RESUMO.-[Influência do Fator de Crescimento Semelhante à Insulina-I (IGF-I) sobre a sobrevivência e o desenvolvimento in vitro de folículos pré-antrais capri-nos.] O objetivo do presente estudo foi investigar os efeitos do fator de crescimento semelhante a insulina-I (IGF-I) na sobrevivência, ativação (transição de folículos primordiais para primários) e crescimento de folículos pré-antrais caprinos cultivados in vitro. Fragmentos de córtex ovariano foram cultivados por um e sete dias na ausência ou presença de IGF-I (0, 50 e 100ng/mL). Os tecidos não cultivados e cultivados foram processados e analisados por histologia e microscopia eletrônica de transmissão. O cultivo por um dia em meio com 100ng/mL de IGF-I apresentou 86,7% de folículos morfologicamente normais. Estes resultados foram semelhantes (P>0,05) ao percentual de folículos normais encontrados no controle (96,7%). Verificou-se ainda que este meio aumentou o percentual de ativação folicular (folículos em desenvolvimento) com um dia de cultivo. Os diâ-metros ovocitário e folicular mantiveram-se semelhantes ao controle ao cultivar por um dia em meio contendo 100ng/ The aim of this study was to investigate the effects of the insulin-like growth factor -I (IGF-I) on survival, activation (transition from primordial to primary follicles) and growth of caprine preantral follicles cultured in vitro. Fragments of ovarian cortex were cultured for one and seven days in the absence or presence of IGF-I (0, 50 and 100ng/ml). The non--cultured and cultured tissues were processed and analyzed by histology and transmission electron microscopy. The culture for one day in a medium with 100ng/ml of IGF-I showed 86.7% of morphologically normal follicles. These results were similar (P>0.05) to the percentage of normal follicles found in the control (96.7%). It was also found that this medium increased the percentage of follicular activation (developing follicles) with one day of culture. The oocyte and follicular diameters remained similar to the control by culturing for one day in a medium containing 100ng/ml of IGF-I. The ultrastructural analysis did not confirm the integrity of the follicular fragments in a medium containing IGF-I (100ng/ml) after one and seven days of culture. In conclusion, this study demonstrated that the addition of 100 ng/ml of IGF-I in the culture medium enables the development of preantral follicles of goats with one day of culture. However, it is not sufficient to maintain the follicular integrity and the follicular survival rate after seven days of culture.

RESUMO O Brasil atualmente é detentor do terceiro maior rebanho de vacas leiteiras do mundo, composto, em sua maioria, de animais mestiços F1 (Gir X Holandês), os quais são bem adaptados às áreas tropicais. O objetivo do presente estudo foi investigar a eficiência da ovum pick-up (OPU) e da produção in vitro de embriões oriundos de doadoras Girolando com sêmen sexado de touros provenientes de duas raças, Gir e Holandesa. Foram utilizados dados referentes a 232 animais de diversos rebanhos, totalizando 4334 oócitos recuperados. Os oócitos foram classificados, e as estruturas viáveis (GI, GII e GIII) foram utilizadas para produção in vitro de embriões (PIVE). Foi observada uma média de oócitos recuperados para as doadoras F1 de 18,14±1,33. Além disso, notou-se que não houve diferença entre os dois cruzamentos utilizados, considerando média de total de embriões (4,96±0,40 e 6,56±0,76) para o cruzamento F1 X HPB (Holandês preto e branco) e F1 X Gir, respectivamente. Portanto, as doadoras F1 apresentaram potencial como doadoras de oócitos no sistema de produção in vitro de embriões, independentemente da raça do touro cujo sêmen foi utilizado.
The aim of this study was to investigate the effects of human follicle stimulating hormone (hFSH) on the in vitro culture of caprine preantral follicles. Fragments of goat ovarian cortex were cultured in α-MEM + supplemented with 0, 10, or 50 ng/mL hFSH for one or seven days. Small fragments of non-cultured and cultured ovarian tissue were processed for classic histology and transmission electron microscopy. The statistical tests used in this study were Lilliefors, Cochran, and Tukey's (when significance was detected, PROC ANOVA, SAS was used). The results revealed a reduction in the percentage of normal follicles after one or seven days of culture under all treatment conditions. The rates of follicular survival were similar to each other, within each day of culture. The medium containing 10 ng/mL hFSH reduced the percentage of primordial follicles following culture for one and seven days and did not increase the percentage of developing follicles. The follicular diameter of ovarian tissue cultured in α-MEM + medium and medium supplemented with 10 ng/mL of hFSH did not change when compared with the control (non-cultured). The ultrastructural analysis confirmed the integrity of follicles cultured for seven days in medium containing 10 ng/mL hFSH. In conclusion, hFSH (10 ng/mL) is capable of promoting the activation of primordial follicles and maintaining the ultrastructural integrity of caprine preantral follicles cultured in vitro for seven days. Fragmentos do córtex ovariano de caprinos foram cultivados em α-MEM + suplementado com 0, 10, ou 50 ng/mL de hFSH por um ou sete dias. Pequenos fragmentos de tecido ovariano cultivado e não cultivados, foram processados por histologia clássica e microscopia eletrônica Costa, S. L. da et al.de transmissão. Os testes estatísticos utilizados na pesquisa foram Lilliefors, Cochran, e Tukey's (caso significância -PROC ANOVA; SAS). Os resultados mostraram uma redução no percentual de folículos normais, após um ou sete dias de cultivo em todas as condições de tratamento. As taxas de sobrevivência folicular foram semelhantes entre si, dentro de cada dia de cultivo. O meio contendo 10 ng/mL de hFSH reduziu a percentagem de folículos primordiais no dia um e sete de cultivo, e não aumentou a percentagem dos folículos em desenvolvimento. O diâmetro folicular de tecido ovariano cultivado em meio α-MEM + e meio suplementado com 10 ng/mL de hFSH não alterou, quando comparado com o controle (não cultivado). As análises ultraestruturais confirmaram a integridade de folículos cultivados por sete dias em meio contendo 10 ng/mL de hFSH. Em conclusão, o hFSH (10 ng/mL) é capaz de promover a ativação de folículos primordiais e manter a integridade ultraestrutural de folículos pré-antrais caprinos cultivados in vitro por até sete dias.
