S. G. Leitão wishes to dedicate this work in memory of Prof. Nikolai Sharapin who kindly introduced the author to the CYTED ProgramRESUMO: "Triagem de extratos de plantas das Américas Central e do Sul para atividade antimicobacteriana pelo teste do Alamar Blue". Unitermos: Mycobacterium tuberculosis, Lantana trifolia, Vitex cooperi, Lippia lacunosa, Lippia rotundifolia, atividade antimicobacteriana, 20-hidroxi-ecdisona.ABSTRACT: Forty eight ethanolic crude extracts and fractions (hexane, dichloromethane, ethyl acetate and n-butanol) from ten Brazilian plants (Leguminosae, Monimiaceae and Verbenaceae), 1 from Costa Rica (Verbenaceae) and 1 from Argentina (Verbenaceae) were screened for antimycobacterium activity against Mycobacterium tuberculosis (ATCC-27294 H 37 Rv), by the Alamar Blue test, at a fi xed concentration of 100 μg/mL. Out of the forty eight, seven were active at this concentration, corresponding to Lantana trifolia (hexane and dichloromethane extracts from leaves), Vitex cooperi (methanol:water, 1:1 extract from barks), Lippia lacunosa (hexane and dichloromethane extracts from leaves) and Lippia rotundifolia (hexane and dichloromethane extracts from leaves), all from the Verbenaceae family.
The specific legislation for the production and use of medicinal plants has been improved in Brazil, as far as its continuous evolution is concerned. However, there is still a failure at surveillance quality of these products. This work showed a panorama of some espinheira-santa samples (Maytenus ilicifolia) purchased from local markets, through moisture, contaminants, epicatechin and total tannin content. Contaminants were found above the maximum limits admitted in 89 % of the total samples (stems, mosses and lichens) above allowable. Regarding water content, only samples 24, 25 and 27, all from group 3, proved to be outside official limits, which must be within 8 and 12 %. For tannin a variation from 0.18 to 2.96 % could be detected (official minimum limit must be 2 %) and, for epicatechin (pharmacopoeial marker), only 37 % of total samples showed its presence, as analyzed by TLC and HPLC. Three main groups were established by Principal Component Analysis (PCA), after laboratory assays. Group 2 with the highest amounts of the characteristic metabolites epicatechin and total tannin, suggested authentic samples. Group 1 and 3 with high level of moisture and contaminants suggested false samples. These results may, in part, suggest that possible deleterious effects caused by the consumption of espinheira-santa available on the market, could be due to the unconformities observed in the analyzed samples.
Leaves of Monteverdia ilicifolia (“espinheira-santa”) are considered a medicinal tea by the Brazilian Sanitary Surveillance Agency (Anvisa), by their anti-dyspeptic, anti-acid and protective of the gastric mucosa properties. Their spiny margins are similar to those of other botanical species, which may lead to misidentifications. The aim of this work was to evaluate the authenticity of 32 samples of herbal drugs commercialized as “espinheira-santa” in the formal trade in Brazil, by macro and microscopic morphological studies of the leaves. The evaluation of the botanical authenticity was based on leaf venation patterns, shape and anatomy of the petiole and midrib region in cross section, vascular system arrangement and epidermal characters. Analysis of these characters compared to literature data suggests that 34% of the samples are M. ilicifolia and the remaining 66% are Sorocea bonplandii, a species with no clinical studies assuring its effective and safe use, representing thus a potential risk to public health.
