Seventy-two cartons of yoghurt were sampled three times at monthly intervals from four different local manufacturers. Total counts were close to 6 x 10 7 cells g -1 of yoghurt. Yeast counts varied from 1 to 2,700 g -1 . There was no evidence of systematic contamination at source but this longitudinal study revealed that ad hoc contamination and improper storage led to the higher yeast counts. Contamination was generally higher in the hotter months but was lower overall than reported from other countries. A total of 577 yeast isolates were identified belonging to ten species. The most abundant yeasts were, in order, Debaryomyces hansenii, Saccharomyces cerevisiae, Mrakia frigida, Hansenula spp., Candida parapsilosis, Debaryomyces castellii and Candida maltosa. The psychrophilic yeast Mrakia frigida is reported for the first time in yoghurts. Low level contamination with Monilia and Penicillium species was found in a few samples. Growth tests suggested that ability to ferment sucrose, growth at 5ºC and in the presence of 300 µg g -1 sorbate preservative, were the three most significant physiological properties to account for these yeasts in yoghurts. The data also suggest that warmer weather and inadequate refrigeration are the principal causes of higher levels of contamination, increased diversity and change in microbial flora.
The effect of gamma irradiation on aflatoxin B1 levels and fungal infection were investigated in peanut samples, Tatu Vermelho cultivar. At a radiation dose of 10 KGy, growth of molds was completely inhibited. Doses of 15, 20, 25 and 30 KGy were sufficient for destruction of aflatoxin B1 by 55-74%. The results suggested that the decontamination of molds by irradiation, before production of aflatoxin B1, is the most acceptable method in the preservation of peanut.
RESUMOA aflatoxina M 1 (AFM 1 ) tem sido detectada em leite de animais alimentados com ração contaminada por aflatoxina B 1 (AFB 1 ), possuindo efeitos tóxicos e carcinogênicos muito próximos. Constitui-se um problema de Saúde Pública, pois sua toxidez é preocupante, quando os indivíduos mais jovens estão entre os maiores consumidores de leite e estes são os mais sensíveis a seus efeitos. O objetivo deste trabalho foi determinar a presença de aflatoxinas em alimentos destinados a bovinos e de AFM 1 em amostras de leite cru e após pasteurização. Trabalhou-se inicialmente com 12 produtores de leite, permanecendo apenas 2 produtores que apresentavam valores significativos de aflatoxina M 1 . A coleta de leite foi realizada 24 h após a coleta do alimento. O método utilizado para a detecção de aflatoxinas no alimento foi a cromatografia em camada delgada. A determinação de AFM 1 em leite foi realizada empregando-se coluna de imunoafinidade para a purificação e detecção por cromatografia líquida de alta eficiência em fase reversa. Não foram detectadas aflatoxinas nas amostras de ração. Entretanto, detectou-se AFM 1 em 19 (52,8%) das 36 amostras de leite cru, em valores que variaram de traços a 74,1 ng L -1. Em leite pasteurizado, a AFM 1 foi detectada em 13 (38,2%) das 34 amostras, em níveis que variaram desde traços a 58,9 ng L -1 . Os valores encontrados de aflatoxina no leite estão dentro dos padrões tolerados pela legislação brasileira. As concentrações de aflatoxina nos leites cru e pasteurizado não diferiram entre si (p>0,05). O fato de não se ter detectado aflatoxinas na ração pode ser explicado pela sua ocorrência em baixas concentrações, isto é, inferiores ao limite de detecção do método utilizado (2 μg/Kg), o que não impediria o aparecimento da AFM 1 no leite e a detecção mediante metodologia mais sensível (2 ng/L).Termos para indexação: aflatoxina M 1 , leite, alimento. Efetuou-se revisão de literatura sobre as condições de crescimento dos fungos toxigênicos Aspergillus flavus e Aspergillus parasiticus em alimentos. Foram abordados os principais fatores que determinam o crescimento de fungos e a produção de aflatoxina (atividade de água, temperatura, potencial hidrogênico, potencial de oxirredução, composição do substrato e aditivos). Concluiu-se que a presença do fungo no alimento não implica, obrigatoriamente, em produção de micotoxina, assim como a toxina pode estar presente no alimento mesmo na ausência do fungo. A contaminação de alimentos por fungos significa perdas econômicas e risco à saúde do consumidor. PALAVRAS-CHAVE: FUNGOS; MICOTOXINAS. , Valbert Nascimento Cardoso 4 RESUMOFoi verificado o efeito da irradiação gama ( 60 Co) na capacidade de destruir a microbiota fúngica, em amendoim em grão, cultivar Tatu Vermelho, da safra 2003 (segundo semestre). Os grãos de amendoim, após a irradiação, foram mantidos à temperatura ambiente em embalagem plástica comercial, durante 180 dias. Para a determinação da porcentagem fúngica foi utilizada a técnica do plaqueamento direto utilizando o meio Ágar Dicloran Rosa de Bengala Cloranfenicol (DRBC), desinfetando ou não os grãos com solução de hipoclorito de sódio. Em grãos de amendoim irradiados e desinfetados externamente, observou-se redução da infecção fúngica a 5 kGy e destruição total de fungos a 10 kGy, após 180 dias de armazenamento à temperatura ambiente. Em grãos irradiados e não desinfetados externamente foram verificados, em função do tempo de armazenamento, aumento da população de fungos com a dose de 1 kGy, redução com a dose de 5 kGy e eliminação total com a aplicação de 10 kGy. The peanuts grains, after irradiation, were kept at room temperature in plastic bags during 180 days. To determine the percentage of fungi, the direct plating technique was used and the grains were plated out on mycological media dicholoran rose bengal chloranphenicol (DRBC), being desinfected or not with sodium hypochlorite solution. Irradiated and desinfected peanuts was observed a reduction of fungi infection at 5 kGy and total fungi destruction at 10 kGy, after 180 days in storage at room temperature. Irradiated and non desinfected grains showed a increase of fungi population with 1 kGy dose, reduction with 5 kGy dose and total destruction with 10 kGy dose. Gamma-irradiation in 10 kGy dose or higher, showed to be an efficient process to reduce the mycoflora of peanuts, Tatu Vermelho cultivar.
