The water phase antioxidant activity of extracts from 23 Bulgarian medicinal plants was studied in relation to their polyphenol content in comparison with mate, black tea, honeybush and rooibos foreign species. Antioxidant activity was measured by the ABTS (2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)) cation radical decolorization assay, and the total polyphenol content was assayed according to the Folin-Ciocalteu method. Five Bulgarian plant extracts exhibited higher antioxidant activity than that of mate, which is 21.7% of all Bulgarian herbs included in this study. These were Alchemilla vulgaris L. (4.79 +/- 0.14 mm), Sambucus ebulus L. (4.03 +/- 0.07 mm), Mentha spicata L. (3.90 +/- 0.03 mm), Fragaria vesca L. (3.74 +/- 0.06 mm), Crataegus monogyna Jacq. (3.63 +/- 0.05 mm). Another eight Bulgarian medicinal plant extracts exhibited an intermediate antioxidant activity - lower than that of mate and higher than that of honeybush, which makes 34.8% of all Bulgarian herbs included in the study. More than half of the herbal extracts included in the present study exhibited antioxidant activity higher than or comparable to the reference foreign plants. A positive correlation (r = 0.92) between antioxidant activity and polyphenol content was found, suggesting that the antioxidant capacity of the aqueous plant extracts is due to a great extent to their polyphenols.

Health has been defined as the capability of the organism to adapt to challenges. In this study, we tested to what extent comprehensively phenotyped individuals reveal differences in metabolic responses to a standardized mixed meal tolerance test (MMTT) and how these responses change when individuals experience moderate weight loss. Metabolome analysis was used in 70 healthy individuals. with profiling of ∼300 plasma metabolites during an MMTT over 8 h. Multivariate analysis of plasma markers of fatty acid catabolism identified 2 distinct metabotype clusters (A and B). Individuals from metabotype B showed slower glucose clearance, had increased intra-abdominal adipose tissue mass and higher hepatic lipid levels when compared with individuals from metabotype A. An NMR-based urine analysis revealed that these individuals also to have a less healthy dietary pattern. After a weight loss of ∼5.6 kg over 12 wk, only the subjects from metabotype B showed positive changes in the glycemic response during the MMTT and in markers of metabolic diseases. Our study in healthy individuals demonstrates that more comprehensive phenotyping can reveal discrete metabotypes with different outcomes in a dietary intervention and that markers of lipid catabolism in plasma could allow early detection of the metabolic syndrome.-Fiamoncini, J., Rundle, M., Gibbons, H., Thomas, E. The applicability of the FTIR attenuated total reflectance technique for in situ monitoring of plant physiological processes such as leaf senescence and aging has been examined. Difference spectra obtained by subtracting the spectrum of the young plant leaf from that of the older one revealed positive bands at 1650-1500 cm(-1), indicating a higher relative concentration of phenolics in the older leaves of both black cherry and sweet pepper bush leaves. Prolonged physiological stress of tobacco leaves exhibited a progressive time-dependent increase of the absorbance at around 3475 cm(-1), corresponding to hydroxyl functional groups. Absorption changes were also observed between 1650 and 1500 cm(-1), which are likely to correspond to phenolics. The characteristic changes of the FTIR absorbance spectra resulting from physiological and induced aging were detected also as a response to treatment with a recombinant alpha-elicitin, cinnamomin. This allowed the first quantification of the biological activity of a recombinant elicitin using a spectroscopic method. We suggest that FTIR spectroscopy provides important information about physiological events occurring in plant tissue in vivo, and it could be useful for the in situ characterization of the plant responsiveness to fungal toxins such as elicitins.

This study aimed to establish the effect of Sambucus ebulus L. (SE) ripe fruit infusion on body weight, blood pressure, glucose levels, lipid profile and antioxidant markers in healthy volunteers in respect of its possible protective activity against cardiovascular diseases and other oxidative stress-related diseases. The study involved 21 healthy volunteers, aged between 20 and 59, BMI 23.12 ± 1.31, who consumed 200 ml SE infusion/day for a period of 30 d. Blood samples were collected before and at the end of the intervention. Significant decrease in triglycerides (14.92%), total cholesterol (15.04%) and LDL-C (24.67%) was established at the end of the study. In addition, HDL-C/LDL-C ratio increased by 42.77%. Improved serum antioxidant capacity and total thiol levels were also established. The results presented in this first human intervention study with SE fruit infusion indicate the potential of the plant to improve lipid profile and serum antioxidant capacity in humans.

Phytotherapy reports strong anti-oxidative and anti-inflammatory potential of agrimony (Agrimonia eupatoria L.). However the effects of agrimony tea consumption are not scientifically proven in humans. We investigated the effects of one month's consumption of agrimony tea in healthy volunteers. Significant elevation of plasma total antioxidant capacity was observed and interleukin 6 levels were significantly lowered at the end of the intervention. An improved lipid profile as estimated by increased high density lipoprotein (HDL) cholesterol was established upon agrimony tea supplementation and HDL cholesterol correlated with adiponectin levels. The results presented in this first human intervention study with agrimony tea indicate that the plant has potential in improving markers of lipid metabolism, oxidative status and inflammation in healthy adults.
