Peptic and/or duodenal ulcers are characterized by diverse acute and chronic ulcerative lesions that commonly arise in any portion of the gastric mucosa that is exposed to the aggressive action of gastric acid. The pathophysiology of peptic ulcers has been attributed to an imbalance between aggressive and protective factors. In Brazil, medicinal plants are commonly used to treat this ailment. A country with great biodiversity, Brazil is considered a rich source of therapeutic products. There have been popular and pharmacological reports on the medicinal relevance of the Brazilian cerrado plant species, including Ananas ananassoides, Celtis iguanaea, Encholirium spectabile, Hymenaea stigonocarpa, Lafoensia pacari, Qualea grandiflora, Qualea parvifora, Mouriri pusa, Solanum lycocarpum, Solanum paniculatum, Serjania erecta, and Vochysia tucanorum, in the treatment of stomach disorders. The aim of the present review was to report on some of the Brazilian cerrado plants that are used in folk medicine because of their gastroprotective potential and to encourage novel studies in the search and preservation of plants with this therapeutic potential.
ABSTRACT:The Celtis iguanaea (Jacq.) Sargent (Cannabaceae) is one of the native species of the Cerrado region of Brazil widely used in folk medicine to treat dyspepsia. The objective of the present study was to evaluate the gastroprotective effect of the Celtis iguanaea (Jacq.) Sargent (HE) hexane leaf extract in the lesion and gastric secretion models.Antiulcerogenic activity of the Celtis iguanaea (HE) hexane leaf extract was observed with the experimental models, such as indomethacin and pyloric ligation-induced gastric ulcers. In order to evaluate the antisecretory activity of this extract, isolated Rana catesbeiana mucosa and pyloric ligation in mice were used. The HE treatment reduced the lesion index of indomethacin and pyloric ligation-induced ulcer. This extract also reduced the gastric acid secretion and total acidity (increasing the gastric pH) in mice. The secretion of H + was reduced in the basal values (15.58 ± 1.99 µEq H + /g/15 min) when isolated Rana catesbeiana mucosa was incubated with HE. Intraduodenal administration of HE reduced the gastric secretion produced by bethanecol or histamine. The antiulcerogenic and antisecretory efficacy of HE in this study suggest anticholinergic and antihistaminergic mechanism or interruption of intracellular events that are linked to acid secretion. RESUMO: Efeito anti-ulcerogênico e antisecretório do extrato hexânico das folhas deCeltis iguanaea (Jacq.) Sargent. Celtis iguanaea (Jacq.) Sargent (Cannabaceae) é uma das espécies nativas do cerrado brasileiro, sendo amplamente utilizada na medicina popular para o tratamento de dispepsia. O objetivo do presente trabalho foi avaliar o efeito gastroprotetor do extrato hexânico das folhas de Celtis iguanaea (Jacq.) Sargent (EH) em modelos de secreção e de lesões gástricas. A atividade antisecretória do extrato hexânico das folhas de Celtis iguanaea (EH) foi avaliada no modelo de mucosa isolada de Rana catesbeiana e pelo modelo de ligadura pilórica em camundongos. A avaliação da atividade anti-ulcerogênica do EH foi determinada no modelo de úlceras gástricas induzidas por indometacina e ligadura pilórica. Quando a mucosa de Rana catesbeiana isolada foi incubada com o EH, a secreção de H + foi reduzida em comparação com o valor basal (15,58 ± 1,99 µEq H + /g/15 min). O tratamento com o EH reduziu o índice de lesões na úlcera induzida por indometacina e ligadura pilórica. O EH também reduziu o volume de secreção, a acidez total, e aumentou o pH gástrico. A administração intraduodenal reduziu a secreção gástrica produzida por betanecol ou histamina. A eficácia anti-ulcerogênica e antisecretória do EH neste estudo, sugere um mecanismo anticolinérgico e antihistaminérgico ou interrupção de eventos intracelulares que estão ligados à secreção ácida. Antiulcerogenic activity of crude ethanolic extract of Celtis iguanaea leaves (CEE) was observed with experimental models such as ethanol, indomethacin, stress and pyloric ligation-induced gastric ulcers. Results obtained from indomethacin-induced ulcer showed the hexane fraction (HF) as the active fraction of CEE. This fraction inhibits the gastric acid secretion, increasing the gastric pH, decreasing the gastric acidity and total gastric contents. Neither the CEE nor the HF alters intestinal motility, thereby excluding a cholinergic antagonist mechanism. Further studies need to be conducted with HF in order to elucidate the active principle and the pharmacological mechanism involved.
Lafoensia pacari A. St.-Hil. can be found from Amapá to Rio Grande do Sul states, and also in Paraguay and Bolivia. It is popularly known as pacari or mangava-brava and is used to promote weight loss, as an anti-thermal or tonic, to treat gastritis, ulcers, scarring, itching, discouragement, and cancer. In the open field tests, the hydroalcoholic extract from L. pacari stem bark (HEP) decreased the number of rearings, number of invaded squares, and increased immobility time compared to control animals. In the pentobarbital-induced sleep time test, HEP decreased latency time to sleep and increased sleeping time. In the rota-rod test, no changes in the studied parameters were observed. In the elevated plus maze, HEP increased the percentage time and percentage entries in the open arms, indicating that this extract exerts an anxiolytic-like activity.Uniterms: Lafoensia pacari A. No teste do campo aberto, o tratamento com o extrato hidro-alcoólico de pacari (HEP) reduziu o número de rearings e o número de quadrados invadidos além de aumentar o tempo de imobilidade dos animais em relação ao controle. No sono induzido por pentobarbital sódico o tratamento com HEP causou redução na latência e aumento na duração do sono. No rota-rod, o tratamento com HEP não alterou os parâmetros observados. No teste de labirinto em cruz elevado, com o tratamento com HEP foi observado aumento do percentual do tempo de permanência e de entradas nos braços abertos, caracterizando uma atividade tipo ansiolítica.
