The objective of this study was to determine the resistance of winter crops of bean (Phaseolus vulgaris) lines with arcelin variants to Zabrotes subfasciatus (Boheman) (Coleoptera: Bruchidae). Two trials, a no-choice test and a free-choice test, were set up under laboratory conditions using genotypes Arc. 1, Arc. 2, Arc. 3 and Arc. 4 (near isogenic lines containing arcelin 1, 2, 3 and 4, respectively), genotypes Arc. 1S, Arc. 3S and Arc. 5S (wild lines containing arcelin 1, 3 and 5, respectively), and genotypes IAPAR MD-808 and Porrillo 70. Plots with 10 g of each line and a proportion of 7 adult pairs/plot were used. Attractiveness, oviposition, emergence and adult weight, mortality, developmental period, and seed weight loss were determined. Nonpreference (attraction and oviposition) was observed only for the wild genotypes Arc. 3S and Arc. 5S. The wild genotypes Arc. 5S and Arc. 1S and the near isogenic lines Arc. 1 and Arc. 2 presented high resistance of the antibiosis type to Z. subfasciatus. Arc. 4 and Arc. 3 presented moderate resistance, probably due to feeding nonpreference.
INFLUENCE OF FRUITING POSITION ON COTTON PRODUCTIONABSTRACT -Cotton production distribution within the plant, seed quality and fruit weight were studied in relation to fruiting position. Data were produced by mapping 360 cotton plants cultivar CNPA 7H which were grown under irrigation in Bom Sucesso, PB. According to the results more than 80% of the total plant yield comes from the botton and middle of the plant and from the first and second fruiting position. Seed quality was affected as the boll position distanced from the first position.
The ability of the stink bug (Nezara viridula) to induce and/or increase production of chemical defenses, i.e., flavonoids, in immature seeds of five genotypes of soybean (BR-16, IAC-100, PI 227687, PI 229358, and PI 274454) was investigated under greenhouse and laboratory conditions. Samples from pods of each genotype damaged by stink bug were analyzed for flavonoid content with high performance liquid chromatography. A dual-choice test was conducted to evaluate the feeding preference of N. viridula comparing BR-16 pods treated with extracts of PI 227687 seeds (with and without stink-bug injury), with water-treated pods. Seeds of PI 227687 damaged by N. viridula presented the highest concentration (352 microg/g) of daidzin (4'-hydroxyisoflavone-7-glucoside). The same trend was observed with genistin (4',5,7-trihydroxyisoflavone-7-glucoside): PI 227687 contained 142.4 microg/g, PI 274454,31,6 microg/g, and PI 229358,38.9 microg/g. Seeds damaged by stink bugs had higher isoflavone contents (daidzin and genistin), compared to controls. However, after being damaged, PI 274454 and PI 229358 produced less genistin than the other genotypes and no differences in concentration between damaged and nondamaged plants of this genotypes were observed. The numbers of observations of the insect feeding and the numbers of stylet sheaths left in water-treated BR-16 pods were greater than in those treated with PI 227687 extracts. The insects fed for longer periods on BR-16 pods treated with extract of PI 227687 without injury compared to those that were treated with extract of PI 227687 previously injured by stink bugs. Extracts of PI 227687 pods (damaged or not) were deterrent to adults of N. viridula, and insect injury increased concentrations of daidzin and genistin in PI 227687 seeds. The deterrence seemed to be more pronounced after pods had suffered stink-bug injury.
The biology of Bemisia tabaci biotype B (Genn.) on bean (Phaseolus vulgaris L.) genotypes containing arcelin in the seeds was evaluated. Also, biochemical screening of seeds and leaves material of these genotypes were carried out in order to verify if traces of arcelin could be found in its leaves. The tests were conducted under greenhouse conditions, in the dry and wet seasons, with the following genotypes: ARC 3s, ARC 5s (wild genotypes containing arcelin in the seeds); ARC 1, ARC 2, ARC 3, ARC 4 (near isogenic lines containing arcelin in the seeds - EMBRAPA) and Porrillo 70, Bolinha, IAPAR MD 808 (commercial genotypes without arcelin). The wild genotypes, ARC 3s and ARC 5s, showed high levels of antibiosis resistance type, mainly for ARC 5s which presented the highest nymphs mortality rates, approximately 90%. Also, the longest development time for nymphs fed on ARC 5s genotype suggest antibiosis and/or feeding nonpreference resistance type. The wild genotype resistance is not related with arcelin presence in the seeds, since no trace of this protein was found in its leaves.
During bean seed storage, yield can be lost due to infestations of Acanthoscelides obtectus Say, the bean weevil. The use of resistant varieties has shown promising results in fighting these insects, reducing infestation levels and eliminating chemical residues from the beans. The expression of resistance to A. obtectus in bean varieties is frequently attributed to the presence of phytohemagglutinins, protease inhibitors and alpha-amylase, and especially to variants of the protein arcelin, which reduce the larval viability of these insects. To evaluate the effect of bean seed storage time on the resistance expression of bean varieties to A. obtectus, tests with seeds of three ages (freshly-harvested, 4-month-old, and 8-monthold) were conducted in the laboratory, using four commercial varieties: Carioca Pitoco, Ipa 6, Porrillo 70, Ônix; four improved varieties containing arcelin protein: Arc.1, Arc.2, Arc. 3, Arc.4; and three wild varieties also containing arcelin protein: Arc.1S, Arc.3S, and Arc. 5S. The Arc.5S, Arc.1S, and Arc.2 varieties expressed high antibiosis levels against the weevil; Arc.1 and Arc3S expressed the same mechanism, but at lower levels. The occurrence of oviposition non-preference was also observed in Arc.5S and Arc.1S. The Arc.3 and Arc. 4 varieties expressed low feeding non-preference levels against A. obtectus. The expression of resistance in arcelin-bearing, wild or improved varieties was affected during the storage of seeds, and was high under some parameters but low in others. The results showed that addition of chemical resistance factors such as protein arcelin via genetic breeding may be beneficial in improving the performance of bean crops.
