ABSTRAKBiosensor merupakan suatu perangkat sensor yang menggabungkan senyawa biologi dengan suatu transduser. Salah satu jenis biosensor yang menggunakan teknik pendeteksian perubahan massa adalah QCM (Quartz Crystal Microbalance). QCM merupakan suatu sensor yang bekerja dengan prinsip pergeseran frekuensi pada kristal kuarsa akibat adanya deposisi massa di permukaan kristal. QCM dapat dipergunakan untuk mendeteksi reaksi antar molekul, sehingga QCM dapat berfungsi sebagai biosensor yang dapat dipergunakan untuk diagnosis suatu penyakit. Dalam pengembangan QCM untuk immunosensor terdapat berbagai permasalahan, salah satunya adalah konstruksi sel. Konstruksi sel dapat dipergunakan sebagai tempat reaksi antara biomolekul. Konstruksi sel terbuat dari bahan teflon putih. Untuk menjaga agar sensor QCM tidak mengalami tekanan fisik yang bisa mengakibatkan pecah karena tekanan dari teflon maka digunakan o-ring silikon sebagai penyekat. Hasil penelitian ini menunjukkan bahwa konstruksi sel yang dibuat dapat digunakan untuk dipergunakan sebagai media pengamatan imobilisasi Bovine Serum Albumin (BSA) di atas permukaan sensor.Kata kunci: Biosensor, QCM, konstruksi sel, immunosensor. ABSTRACTThe biosensor is a sensor device that combines biological compounds with a transducer. One type of biosensor that uses mass change detection techniques are QCM (Quartz Crystal Microbalance). QCM is a sensor that works with the principle of quartz crystal frequency shift due to mass deposition on the surface of the crystal. QCM can be used to detect the reaction between the molecules, so that the QCM can serve as biosensors that can be used for the diagnosis of a disease. In the development of QCM immunosensor for there are many problems, one of which is the construction of the cell. Construction cells can be used as a reaction between biomolecules. Construction of cells made of white Teflon. To keep QCM sensors are not experiencing physical stress that can lead to rupture due to pressure from the Teflon o-ring is used as an insulating silicon. The results of this study indicate that the construction of the cells created can be used as a medium for immobilization of Bovine Serum Albumin observations (BSA) on the surface of the sensor.
Perkembangan peradaban manusia tidak pernah lepas dari proses pemanfaatan energi. Saat ini hampir seluruh aspek kehidupan manusia memiliki ketergantungan terhadap energi. Secara umum sumber energi dikategorikan menjadi dua, yaitu tak terbarukan dan terbarukan. Pengenalan akan pentingnya energi terbarukan sebagai pengganti energi fosil perlu ditanamkan pada anak sejak dini. Akan tetapi, umumnya guru-guru di sekolah dasar maupun menengah mengalami kesulitan dalam menyampaikan materi tentang energi terbarukan secara mudah dan menarik. Permasalahan inilah yang diangkat oleh Pusat Studi Energi Terbarukan (PSET) Universitas Surabaya (Ubaya) untuk melakukan program pengabdian pada masyarakat dalam bentuk sosialisasi dan pengenalan energi terbarukan pada siswa sekolah mitra. Solusi permasalahan dilakukan dengan memberikan program pengenalan energi terbarukan kepada mitra pengabdian. Metode kegiatan pengabdian adalah mengunjungi/mengundang siswa sekolah mitra untuk diberi paparan interaktif tentang energi terbarukan khususnya sel surya oleh narasumber. Setelah itu siswa diminta membuat modul peraga sel surya sederhana dan mempraktikkannya secara langsung untuk mengamati energi yang dihasilkan oleh sel surya. Pemahaman siswa dievaluasi melalui survei di akhir kegiatan. Hasil survei menunjukkan bahwa program ini dapat membantu meningkatkan pemahaman siswa akan energi terbarukan khususnya tentang sel surya. Pengetahuan dasar yang diberikan pada siswa sekolah mitra diharapkan dapat menjadi bekal pemahaman terkait program energi yang berwawasan lingkungan.
Phalaenopsis sp. is the most-produced orchid species in Indonesia. Compared to conventional breeding, mutation induction by using mutagens, such as Ultraviolet Light-C (λ = 254 nm) (UV254) and Ethyl Methane Sulfonate (EMS), could probably result in new superior orchid variants. This research aims to get some mutants with phenotypes that have visual differences in the Phalaenopsis spp. hybrids wild type. There were 4 durations of UV254 irradiation: 5’ on, 85’ off; 10’ on, 80’ off (1 day and 7 days for each treatment); 4 concentrations of EMS used in this research: 0.05%; 0.06%; 0.07%; 0.08% for 6 hours of immersion; selected UV254 irradiation (5’ on, 85’ off (7 days)) combined with these concentrations. UV254 irradiation treatment (5’ on, 85’ off (1 day and 7 days); 10’ on, 80’ off (7 days)) resulted in some mutants with leaf phenotypes that were visually different from the wild type; 0.05% EMS treatment resulted in PLBs mutant with a visually larger size than the wild type; 0.08% EMS treatment and combination treatments (for EMS 0.05% and 0.08% for each treatment) resulted in non-growing albino PLBs. Hence, mutation induction using UV254 and EMS in this research produced several most likely mutants having visual differences that may be more desirable than the wild type.
Porang (Amorphophallus muelleri Blume.) usually used to functional food raw materials, caused its high glucomant content. Limitation of conventional propagation both vegetative and generative affected to decreasing of porang production to fulfil market demand. To solve the problem, plant tissue culture technique was chosen to rapid propagate bulbils of porang. So, the aim of this research to determine the combination and concentration of Plant growth Regulators (PGRs), such as 6-Benzylaminopurine (BAP), Naphthaleneacetic Acid (NAA), and Thidiazuron (TDZ) for quality of callus, also growth of callus and shoots for rapid propagation of porang, cause PGRs are one of factors that affect the success of in vitro propagation. Experimental design of this research was completely randomized designed with statistical analysis using analysis of variance (Anova) one way. Difference combination and concentration of cytokinin and auxin as manipulation variables, which several parameters growth of callus, shoots, and roots was observed to indicate the rapid organogenic of porang. The optimal shoot induction medium was Murashige and Skoog (MS) media with 5.0 mg.L-1 BAP and 0.2 mg.L-1 NAA on which number of shoots and shoot height reached 15±1.8 and (7.2±1.86) cm. After shoot formation stage, the proper media for rooting stage was determined, which was Murashige and Skoog (MS) media with 1.0 mg.L-1 NAA resulted the rooting rate reached 100% and roots appear earlier. Optimal combination and concentration of PGRs on micropropagation of Amorphophallus muelleri Blume. from bulbils explant should be known to increase the production of porang, which can fulfil the market demand.
Utilization of Solanum lycopersicum L. and its varieties contribute to plant biotechnology to study the effect of genes and flower transition events. This study aims to optimize media and light intensity for in vitro flowering of a cherry tomato variety. Induction of flowering was carried out using seven types of media combined with 3 light intensities, followed by feeding by adding liquid medium MS+PBZ 1 mg/L. A positive response is shown at the light intensity of 2,600 lux; there are 5 explants that have flower buds. The percentage of flower buds for each treatment combination was 50% from two replications. The first shoots occurred at 142 days after planting (DAP) on explants grown on media containing 0.5 mg/L BA, 0.75 mg/L PBZ, with feeding treatment. It can also be assumed that the emergence of flower buds occurred not because of the PGR induction treatment, but because at that time the explants had entered the generative phase, considering that the plantlets had been growing for a long time. Although the effects of BA and PBZ have not been confirmed, the use of 2,600 lux light appears to be the best condition for cherry tomato in vitro flowering induction.
Buah tomat merupakan salah satu buah yang sering dijumpai di Indonesia. Permintaan tomat juga terus meningkat dari tahun ke tahun di Indonesia. Keterbatasan lahan menjadi kendala dalam memenuhi permintaan tersebut. Oleh karena itu, tomat dapat dikembangkan melalui media kultur jaringan untuk menghasilkan buah yang sehat dan cepat. Salah satu faktor yang menentukan keberhasilan dalam budidaya tomat melalui media kultur jaringan adalah intensitas cahaya yang digunakan. Metode multiple linear regression diimplementasikan untuk dapat memperkirakan intensitas cahaya yang tepat. Fitur yang akan menjadi prediktor dalam penelitian ini adalah jenis media tanam dan panjang kecambah yang diharapkan. Berdasarkan evaluasi yang telah dilakukan, nilai mean absolute error (MAE) yang dihasilkan yaitu 0,87 dengan rasio data training dan testing adalah 90:10. Hal ini menandakan bahwa sistem cukup akurat dalam melakukan prediksi.
Telah dilakukan studi konsumsi energi di kantin Keluwih Universitas Surabaya (UBAYA) sebagai langkah awal untuk menentukan desain pemanfaatan energi alternatif yang tepat guna di kantin tersebut. Kajian diawali dengan melakukan survei konsumsi energi di kantin Keluwih untuk mengetahui kebutuhan energi di kantin. Konsumsi energi ini diukur dari energi yang dipergunakan untuk penerangan (lampu), pendingin ruangan (AC), dan peralatan elektronik lainnya (televisi, blender, rice cooker, kompor listrik, dll.) serta bahan bakar yang dipakai untuk memasak. Data diperoleh dari kuesioner yang diberikan kepada pengelola kantin serta penyewa stan di kantin Keluwih. Dari data yang diperoleh dapat diketahui peta konsumsi energi di Kantin Keluwih dan berdasarkan survei yang telah dilakukan dapat disimpulkan bahwa konsumsi energi di Kantin Keluwih didominasi oleh energi listrik yang jumlahnya berkisar 600 kWh/hari.
