The effects of DNA therapy are reflected in gene expression modulation; therefore, the genes identified as differentially expressed could be considered as transcriptional biomarkers of DNAhsp65 immunotherapy against TB. The data have important implications for achieving a better understanding of gene-based therapies.

Petromica ciocalyptoides AND Topsentia ophiraphidites, FROM THE ASCIDIAN Didemnum ligulum AND FROM THE OCTOCORAL Carijoa riisei. The investigation of extracts from six species of marine invertebrates yielded one new and several known natural products. Isoptilocaulin from the sponge Monanchora aff. arbuscula displayed antimicrobial activity at 1.3 μg/mL against an oxacillin-resistant strain of Staphylococcus aureus. Five inactive known dibromotyrosine derivatives, 2 -6, were isolated from a new species of marine sponge, Aplysina sp. The sponges Petromica ciocalyptoides and Topsentia ophiraphidites yielded the known halistanol sulfate A (7) as an inhibitor of the antileishmanial target adenosine phosphoribosyl transferase. The ascidian Didemnum ligulum yielded asterubin (10) and the new N,N-dimethyl-O-methylethanolamine (11). The octocoral Carijoa riisei yielded the known 18-acetoxypregna-1,4,20-trien-3-one (12), which displayed cytotoxic activity against the cancer cell lines SF295, MDA-MB435, HCT8 and HL60.Keywords: marine sponge; ascidian; octocoral. INTRODUÇÃONos últimos anos, investigações químicas e farmacológicas de organismos marinhos têm contribuído significativamente para a descoberta de novas substâncias químicas potencialmente bioativas para fins terapêuticos 1 . Embora o litoral brasileiro seja o segundo mais extenso depois da Austrália, o desenvolvimento da química de produtos naturais de organismos marinhos no Brasil foi negligenciado por muitos anos, porque o principal foco da química de produtos naturais brasileira foi direcionado ao estudo de plantas. Desta maneira, a fauna marinha brasileira ainda é pouco explorada na pesquisa por produtos naturais biologicamente ativos, estruturalmente inéditos ou taxonomicamente relevantes 2 .No presente trabalho relata-se a investigação química dos extratos brutos bioativos de quatro esponjas, uma ascídia e um octocoral. Descreve-se o isolamento de um alcalóide guanidínico a partir da esponja Monanchora aff. arbuscula, de cinco derivados da dibromotirosina, a partir da esponja Aplysina sp., o isolamento do mesmo esterol trissulfatado a partir de ambas esponjas Petromica ciocalyptoides e Topsentia ophiraphidites, o primeiro isolamento da asterubina, a partir de uma ascídia, Didemnum ligulum, da qual também foi isolada a N-N-dimetil-O-metiletanoamina, inédito na literatura, além do 18-acetoxipregna-1,4,20-trien-3-ona a partir do octocoral Carijoa riisei. Também são apresentados os resultados de testes de atividade biológica realizados com estes metabólitos secundários.

The present investigation reports the isolation of aeroplysinin-2, 2-(3,5-dibromo-4-methoxyphenyl)-N,N,N-trimethyletanamonium, 7,9-dibromo-10-hydroxy-8-methoxy-1-oxa-2-azaspiro[4.5]deca-2,6,8-trien-3-carboxylic acid and its methyl ester, 11-oxoaerothionin, aerothionin, 11-keto-12-hydroxyaerothionin, 11-ketofistularin-3 and fistularin-3 from Aplysina fistularis, as well as of furodysinin lactone and 9a,11a-epoxicholest-7-en-3b,5a,6a,10-tetrol-6-acetate from Dysidea sp. Although the extracts of both sponges displayed antituberculosis activity, only 11-ketofistularin-3 isolated from A. fistularis displayed antimycobacterial activity against Mycobacterium tuberculosis H34Rv, with MIC at 16 mg/mL and SI of 40, a result that reinforce that fistularin-3 derivatives are interesting leads for the development of antituberculosis drugs.Keywords: marine sponge; Aplysina fistularis; Dysidea sp.. INTRODUÇÃOA tuberculose é uma doença respiratória prevalente em todo o mundo. Estima-se que um terço da população mundial esteja infectada com o agente etiológico causador da tuberculose, a micobactéria Mycobacterium tuberculosis. Considerada doença erradicada durante os anos 60-70, a tuberculose ressurgiu nos anos 80 associada a linhagens resistentes a antibióticos, de difícil tratamento, que exige 6 meses ininterruptos de quimioterapia. Considera-se que a tuberculose é um dos maiores problemas de saúde pública, especialmente quando associada com doenças que promovem deficiência de imunidade, tais como AIDS.1,2 Neste contexto, a busca por novos agentes terapêuticos para o tratamento da tuberculose tem chamado a atenção.Dois dentre os cinco principais antibióticos utilizados no tratamento de tuberculose são de origem natural. A estreptomicina (1), originalmente isolada das actinobactérias Streptomyces lavendulae e S. griseus, atua sobre M. tuberculosis inibindo a síntese proteica por parte da subunidade ribossomal 30S. Já a rifampicina (2) é um derivado da rifamicina (3), a qual foi originalmente isolada de S. mediterranei. A rifampicina atua sobre M. tuberculosis inibindo a transcrição da subunidade b da RNA polimerase.3 Além da estreptomicina e da rifampicina, dentre os sete antibióticos de segunda linha utilizados no tratamento de tuberculose, quatro são produtos naturais ou derivados de produtos naturais.3 Sendo assim, é evidente a importância de produtos do metabolismo secundário na descoberta e desenvolvimento de medicamentos para o tratamento da tuberculose. Ottonia martiana is a plant popularly known in Brazil by the use for toothache. Ethanolic extract (EE), hexane fraction (HF), dichloromethane fraction (DF) and piperovatine obtained from O. martiana were assayed in vitro and in vivo. The acute toxicity of EE was determined, and LD 50 values of 164.5 and 65.0 mg/ kg by the oral and intraperitoneal routes, respectively, indicated a high toxicity for EE in vivo, explaining its popular use by topical administration only. A local anesthetic-like effect of EE and its fractions was observed in experimental models using pain induction, and such effect involved an analgesic action. The antimycobacterial activity of EE, HF, DF and piperovatine was evaluated against Mycobacterium tuberculosis H37Rv ATCC 27924. EE, HF, DF, and piperovatine showed a potential antimycobacterial effect with MICs of 16.0, 62.0, 62.0 and 8.0 μg/mL, respectively. Piperovatine was more effective than the EE or the other fractions. The selectivity index (SI=IC 50 /MIC) values calculated for EE, HF, DF and piperovatine based on the MICs and the cytotoxicity against J774 macrophages (IC 50 by MTT assay) revealed values of 6.43, 2.34, 1.5 and 9.66, respectively.
