The problem of illegal circulation of drugs, their support and falsification are relevant not only in Ukraine. Experts believe that about 10% of the global medicinal drug market is fake and fraud. Counterfeit drugs may contain the active ingredient with its exact content as well as non-relevant ingredients, may not contain the active ingredient at all or may contain unrefined ingredients. In any case, this leads to a lack of therapeutic effect in the treatment, poisoning and, even, death of patients. As a result of our research, the presence of highly dangerous and toxic components in the composition of drugs (antifreeze, tar, arsenic, mercury and others) was established. In case of poisoning or death of a patient after the use of a counterfeit medicinal drug, an examination is carried out, and the presence of the active and additional ingredients of the drug is confirmed by chemical toxicological analysis. Chemical-toxicological analysis is a part of the science "Forensic chemistry" and allows us to confirm the presence of the drug in the body, the purity of the drug, the presence of the active ingredient of the drug, and therefore allows us to confirm the fact of poisoning or death of a person under the action of a chemical substance (for example, a falsified drug).
Abstract:The synthesis was performed using 5-methyluracil mono-and bis-derivatives with the halogencontaining pharmacophore groups in the composition of molecules; their physiochemical and biological properties were studied. These reactions are a typical example of substitution reactions on heteroatom N (1) of uracil molecule using as the second component the reaction of halothane-the known inhaled anesthetic. The reactions were carried out in a solvent system: benzene-dimethyl formamide and diethyl ether, under conditions of phase transfer catalysis DB-18-crown-6-complex (alkaline medium), by heating from 2 to 11 hours, followed by purification, and drying of the resulting products.The structure of the synthesized compounds was confirmed by elemental analysis, IR and 1 HNMR spectra. The purity was controlled by the methods of thin-layer and gas-liquid chromatography. Chromatography, IR and 1 HNMR spectra of the final products were identified in comparison with the chromatograms, IR and 1 HNMR spectra of the initial compounds. Variation of the reaction conditions and changes in methods of synthesis enabled to receive the synthesized compounds of high purity and to increase their practical output up to 43-80%. Molecular complex of Bacterial lectin and 5-methyluracil bis-derivative was obtained.The toxicity and anti-tumor activity of some of the synthesized compounds were investigated. We have found that the new molecular complex of Bacterial lectin and 5-methyluracil bis-derivative has high antitumor activity in Limphosarcoma Plissa-62.8% (activity criterion > 50%), which allows us to consider the synthesized compound as a physiologically active compound with the prospect for further study as a potential vehicle for anti-tumoral treatment in patients.
The quantity of pharmaceutical preparations, which are used in medicine and different areas of the economy increased because of the development of the chemical and pharmaceutical industries. These substances can caise poisonings in certain situations. The sources of poisonings are: sewage of industrial enterprises; pesticides (chemical poisons) for fighting agricultural pests; soil; vegetables and fruits; chemical substances for fights against rodents and insects. The use of chemical substances in the economy increases the quantity of poisons as objects of forensic toxicological analysis. “Forensic chemistry” — is a science which studies methods for the isolation, purification, qualitative analysis and quantitative analysis of toxic and poisonous substances and their metabolites in objects of various origins: animal and plant biological materials, industrial waste, emissions of waste water, the air of industrial enterprises, soil and agricultural crops, etc. The aim of this work is to study the importance of chemicals, namely fluorine-containing organic compounds, which are strong toxic substances, during the study of "Forensic Chemistry" throughout the academic period at the Pharmaceutical Faculty in Ukraine.
Синтез біологічно активних сполукSynthesis of the biologically active compounds П ошук нових біологічно активних сполук нині здійснюють за двома напрямами: пошук неспе-цифічних фармакологічних агентів з широким спектром реалізації біологічного ефекту (із невстановленим меха-нізмом дії чи з двома та більше метаболічними шляхами) -неспецифічна дія; пошук високоспецифічних, так зва-них «малих молекул» із чітко окресленим механізмом дії, впливом на конкретну біологічну мішень (інгібування/ активація певного ферменту, рецептора, інтеркаляція в конкретну послідовність нуклеотидів тощо) [1][2][3][4].У попередніх дослідженнях показали, що бактерійні лектини цих штамів бацил виявляють виражений і вибір-ковий протипухлинний ефект щодо пухлин різного виду.Протипухлинна активність цих біологічно активних речовин (БАР) та їх вузька вуглеводна специфічність дають можливість використовувати бактерійні лектини як базисні речовини для конструювання медико-біо-логічних препаратів спрямованої дії [5]. Молекулярні комплекси з прогнозованою біологічною активністю, що утворюються на основі біологічних структур -бактерійних лектинів та гетероциклічної компоненти, -у фаховій літературі не описані. Тому певний інтерес становило створення та дослідження біологічної актив-ності, зокрема протипухлинної активності молекулярних Один з актуальних напрямів пошуку нових протипухлинних засобів -дослідження антиметаболітів піримідинового та пуринового обміну, які є преформованими спо-луками в низці біосинтетичних процесів (ДНК, РНК, специфічні білки), гальмуючи пухлинний ріст. З метою одержання нових потенційних протипухлинних засобів створили молекулярні комплекси на основі заміщених 1,1-біс-(піримідин-2,4-діон-1-іл)-2-бромо-2-хлороетиленів і бактерійного лектину Bacillus polymyxa. Встановили, що молекулярні комплекси біс-похідних піримідинів і бактерійного лектину є мало-токсичними (ЛД 50 від 338 мг/кг до 335 мг/кг) і мають значний протипухлинний ефект (до 62,8% на лімфосаркомі Пліса). Це свідчить, що нові молекулярні комплекси є перспективними для наступного вивчення як малотоксичні речовини з високою протипухлинною активністю.
