The main properties and biological effects of the antioxidant carnosine, the natural dipeptide β-alanyl-L-histidine, are considered. Data on the effective use of carnosine in different pathologies are presented. Special attention is paid to issues of use of carnosine in neurologic and mental diseases, in alcoholism as well as in physiological states accompanied by activation of free-radical processes and formation of oxidative stress.

Background: Alcoholism is an acute social problem worldwide for negative impact on health. This problem is closely associated with social stress and drinking traditions. Previously, it was established exhaustion of blood antioxidant in alcoholism patients. It requires development and investigation of substances with protective properties under ethanol impact, on both cells and biomolecules. Aims and Objectives: Lithium salts are widely used in medicine as mood stabilizers for mental pathologies, including alcoholism. In this work, we investigate lithium ascorbate for the protection of human blood plasma lipids and proteins under ethanol impact. Materials and Methods: Well-known antioxidants -carnosine and ascorbic acid -were used as reference drugs. Heparinized venous blood was used as an experimental substrate. The ethanol and tested substances in the form of water (physiological)solutions were added to blood in vitro. Protection effects were measured and estimated as concentrations of carbonyls proteins and products of lipid peroxidation in blood plasma. Results: It was shown similar protective action of lithium ascorbate and carnosine on human plasma biomolecules against damaging action of ethanol. No protective effect revealed for ascorbic acid. Conclusion: Lithium ascorbate and carnosine were proved to possess the protection effects for blood lipids and proteins under ethanol impact. Normothymic and bloodprotective properties are desirable for psychotropic drugs, and thus, lithium salts could be considered as prospective agents in the treatment of addictions and other pathologies associated with oxidative stress.

Organic lithium salts containing anionic components (succinate, fumarate, pyruvate and antioxidant ascorbate) were tested for protection of blood plasma proteins and lipids against ethanol-induced oxidation in vitro. We used normothymic lithium carbonate and well-known antioxidant dipeptide carnosine (b-alanyl-L-histidine) as the reference drugs. The oxidized proteins and lipids were determined by the level of carbonylated proteins (CP) and TBA-reactive products (TBA-RP), respectively. In alcoholic patients the level of oxidized proteins and lipids was higher than in healthy persons. Incubation of blood with ethanol resulted in an increase in oxidized proteins and lipids in blood plasma of healthy persons but had no influence on the level of CP and TBA-RP in blood plasma of alcoholic patients. Lithium carbonate, lithium ascorbate, and lithium succinate exhibited protective action against ethanol-induced oxidation of biomolecules of blood plasma of healthy people. These effects were comparable with carnosine action. The studied compounds had no effect on the level of CP and TBA-RP of blood plasma of alcoholic patients.

Исследовано влияние солей лития, содержащих разные анионные компоненты (карбонат лития-Li-CAR, пируват лития-Li-PYR и фумарат лития-Li-FUM), на продукт окислительной модификации ДНК-8-гидрокси-2'-дезоксигуанозин (8-OHdG)-в плазме после инкубации крови здоровых лиц (группа ЗД), больных алкогольной зависимостью (группа АЗ) и больных биполярным аффективным расстройством (группа БАР) с этими соединениями в присутствии 0,5 % этанола in vitro. Для оценки концентрации 8-OHdG в плазме крови применяли иммуноферментный метод. Этанол в используемых экспериментальных условиях не влиял на уровень 8-OHdG в пробах всех обследуемых групп. В пробах без соединений лития (контроль) у больных АЗ концентрация 8-OHdG была выше, чем у здоровых лиц, а у больных БАР не отличалась от ЗД. Соли лития не оказывали влияние на концентрацию 8-OHdG в пробах здоровых лиц и больных алкогольной зависимостью, тогда как в пробах группы больных БАР Li-PYR статистически значимо снижал уровень 8-OHdG. Сделано заключение о перспективности пирувата лития для создания новых фармакологических препаратов, сочетающих нормотимические, антиоксидантные и цитопротекторные свойства. ключевые слова: 8-гидрокси-2'-дезоксигуанозин (8-OHdG), карбонат лития, пируват лития, фумарат лития, алкоголизм, биполярное аффективное расстройство

IntroductionThe creation of new lithium compounds with antioxidant activity is relevant problem for psychiatry. The aim of this work was study of the protective effect of lithium salts against ethanol-induced oxidative damage to proteins and lipids of human blood plasma in vitro.MethodsWe used lithium ascorbate and lithium carbonate 0.6 mmol/L which correspond to the therapeutic dose (in terms of lithium ions). Antioxidant carnosine (β-Ala-L-His) was used as comparison drug. We used the blood of 12 healthy donors. The heparinized blood samples were incubated in presence of tested preparations for one hour at 37 °C. The final ethanol concentration in samples was 0.5%. Oxidative modification of proteins was determined as the level of carbonylated proteins with 2.4–dinitrophenilhydrazine, lipid peroxidation products–as the level of TBA-reactive products by spectrometry. Statistical analysis was performed with “Statistika 10” program.ResultsThe addition of ethanol in the blood led to a significant increase in carbonylated proteins and TBA-reactive products in the plasma (carbonylated proteins: without ethanol 0.26 ± 0.01 nmol/mg of protein; with ethanol 0.33 ± 0.02 nmol/mg; TBA-reactive products: without–3.2 ± 0.1 nmol/mL; with–4.0 ± 0.2 nmol/mL, P < 0.05). In the presence of carnosine such increase of oxidized products of biomolecules is not observed, i.e. carnosine had a protective effect against ethanol-induced oxidative damage. Lithium ascorbate showed a protective effect like carnosine. Lithium carbonate revealed no detectable influence on biomolecules in the conditions of our experiment.ConclusionLithium ascorbate has a protective effect on blood plasma proteins and lipids under ethanol-induced oxidative damage of biomolecules.Disclosure of interestThe authors have not supplied their declaration of competing interest.

Исследован уровень окислительной модификации белков (карбонилированных белков, КБ) и липидов (ТБК-реактивных продуктов, ТБК-РП) плазмы крови, а также активность супероксиддисмутазы (СОД) в плазме и гемолизате эритроцитов крови больных алкогольной зависимостью в динамике стандартной антиалкогольной терапии: на 3-4 день после поступления пациента в стационар (1-я точка) и на 12-14 день терапии (2-я точка). Обнаружено статистически значимое повышение уровня КБ и ТБК-РП у больных в 1-ой точке обследования относительно соответствующих показателей здоровых лиц (контроля). Активность СОД как в плазме крови, так и в гемолизате у больных алкоголизмом в этот период (в 1-й точке) практически не отличалась от контроля. Дальнейшее лечение приводило к снижению карбонилированных белков и ТБКреактивных продуктов плазмы крови до величин, статистически не отличающихся от контрольных. Активность СОД в плазме крови в группе пациентов через 12-14 дней лечения оставалась практически такой, как и в 1-й точке обследования, а в гемолизате эритроцитов достоверно увеличилась. Полученные данные позволяют рассматривать карбонилированные белки и ТБК-реактивные продукты, как и СОД, в качестве кандидатов для включения в состав комплексного биомаркера оценки эффективности терапии и устойчивости терапевтической ремиссии у больных алкогольной зависимостью.
