The present study investigated the growth inhibition effect of the fluoroquinolone antibiotics enrofloxacin and ciprofloxacin on four photoautotrophic aquatic species: the freshwater microalga Desmodesmus subspicatus, the cyanobacterium Anabaena flos-aquae, the monocotyledonous macrophyte Lemna minor, and the dicotyledonous macrophyte Myriophyllum spicatum. Both antibiotics, which act by inhibiting the bacterial DNA gyrase, demonstrated high toxicity to A. flos-aquae and L. minor and moderate to slight toxicity to D. subspicatus and M. spicatum. The cyanobacterium was the most sensitive species with median effective concentration (EC50) values of 173 and 10.2 µg/L for enrofloxacin and ciprofloxacin, respectively. Lemna minor proved to be similarly sensitive, with EC50 values of 107 and 62.5 µg/L for enrofloxacin and ciprofloxacin, respectively. While enrofloxacin was more toxic to green algae, ciprofloxacin was more toxic to cyanobacteria. Calculated EC50s for D. subspicatus were 5,568 µg/L and >8,042 µg/L for enrofloxacin and ciprofloxacin, respectively. These data, as well as effect data from the literature, were compared with predicted and reported environmental concentrations. For two of the four species, a risk was identified at ciprofloxacin concentrations found in surface waters, sewage treatment plant influents and effluents, as well as in hospital effluents. For ciprofloxacin the results of the present study indicate a risk even at the predicted environmental concentration. In contrast, for enrofloxacin no risk was identified at predicted and measured concentrations.
In ecotoxicology species reproduction tests and multiple testing of reproduction data are wide spread. While normal approximation of the data is a minor problem often the requirement of variance homogeneity is not fulfilled. Variance homogeneity is necessary to assure the proper application of statistical procedures like pairwise t tests, Dunnett t test, and Williams t test. A Poisson model can solve this issue preserving meaningful results and rendering statistical analysis more reliable. Moreover, sequential application of pairwise statistical ''control vs. treatment'' tests is a drawback concerning a-inflation. The closure principle (CP) for hypothesis testing is used to generate a step-wise approach for detection of the No/Lowest Observed Effect Concentration using the computational approach test (CAT). The advantages and disadvantages of the combined CPCAT approach compared to the widely used t tests are pointed out and results of real data and fictitious data analysis are compared revealing the superiority of the Poisson model and CPCAT.
Zusammenfassung Hintergrund und Ziel In der modernen Land-und Forstwirtschaft ist die Verwendung von Pflanzenschutzmitteln (PSM) weit verbreitet. Ihr Einsatz birgt jedoch auch Risiken für die Umwelt, da im Rahmen ihrer bestimmungsgemäßen Verwendung auch ein Eintrag in Nichtzielflächen nicht auszuschließen ist. So kann z. B. durch Abdrift, durch Niederschlagswasser sowie durch Run off und Dränagen von behandelten Flächen ein Eintrag in Oberflächengewässer erfolgen. Obwohl wurzelnde dikotyle Makrophyten einen wichtigen Bestandteil des Ökosystems darstellen, werden sie bei der Bewertung des Risikos von PSM-Anwendungen für Wasserorganismen im Rahmen der Eingangsuntersuchung standardmäßig nicht berücksichtigt. Ziel der vorliegenden Arbeit war die Entwicklung eines standardisierten Einphasentestsystems mit dem zweikeimblättrigen Ährigen Tausendblatt Myriophyllum spicatum (Haloragaceae). Methoden Um substanzspezifische Daten zur Ökotoxizität zu erhalten, wurde ein sedimentfreies Einphasentestsystem gewählt, das Bedingungen bietet, die unabhängig vom Verteilungsverhalten der Testsubstanzen zwischen der Sediment-und Wasserphase sind. Die ausschließliche Exposition der Pflanzen über die Wasserphase beschränkt den analytischen Aufwand und erleichtert die Modellierung der Ergebnisse, da nur die Wasserphase berücksichtigt werden muss. Zudem ermöglicht diese Methode den direkten Vergleich mit Testergebnissen mit der monokotyledonen Wasserlinse Lemna sp. (OECD-Prüfrichtlinie 221, OECD 2006); Studien zur Toxizität gegenüber Lemna stellen eine Standardtestanforderung für Herbizide dar. Ergebnisse, Diskussion, Schlussfolgerungen Die Exposition von Myriophyllum spicatum erfolgt über 14 Tage unter axenischen Bedingungen im Klimaschrank. Mit dem Test wird die Ökotoxizität gegenüber Myriophyllum spicatum bezogen auf die Wachstumshemmung in Form einer Dosis-Wirkungs-Beziehung ermittelt. Zur Auswertung werden die Endpunkte Zunahme der Hauptsprosslänge, Wachstum der Seitentriebe, Zunahme der Gesamtsprosslänge (Summe aus Hauptsprosslänge und Seitentrieben) und Wachstum der Wurzeln sowie die Zunahme des Frisch-und Trockengewichtes der Pflanzen herangezogen. In dieser Arbeit werden das Testdesign beschrieben und Ergebnisse mit 3,5-Dichlorphenol vorgestellt.
