To determine the period of harvest that optimizes the antimicrobial activities of the essential oil of Ocimum gratissimum L. from Benin, aerial plant parts were collected at two vegetative stages (pre- and full-flowering) and three sampling times (7 am, 1 pm, and 7 pm). Extraction by hydrodistillation yielded between 0.65 and 0.78% of essential oils. Characterization of the oils by GC-FID and GC/MS analysis revealed the presence of monoterpenes (87.26-93.81%), sesquiterpenes (5.57-11.34%), and aliphatic compounds (0.15-0.18%), with p-cymene (1; 28.08-53.82%), thymol (2; 3.32-29.13%), γ-terpinene (3; 1.11-10.91%), α-thujene (4; 3.37-10.77%), and β-myrcene (5; 4.24-8.28%) as major components. Two chemotypes were observed, i.e., a p-cymene/thymol and a p-cymene chemotype, for plants harvested at 7 am for the former and at 1 pm or 7 pm for the latter, respectively. The oils were fungicidal against Candida albicans, with the sample from full-flowering plants collected at 7 am being the most active (MIC = 0.06±0.00 mg/ml). The chemical variation of the oils also influenced the antimicrobial effect against Staphylococcus aureus; the most active oil was obtained from plants at the pre-flowering stage collected at 7 am (MIC=0.24±0.01 mg/ml). Escherichia coli was insensitive to the chemical variation of the oils (MICs of ca. 0.48±0.02 mg/ml for all oils). Moreover, the essential oils showed low toxicity against Artemia salina Leach larvae, with LC(50) values in the range of 43-146 μg/ml. This is the first study of the interaction between the daytime of collection and vegetative stage of the plants and the antimicrobial properties and toxicity of the essential oil of O. gratissimum from Benin.

A chemical study of the hydro-ethanol extract of the leaves of Combretum glutinosum resulted in the isolation of nine compounds, including 5-demethylsinensetin (1), umuhengerin (2), (20S,24R)-ocotillone (3), lupeol (4), β-sitosterol (5), oleanolic acid (6), betulinic acid (7), corymbosin (8) and β-sitosterol glucoside (9). Four compounds have been isolated for the first time from the genus Combretum [viz. (1), (2), (3) and (8)]. The crystal structures of flavonoid (2), C20H20O8, Z′ = 2, and triterpene (3), C30H50O3, Z′ = 1, have been determined for the first time; the latter confirmed the absolute configuration of native (20S,24R)-ocotillone previously derived from the crystal structures of related derivatives. The molecules of (3) are linked into supramolecular chains by intermolecular O—H...O hydrogen bonds. The crude extracts obtained by aqueous decoction and hydro-ethanolic maceration, as well as the nine isolated compounds, were tested for their anthelmintic activity on the larvae and adult worms of Haemonchus contortus, a hematophage that causes parasitic disorders in small ruminants. The evaluated anthelmintic activity showed that the extracts at different doses, as well as all the compounds tested at 150 µg ml−1, inhibited the migration of the larvae and the motility of the adult worms of the parasite compared with the phosphate buffer solution negative reference control. The best activity was obtained with flavonoids (1), (2) and (8) on both stages of the parasite. The flavones that showed good activity can be used for the further development of other derivatives, which could increase the anthelmintic efficacy.

Introduction: The presence of microbes in our environment is always a permanent public health problem. In this context, research on natural treatment, less expensive and accessible to fight these microbial germs would be beneficial. Methods: During this work, molecules of thiosemicarbazones due to their numerous biological activities were hemi-synthesized in situ in the essential oil of Cymbopogon citratus in order to evaluate their antimicrobial activities. Results: Analysis of the essential oil extracted by hydrodistillation revealed the presence of 72.91% of citral. Citralthiosemicarbazone (CThio) and citral 4-phenyl-3-thiosemicarbazone (CPthio) were hemi-synthesized in this oil with interesting yields of 83% and 91%, respectively. After purification and confirmation of the structures of these molecules, the three substances were tested on eleven strains of microbes. Determination of the inhibition diameters showed that the activity of the essential oil is best in over 80% of strains. However, the largest diameter of inhibition (26 mm) was noted with CPthio against Salmonella typhi R 30951401. The determination of the minimum inhibitory concentrations showed that the oil remains more active with the smallest value of 0.3125 mg / ml against Micrococcus luteus. The lower value of minimum bactericidal concentrations was also obtained with the essential oil against Enterococcus foecalis ATCC 29212. Conclusion: The essential oil of C. citratus remains more active in the majority cases. It could be a great alternative in the fight against bacteria, and the advantage is that it remains a natural substance.
