The mammalian target of rapamycin (mTOR) is a nutrient and ATP sensor suggested to play an important role in tumorigenesis, particularly in the setting of PTEN loss or activated Akt/PKB. Of mTOR's two known effectors, eIF4E has been implicated in tumorigenesis, whereas the role of S6 kinase (S6K1) in transformation is less understood. To assess the contribution of S6K1 to the transformed phenotype, we pharmacologically and genetically manipulated the mTOR-S6K pathway in glioma cells and monitored its effects on growth in soft agar, a hallmark of cellular transformation, and also assessed in vivo intracranial growth. Anchorage-independent growth by HRas V12 -transformed human astrocytes as well as by U251 and U373 human glioma cells was inhibited by pharmacologic mTOR inhibition. Similarly, short hairpin RNA-mediated suppression of mTOR also reduced anchorage-independent growth of glioma cell lines. Expression of wild-type eIF4E in rapamycin-treated E6/E7/hTert/HRas V12 and U373 cells failed to rescue colony formation, although expression of wild-type S6K1 or rapamycin-resistant S6K1 in rapamycin-treated U373 and U251 provided partial rescue. Consistent with the latter observation, small interfering RNAmediated suppression of S6K1 in HRas V12 -transformed human astrocytes, U251, and U373 cells resulted in a significant loss of anchorage-independent growth. Furthermore, we found that in vivo short hairpin RNA-mediated suppression of S6K1 in HRas V12 -transformed human astrocytes reduced intracranial tumor size, in association with reduced tumor levels of phosphorylated ribosomal protein S6. These findings implicate the mTOR-S6K pathway as a critical mediator of glial cell transformation. [Cancer Res 2008;68(16):6516-23]

Background and Objectives:The increasing worldwide incidence of obesity shows it is a global epidemic, emphasizing its status of public health problem. Obesity also represents a childhood disease, requiring health professionals to carry out preventive actions that include the entire context in which children and adults are considered. Based on this subject, we aimed to discuss the problem of childhood obesity and its association with three agents that influence the lifestyle of children with overweight and obesity: audiovisual media, family relations and academic institutions. Content: This was a literature review article, carried out at the search engines (SciELO and PubMed), seeking for articles that had in their context at least two of the following descriptors: academic institutions, family relations, audiovisual media and childhood obesity. The period considered was from 2009 to 2013. Conclusion: we verified the need for public policies that regulate advertising aimed at children, adapting them to the preventive/educational profile. Additionally, it was observed that the school plays a key role in the practice of healthy eating habits. It is clear, therefore, the need to adapt to this reality, with the family exercising the role of mediating and controlling such actions.
Obesidade. Promoção da saúde. ABSTRACT Background and objectives:Currently, obesity is considered a major public health problem because its prevalence set up at an alarming rate. The costs allocated to spending on obesity are very high and in the long run, can become obstacles to the sustainability of treatment of individuals, whether in the public, as in private. The objective was to reflect on the importance of public policies aimed at combating the epidemic of obesity and its costs to
The integration of professional physical education in multidisciplinary teams of family health strategy ABSTRACTThe present study aimed to reflect on the importance of multidisciplinary teams in the Family Health Strategy, focusing on the importance of physical education professional. The methodology consisted of literature searches, based on the Ministerial Decree 154 of January 2008 that created the Centers of Support for Family Health (CSFH) and guidelines. The location of the data sources occurred in conventional library systems and web search, as LILACS and SciELO, in the period April-June 2013. The following key words were used: family health, physical education, health promotion; prevention of diseases. The results point to the importance of the role of physical education professional in Centers of Support for Family Health is recognized, however, it is necessary to consolidate their involvement in multidisciplinary teams, as it plays a key role in disease prevention and health promotion.
Objective: To verify if there is an association between cardiometabolic risk factors and active daily commuting to school among children and adolescents. Methods: A total of 1,743 schoolchildren aged 7 to 17 years old were evaluated in the city of Santa Cruz do Sul (RS). The way of commuting to school was investigated with a questionnaire, and the cardiometabolic risk factors analyzed were body mass index (BMI), waist circumference (WC), systolic (SBP) and diastolic (DBP) blood pressure, blood glucose, triglycerides, total cholesterol (TC) and fractions, LDL and HDL. Results: The prevalence of active commuting among schoolchildren was 48.0% (95%CI 45.7-50.4), and it was associated, in the crude analysis, with blood glucose and LDL cholesterol levels. Passive schoolchildren had a 1.1 higher prevalence ratio of high glucose and LDL cholesterol levels. However, when sociodemographic variables were included in the model, these associations were not maintained. Conclusions: The prevalence of active commuting in the sample studied is low and it was shown to have a crude association with glucose and LDL cholesterol levels in students. However, sociodemographic factors seem to influence these associations.
