A new chromone, 2-(2-hydroxy-2-phenylethyl)chromone (), was isolated together with ten known phenylethyl chromones from MeOH extracts of agarwood (). The selected compounds were evaluated in an antiproliferative assay against five human tumor cell lines, including a multidrug-resistant cell line. They were also tested for antitumor promoting activity, as mediated by 12--tetradecanoylphorbol-13-acetate-induced activation of the Epstein-Barr virus early antigen in Raji cells. Among all compounds, 4',7-dimethyoxy-6-hydroxychromone () displayed broad spectrum antiproliferative activity against all tumor cell lines tested with IC values of 25-38 µM, while was selectively inhibitory against multidrug-resistant cells. All tested compounds suppressed tumor promotion at noncytotoxic concentrations. 4',6-Dihydroxyphenylethylchromone () exhibited the most potent effect with an IC value of 319 mol ratio relative to 12--tetradecanoylphorbol-13-acetate. This study is the first to report the antitumor promoting activity of 2-(2-phenylethyl)chromone derivatives, as well as the selective antiproliferative activity of against a multidrug-resistant tumor cell line.
Ficus elastica leaf is one of the Ficus species assumed to be rich in polyphenols so that it can act as an antioxidant. The research aimed to determine the antioxidant activity and IC50 value of methanol and water extracts of rubber leaf. In this research, the extraction was conducted by maceration using methanol and obtained the yield value of 7,03%, while the infundation using water obtained the yield value of 2,82%. The antioxidant activity test was conducted by DPPH free radical scavenging (1,1-diphenyl-2-phycrilhydrazyl) using a quercetin comparator. The qualitative tests were conducted by TLC method using dicloromethane:methanol eluent with a ratio of 99:1 for methanol extracts, and a ratio of 95:5 for water extracts, where all extracts of both methanol and water positively container antioxidants characterized by the formation of yellow spots with a purple background on TLC plates. The results of the quantitative test shower that the methanol extract had medium antioxidant activity with an IC50 value of 78,39 µg/mL, and water extract had an inactive antioxidant activity with an IC50 value of 319,11 µg/mL. While quercetin was classified as a very strong antioxidant with an IC50 value of 7,62 µg/mL.
Biancaea sappan (BS) is traditionally used to treat anticonvulsants, anti-inflammatory, antiproliferative, anticoagulant, antiviral, immunostimulant, antioxidant, and antimicrobial treatments. Flavonoids are found in Secang; flavonoids are secondary metabolites that have antioxidant activity. This study aims to identify the flavonoids using TLC and determination of flavonoids content in BS leaves. Initially, The methanol extract of BS was obtained by maceration with ethanol. The qualitative analysis of flavonoid was using TLC and visualization by sprayed with AlCl3. The determination of total flavonoid content is conducted based on the AlCl3 method with total flavonoids expressed in QE (Quercetin equivalent) at the maximum wavelength of 431 nm. The research results showed that BS leaves contain flavonoids and the total flavonoid content of BS leaf extract is 1.0318 mg QE / g extract.Keywords: Caesalpinia sappan; Flavonoid content, Spectrophotometric
