pela pesquisa científica. Nos últimos anos, foi gerado um significativo progresso no entendimento das bases dos mecanismos de tolerância ao Al, assim como no desenvolvimento de cultivares mais adaptados as condições de solos ácidos. Os mecanismos de tolerância ao Al conhecidos se resumem basicamente em duas classes: os que agem no sentido de expulsar o Al depois de absorvido ou de impedir sua entrada pela raiz e os mecanismos de desintoxicação, complexando o Al em organelas específicas da planta, principalmente nos vacúolos. Em inúmeras espécies, mecanismos fisiológicos tem sido reportados como responsáveis pela ativação de ácidos orgânicos (principalmente citrato e malato) que atuam como agentes quelantes do Al, porém muitos processos ainda não são bem entendidos e esclarecidos. Atualmente, se começa a entender melhor um segundo mecanismo de tolerância ao Al que envolve a desintoxicação interna do Al através da complexação por ácidos orgânicos e o seqüestro destes complexos pelos vacúolos. Outros mecanismos potenciais são alvo de especulações e discussões. The use of liming for correcting soil pH is not viable for some of acid soil areas (technique or economic reasons), making the development of Al tolerant genotypes the best alternative.
